A Single-molecule Manipulation Instrument Integrated with in Situ Nanopore Fabrication

摘要
Nanopore sequencing is a powerful tool in the field of Genomics and Proteomics due to its long reads and identification of molecular modifications. However, there is still a challenge for locating a nanopore in manipulation-based sequencing with solid-state nanopore. In this paper, we developed an instrument for performing in situ nanopore fabrication and precise molecule manipulation in a seamless way. We successfully controlled a single λDNA molecule exiting a nanopore at a speed about 1 bp/s. The proof-of-utility of the instrument provides a strong support for the realization of solid-state nanopore-based sequencing.
