•LC-MS offers an alternative to invasive methods in the bone marrow to measure MRD in the blood of patients with MM.•This LC-HRMS responds to two objectives: overcoming the potential interference of biotherapeutics and assessing the MRD.•Highly sensitive and specific quantification of intact M-protein light chains in patient serum was achieved.
