Study on the anti-tumor mechanism of Poria triterpenoids on mouse ascites tumor based on Lable-free proteomics

Abstract In order to understand the mechanism of tumor suppression of Poria triterpene on H22 ascites tumor, this paper used HPLC-LTQ-Orbitrap method to detect and identify the chemical composition of Poria triterpene site, based on the use of Lable-free proteomics, so as to explore the potential mechanism of action of Poria triterpene on tumor suppression of mouse ascites tumor. A total of 19 triterpenoids were identified by chemical composition profiling of Poria triterpenes, and 188 differentially expressed proteins were found in Poria triterpene group compared with the model group, of which 86 differentially expressed proteins were upregulated and 102 differentially expressed proteins were downregulated by GO database analysis; the differentially expressed proteins were mainly involved in the regulation of leukocyte GO database analysis showed that differentially expressed proteins were mainly involved in the regulationof interleukin-1 production, arachidonic acid secretion, autophagy and other biological processes; KEGG database analysis showed that differentially expressed proteins were involved in signaling pathways that were closely related to tumors, mainly MAPK, Apoptotic acid, and other proteins. Mainly MAPK, Apoptosis, mTOR, Wnt and p53 pathways; RT-qPCR was used to validate the genes encoding seven representative differential proteins at the mRNA level, and then Western blot was used to validate the differential proteins, and the results were consistent with the proteomic results. The mechanism of tumor suppressive effect of Poria triterpenes on mouse ascites tumor was found to be mainly involved in apoptosis, inflammatory response and immune process.
