Antigenic Nanostructures based on Amoxicilloyl-bidendrons for Effector Cell Activation to Study Allergic Reactions to Amoxicillin

Amoxicillin (AX) is the most common betalactam that causes allergic reactions. To be immunogenic, AX requires protein haptenation to form multivalent conjugates of increased size, suitable for crosslinking of specific IgE (sIgE) bound on basophils or mast cells (MCs). Using well-defined nanostructures allowed the identification of the shortest distance between antigenic determinants for cell activation in a realistic model using AX as a hapten. In this study, we designed nanostructures that varied both the number of antigenic determinants and the distance between them in order to assess the role of valence in promoting effector cell activation in the context of allergy to AX.
