Matrix metalloproteases (MMPs) are enzymes involved in the degradation of the extracellular matrix and display a critical role in a variety of physiological and pathological processes. They are of primary interest as therapeutic targets particularly for their role in carcinogenesis, but also in fibrosis, inflammation, abnormal angiogenesis, weakening of the matrix, microglial activation, autoimmune diseases, and wound healing. Herein the features of MMPs and major advances in the development of specific MMP inhibitors are discussed, reporting main drug candidates developed over last years that are currently under clinical trials or have been already approved by FDA for the treatment of cancer and other disorders, such as arthritis, neurological disorders, and periodontal disease.
