•Comprehensive 2D RPCCC × RPLC chromatographic analysis of Rheum palmatum L.•2D bioassay profilings were combined with 2D chromatogram for 2D bioscreening.•Separation and characterization of 11 constituents from Rheum palmatum L.•Two components were identified as natural α-glucosidase inhibitors for the first time.
