Serine is a crucial amino acid that is involved in numerous biological processes. It serves as a primary source of one-carbon (1C) units, which are critical for the biosynthesis of macromolecules and the regulation of gene expression through epigenetic mechanisms. Studies utilizing isotopic tracing have revealed that almost all of the methyl groups necessary for replenishing the 1C pool are derived from serine. Serine hydroxymethyltransferase 2 (SHMT2) is a pivotal enzyme in 1C metabolism that catalyzes the conversion of serine to glycine and folate-mediated 1C units.
