Abstract
Microfluidic diffusional sizing (MDS) is a powerful lab-on-a-chip method for determining the particle sizes, interactions, and association states of fluorescent analytes. MDS requires only microliter volumes and works even in heterogeneous, unpurified samples. Here we describe how MDS can be used to study (i) antibody binding and cellular copy numbers of oncoproteins, (ii) charge-dependent interactions of proteins with lipid membranes, and (iii) the size of native nanodiscs. We also show how single-molecule MDS (smMDS) achieves ultimate sensitivity and superior size resolution through digital counting.
