Molecular Cloning, Tissue Expression, and Bioinformatics Analysis of STMN1 Gene from Anhui White Goats

This study cloned the coding sequence region of the Stathmin 1 ( STMN1 ) gene of the local breed Anhui White Goats (AWGs), and preliminarily explored its protein structure and biological characteristics, and elucidated its expression patterns in various tissues of AWGs. The 450 bp coding region of the AWGs STMN1 gene encodes a 149 amino acid polypeptide with an unstable structure. Its amino acid sequence was identical to that of seven species including goat ( Capra hircus ), sheep ( Ovis aries ), and cattle ( Bos taurus ), and the lowest homology with human ( Homo sapiens ) was 86.6 %. Phylogenetic tree analysis showed that STMN1 was highly conserved in evolution. STMN1 was a hydrophilic protein without transmembrane structure and signal peptide, and its subcellular localization was mainly in the nucleus, followed by the cytoplasm. There were 12 phosphorylation sites, 2 threonine sites and 10 serine sites. STMN1 protein was mainly composed of alpha-helices, random coils, and beta turns, which belongs to beta beta alpha structure. The STMN1 expression profile in AWGs tissues detected by RT-qPCR and WB showed that it was ubiquitously expressed, with the highest expression in the hypothalamus, followed by the pituitary and ovary, and the lowest in the uterus, spleen, lung, and liver. It was speculated that STMN1 may regulate the AWG ' s growth and development by affecting the endocrine and nervous systems through the hypothalamus. In short, this study laid the foundation for exploring its possible biological function.
