Objective The repair of bone defects caused by periodontal diseases is an arduous challenge in clinical treatment. ETV2 transcription factors belong to the E26 transformation specific(ETS) family. It is reported that ETV2 signaling molecules can play a key role in promoting angiogenesis and are ideal biological signaling molecules for periodontal tissue regeneration. However, the role of ETV2 in the osteogenesis of human dental pulp stem cells(hDPSCs) remains unclear and needs to be verified. Methods The lentivirus vector based on the dox induction system was used to realize the transgene overexpression of ETV2. Real time quantitative polymerase chain reaction(qRT-PCR), western blot, immunofluorescence staining, alkaline phosphatase(ALP) staining and alizarin red S(ARS) staining were used to evaluate the effect of dox induced overexpression of ETV2 on hDPSCs osteogenesis. In addition, we also analyzed the potential mechanism of Etv2 induced osteogenesis by RNA sequencing(RNA Seq). Results The experiment proved that ETV2 significantly overexpression up-regulated the osteogenic markers ALP, COL1A1 and OSX, significantly enhanced the activity of ALP, and promoted matrix mineralization hDPSCs. In addition, RNA sequencing and Western blot analysis showed that ETV2 was activated after the ERK/MAPK and PI3K-Akt signaling pathways were transduced. The overexpression of ETV2 can enhance the osteogenic differentiation of hDPSCs, which is partially reversed by the treatment of ERK/MAPK and PI3KAKT signal inhibitors. Conclusion The experimental results show that the overexpression of ETV2 has osteogenic effect on hDPSCs, which provides a new idea for periodontal tissue reconstruction.
