Sequencing technologies such as targeted amplicon sequencing are highly effective approaches for studying genetic variation in specific genomic regions as they allow efficient variant identification and characterization.They are useful in studying specific biosynthetic pathways in plants that will allow the breeding of new cultivars with targeted composition.Given that flax seed is a major source of omega-3 fatty acids, we designed a study of the polymorphisms in genes involved in fatty acid biosynthesis.In this study, we report on the library preparation for the targeted amplicon sequencing of these genes.
Pathogenic isolates of Fusarium spp. associated with babaco (Vasconcellea heilbornii var. pentagona Badillo) were characterized morphologically and molecularly. Babaco roots with symptoms of root rot were collected in major areas in southern Ecuador, and monosporic isolates were obtained. Twenty-one isolates associated with the vascular wilt of babaco (VWB) were characterized culturally, morphologicaly, and molecularly. Amplification of ITS (Internal Transcribed Spacer) subregions placed between the ribosomal genes (rDNA) 18S was performed using specific primers for Fusarium identification. Based on morphological and cultural characteristics, 12 isolates were identified as belonging to Fusarium. Analysis of nucleotide sequences confirmed that all of them belonged to the Fusarium genus: Fusarium oxysporum (Schlechtendahl emend. Snyder & Hansen) (nine isolates), Fusarium camptoceras (Wollenweber & Reinking emend. Marasas & Logrieco) (one isolate), Fusarium temperatum (Scaufl. & Munaut) (one isolate), and Fusarium sp. (one isolate). This is the first report of these four Fusarium species causing VWB in the south of Ecuador. Key words: Fusarium, species, babaco, indentification.
This work aimed to investigate the effect of solvents on the extraction of some phythochemical from Melastoma malabathricum leaves. Melastoma malabathricum or locally known as Senduduk belongs tothe family of Melastomataceae and rich in flavonoid contents. The solvents tested were deionized water (H2O), ethanol (EtOH), ethyl acetate (EA) and hexane (Hex) and six hour of extraction time using Soxhlet extraction (SOX). The main compounds (quercetin and rutin) of Melastoma malabathricum were identified by High Performance Liquid Chromatography (HPLC) in specific solvents of extraction such as H2O,EtOH, EA and Hex. The results revealed that water was pronounced than the other solvents tested. The highest yield of extraction can be obtained during water extract(28.03 wt.%), followed by ethanol(26.61 wt.%), ethyl acetate (4.61 wt.%) and hexane (1.92 wt.%), respectively.In addition, Quercetin content was found to be higher in EtOH extract with 0.77 ppm, EA extract with 0.68 ppm, followed by Hex with 0.49 ppm. However, rutin content was found to be higher in EtOH extract (83.13 ppm), followed by EA extract (56.53 ppm) and Hex extract (0.66 ppm), respectively. The data generated in this work, provide an insight on phytochemical content of Melastomamalbathricum leaves at different solvents polarities.
