The Nathan S. Kline Institute for Psychiatric Research (NKI) is a New York State-funded research Institute, located in Orangeburg, New York.NKI is dedicated to research designed to better understand the causes of mental illnesses, and to improving the lives of people with mental illness. The Institute is named after psychiatrist Nathan S. Kline, MD who died in 1982.NKI is a facility of the New York State Office of Mental Health that has earned a national and international reputation for its pioneering contributions in psychiatric research, especially in the areas of psychopharmacological treatments for schizophrenia and major mood disorders, and in the application of computer technology to mental health services. Since 1952, interdisciplinary teams of distinguished NKI scientists have applied their talents and expertise to study the etiology, treatment, prevention, and rehabilitation of severe and persistent mental illnesses.Located on the grounds of Rockland Psychiatric Center in Orangeburg, New York (20 miles north of New York City), NKI receives additional operating support from federal, municipal, and private sources through the Research Foundation for Mental Hygiene. NKI has a strong academic collaboration with the Department of Psychiatry of New York University.A broad range of studies are conducted at NKI, including basic, clinical, and services research, intended to improve care for people suffering from complex, psychobiologically based, severely disabling mental disorders. They focus primarily on patient-oriented research programs emphasizing the causes, diagnosis, treatment, prevention, and care of severe and long-term mental disorders; clinically relevant, basic research on physiological and biochemical aspects of mental disease; and research on the cost, quality, and effectiveness of services for patients in mental health programs certified, operated, and/or funded by New York State..
Deep brain stimulation (DBS) is an emerging therapy for treatment-resistant neurological and psychiatric disorders. Despite this, little is known about the anatomical and functional mechanisms that underlie this therapy. We targeted DBS to white matter adjacent to the subcallosal anterior cingulate cortex (SCC-DBS) in macaques, modeling the approach proven effective for depression in humans. SCC-DBS caused a selective increase in fractional anisotropy, linked to white matter microstructure, in the cingulum bundle. At the cellular level, this was associated with an increase in both myelinated oligodendrocytes and the degree of myelination in the mid-cingulum bundle. SCC-DBS also altered brain-wide functional connectivity, changing interactions between the SCC and multiple brain networks, most notably the default mode network that has been implicated in depression. Overall, our data indicate that white matter remodeling as well as selective changes in multiple brain networks may contribute to DBS's therapeutic efficacy.
Despite decades of neuroimaging research, how white matter develops along the length of major tracts in humans remains unknown. Here, we identify fundamental patterns of white matter maturation by examining developmental variation along major, long-range cortico-cortical tracts in youth ages 5-23 years using diffusion MRI from three large-scale, cross-sectional datasets (total N = 2,716). Across datasets, we delineate two replicable axes of human white matter development. First, we find a deep-to-superficial axis, in which superficial tract regions near the cortical surface exhibit greater age-related change than deep tract regions. Second, we demonstrate that the development of superficial tract regions aligns with the cortical hierarchy defined by the sensorimotor-association axis, with tract ends adjacent to sensorimotor cortices maturing earlier than those adjacent to association cortices. These results reveal developmental variation along tracts that conventional tract-average analyses have previously obscured, challenging the implicit assumption that white matter tracts mature uniformly along their length. Such developmental variation along tracts may have functional implications, including mitigating ephaptic coupling in densely packed deep tract regions and tuning neural synchrony through hierarchical development in superficial tract regions - ultimately refining neural transmission in youth.
To evaluate the diagnostic performance of semi-supervised learning models for aggressive prostate cancer detection on MRI compared to fully supervised models trained with additional expert annotations. We used 1500 MRI scans from the PI-CAI challenge training subset. Positive scans had 220 human and 205 AI-generated annotations. The mtU-Net (proposed teacher-student semi-supervised approach) was compared to supervised (trained using only 220 human annotations) and semi-supervised (trained on human and AI-generated annotations) nnU-Net. The 205 AI-annotated scans were manually annotated, and a fully supervised model was trained. External validation was performed on a newly annotated dataset from the PROMIS study (n = 574, 403 lesions) and the Prostate158 dataset (n = 158, 126 lesions). Patient-level performance was evaluated using the area under the curve (AUC) and lesion-level detection (overlap > 0.10) using average precision (AP), along with 95
The Biofluid Biomarkers Best Practices Workgroup of the National Alzheimer's Coordinating Center-Alzheimer's Disease Research Center (ADRC) Biomarker Core Steering Committee was convened to update pre-analytical handling guidelines for biofluid biomarkers, focusing on cerebrospinal fluid (CSF) and blood. We reviewed current literature pertinent to best practices for biomarker studies and surveyed the ADRCs for biomarker analytes, platforms, and protocols used at each center. Across 37 ADRCs, 16 CSF and 28 plasma/serum analytes were reported to be studied at multiple centers. The pre-analytical handling steps and concerns related to each, as supported by empirical studies and expert opinion, were integrated to generate a revised guideline document. The guideline aimed to standardize steps in biospecimen and biomarker analyte collection, storage, and pre-analytical handling across the ADRCs. The 2025 ADRC guidelines represent the current working knowledge on biomarker best practices, providing guidance and harmonized protocols, and promoting robust analysis and reporting of composite data.
