
Objective To investigate the status of plaque control in patients with periodontal disease and its related in-fluencing factors.Methods A total of 350 periodontal disease patients admitted to the Hospital of Stomatological Xi'an Jiao-tong University from May 2018 to May 2022 were selected as the research subjects by the random sampling method.All patients underwent total supragingival oral prophylaxis.The total oral plaque assessment and detection,general oral examination,and a questionnaire survey on oral hygiene behavior will be conducted when patients came back for return visit at 7-10 days after operation.Single factor and multivariate logistic regression analysis were used to analyze the factors affecting the effectiveness of plaque control.Results A total of 350 questionnaires were distributed in this study,and 340 valid questionnaires(97.14%)were collected.The total plaque index of the patient was(68.42±16.93)%;the plaque index in maxillary was significantly lower than that in mandibular(P<0.05);the plaque index in anterior tooth area was significantly lower than that in the poste-rior tooth area(P<0.05);there was no significant difference in plaque index between the left and right halves of the mouth(P>0.05);there was no significant difference in plaque index between zone 1 and zone 2(P>0.05);there was no signifi-cant difference in plaque index between zone 3 and zone 4(P>0.05);the plaque index in zone 3 and zone 4 was significantly higher than that in zone 1 and zone 2,respectively(P<0.05).There was no significance difference in the plaque index be-tween distal and lingual surfaces(P>0.05),while there was significance difference in the plaque index between the other dental surfaces(P<0.05).The results of univariate analysis showed that there were significant difference in plaque index among patients with different genders,educational levels,disease severity,brushing duration,and brushing method(P<0.05);the results of multivariate logistic regression analysis showed that the severity of the disease and brushing method were the factors affecting the plaque index(P<0.05).Conclusion The overall control situation of plaque control in patients with periodontal disease is not ideal.The severity of the disease and the method of brushing teeth are factors affecting the effectiveness of plaque index control.Oral health education should be strengthened to promote plaque control in patients with periodontal disease.
Objective To investigate the diagnostic value of cervical vagus nerve cross-sectional area(CAS)for Parkinson's disease(PD).Methods Thirty patients with PD admitted to the People's Hospital of Zhengzhou University from October 2019 to October 2022 were selected as PD group,25 patients with multiple system atrophy(MSA)admitted to the People's Hospital of Zhengzhou University during the same period were selected as the MSA group,and 30 healthy individuals who underwent physical examination in the People's Hospital of Zhengzhou University during the same period were selected as healthy control group.Cervical vagus CAS of subjects in the three groups were measured by high-resolution ultrasound,and the difference of CAS of cervical vagus nerve was compared among the three groups.The degree of impairment of autonomic nervous function of subjects in the three groups was evaluated by PD autonomic symptom scale(SCOPA-AUT).The diagnostic value of cervical vagus nerve CAS for PD was analyzed by receiver operating characteristic(ROC)curve.Results The CAS of the right cervical vagus nerve of subjects was significantly larger than that of the left in the healthy control group and PD group(P<0.05);there was no significant difference in CAS of bilateral cervical vagus nerve of subjects in the MSA group(P>0.05).The CAS and average CAS of bilateral cervical vagus nerve of subjects in the PD group and MSA group were significantly lower than those in the healthy control group(P<0.01).The CAS of the right vagus nerve of subjects in the MSA group was significantly lower than that in the PD group(P<0.05);there was no significant difference in CAS and the average CAS of the left vagus nerve between the MSA group and the PD group(P>0.05).The total score of SCOPA-AUT and gastrointestinal(GI),cardiovascular(CV),urinary(UR)and sexual(SX)scores of subjects in the PD group and MSA group were significantly higher than those in the healthy control group(P<0.01).The total score of SCOPA-AUT and UR,SX scores of subjects in the MSA group were significantly higher than those in the PD group(P<0.05).There was no significant difference in temperature(TH)and pupil(PU)of subjects among the three groups(P>0.05).Pearson correlation analysis showed that the CAS of cervical vagus nerve of PD patients was not correlated with the total score of SCOPA-AUT and the UR,TH,PU,SX scores(r=-0.143,0.281,0.297,0.265,0.312;P>0.05).The CAS of cervical vagus nerve of PD patients was negatively correlated with GI and CV scores(r=-0.683,-0.373;P<0.05).ROC curve analysis showed that the area under the curve of cervical vagus nerve for diagnosing PD was 0.870(95%confidence interval:0.773-0.966,P<0.05);the critical value was 3.064 mm2,the sensitivity was 96%,and the specificity was 67%.The area under the curve of CAS of cervical vagus nerve in differential diagnosis of PD,MSA was 0.680(95%confidence interval:0.537-0.823,P<0.05).The sensitivity and specificity for the diagnosis of MSA were 68%and 70%when the CAS of the cervical vagus nerve<2.709 mm2.Conclusion The CAS of cervical vagal nerve has high clinical diagnostic value for PD,and it provides a new way to improve the diagnosis rate of PD.
Objective To investigate the effect and mechanism of sodium cantharidate on the proliferation,migration and invasion of esophageal carcinoma EC9706 cells.Methods Esophageal cancer EC9706 cells were randomly divided into blank control group,low-dose sodium cantharidate group,medium-dose sodium cantharidate group,high-dose sodium canthari-date group and cisplatin group.The EC9706 cells in the low-,medium-and high-dose sodium cantharidate groups were given fi-nal mass concentration of 1.0,2.5 and 5.0 mg·L-1 sodium cantharidate intervention,respectively.The EC9706 cells in the cisplatin group were treated with the final mass concentration of 140 mg·L-1 cisplatin,and the cells in the control group was cultured in Dulbecco's modified Eagle's medium.The cell proliferation rate in each group was detected by cell counting reagent-8 method,the mobility of EC9706 cells in each group was detected by scratch test,the invasion rate of EC9706 cells in each group was detected by Transwell method,and the levels of Wnt3a and β-catenin mRNA in EC9706 cells in each group were detected by real-time quantitative polymerase chain reaction method,and the levels of Wnt3a and β-catenin protein in EC9706 cells in each group were detected by Western blot.Results At 24,48 and 72 h of cultivation,the proliferation rate of EC9706 cells in the low-dose sodium cantharidate group,medium-dose sodium cantharidate group,high-dose sodium canthari-date group and cisplatin group was significantly lower than that in the blank control group(P<0.05);the proliferation rate of EC9706 cells in the low-dose sodium cantharidate group and medium-dose of sodium cantharidate group was significantly higher than that in the high-dose sodium cantharidate group and cisplatin group(P<0.05);the proliferation rate of EC9706 cells in the low-dose sodium cantharidate group was significantly higher than that in the medium-dose sodium cantharidate group(P<0.05);there was no significant difference in the proliferation rate of EC9706 cells between the high-dose sodium cantharidate group and cisplatin group(P>0.05);the proliferation rate of EC9706 cells in the low-dose sodium cantharidate group,medium-dose sodium cantharidate group,high-dose sodium cantharidate group and cisplatin group was significantly decreased with the extension of culture time(P<0.05).The mobility rate and invasion rate of EC9706 cells in the low-dose sodium cantharidate group,medium-dose sodium cantharidate group,high-dose sodium cantharidate group and cisplatin group were significantly lower than those in the blank control group(P<0.05);the mobility rate and invasion rate of EC9706 cells in the low-dose sodium cantharidate group and medium-dose sodium cantharidate group were significantly higher than those in the high-dose sodium cantharidate group and cisplatin group(P<0.05);the migration rate and invasion rate of EC9706 cells in the low-dose sodium cantharidate group were significantly higher than those in the medium-dose sodium cantharidate group(P<0.05);there was no significant difference in the mobility rate and invasion rate of EC9706 cells between the high-dose sodium cantharidate group and cisplatin group(P>0.05).The relative expression levels of Wnt3a and β-catenin mRNA and protein in EC9706 cells in the low-dose sodium cantharidate group,medium-dose sodium cantharidate group,high-dose sodium cantharidate group and cisplatin group were significantly lower than those in the blank control group(P<0.05);the relative expression levels of Wnt3a and β-catenin mRNA and protein in EC9706 cells in the low-dose sodium cantharidate group and medium-dose sodium cantharidate group were significantly higher than those in the high-dose sodium cantharidate group and cisplatin group(P<0.05);the relative expressions levels of Wnt3a and β-catenin mRNA and protein in EC9706 cells in the low-dose sodium cantharidate group were significantly higher than those in the medium-dose sodium cantharidate group(P<0.05);there was no significant difference in the relative expression levels of Wnt3a and β-catenin mRNA and protein in EC9706 cells between the high-dose sodium cantharidate group and cisplatin group(P>0.05).Conclusion Sodium cantharidate can significantly inhibit the proliferation,migration and invasion of esophageal carcinoma EC9706 cells,and its mechanism may be related to the inhibition of the Wnt3a/β-catenin pathway.
Diabetic cardiomyopathy is one of the leading causes of heart failure and death in diabetic patients.There is classic evidence of ferroptosis in the development of diabetic cardiomyopathy,which proves that ferroptosis is correlated with the diabetic cardiomyopathy.Oxidative stress caused by excessive production of reactive oxygen species and loss of antioxidant capacity is considered to be the main mechanism leading to diabetic cardiomyopathy.As one of the key regulatory factors of oxi-dative stress,nuclear factor E2-related factor 2(NRF2)and its target gene play an important role in the prevention and treat-ment of diabetic cardiomyopathy.This article summarizes the mechanism of ferroptosis and the pathogenesis of diabetes cardio-myopathy,and expounds the relationship between them,and focuses on the molecular mechanism of NRF2 in the development of diabetic cardiomyopathy and its targeted treatment effect.
Objective To explore the correlation between polarization status of microglia/macrophages(MG/MP)in brain tissue and edema around hematoma in patients with acute cerebral hemorrhage.Methods A total of 52 patients with acute intracerebral hemorrhage admitted to Anyang People's Hospital from December 2020 to November 2022 were selected as the research subjects.All patients underwent craniotomy to remove hematoma,and the normal brain tissue in the cortical area that was not invaded by the hematoma and the fragmented brain tissue around the hematoma(brain tissue around the hematoma)on the surgical pathway were obtained.The expression levels of inflammatory factors such as interleukin(IL)-1β,IL-6,tumor necrosis factor-α(TNF-α),IL-10 and transforming growth factor-β(TGF-β)protein in brain tissue were detected by Western blot.The expression levels of IL-1 β,IL-6,TNF-α,IL-10 and TGF-β mRNA in brain tissue were detected by fluorescence quantitative polymerase chain reaction.The levels of M1-type and M2-type MG/MP in brain tissue was detected by immunofluorescence confocal technique.CT images data of patients before operation were collected and the relative-erihema-tomal edema(r-PHE)was calculated.The patients were divided into high r-PHE group(2.0≤ r-PHE<2.5)and low r-PHE group(1.5<r-PHE<2.0)according to r-PHE.The relative expression of IL-1 β,IL-6,TNF-α,IL-10 and TGF-β mRNA in brain tissue around the hematoma of patients between the high r-PHE group and the low r-PHE group was compared.Results The relative expressions of IL-1 β,IL-6,TNF-α protein and mRNA in brain tissue around the hematoma were significantly higher than those in the normal brain tissues(P<0.05),but there was no significant difference in the relative expressions of IL-10 and TGF-β protein and mRNA between the brain tissue around the hematoma and the normal brain tissue(P>0.05).The levels of M1 type and M2 type MG/MP in the brain tissue around the hematoma were significantly higher than those in normal brain tissue(P<0.05).The relative expressions of IL-1β,IL-6 and TNF-α mRNA in the brain tissue around the hematoma of patients in the high r-PHE group were significantly higher than those in the low r-PHE group(P<0.05),and there was no significant difference in the relative expressions of TGF-β and IL-10 mRNA in the brain tissue around the hematoma of patients between the two groups(P>0.05).Conclusion The levels of pro-inflammatory factors and M1-type MG/MP are increased in the brain tissue around the hematoma in patients with acute cerebral hemorrhage,and the degree of polarization of M1-type MG/MP is consistent with the degree of edema around hematoma after cerebral hemorrhage.
Objective To investigate the effect of microglia inhibitor Pexidartinib on reconsolidation of remote contextual fear memory in mice.Methods Twelve healthy C57BL/6 male mice were randomly divided into experimental group and control group,with 6 mice in each group.The mice in the two groups underwent contextual fear conditioning training in the contextual fear response box to establish the contextual fear models,and the freezing time of mice after each footshock was recorded.After 7 days,the mice in the experimental group were fed with food containing Pexidartinib formulation PLX3397,while the mice in the control group were fed with regular food until the end of behavioral experiment.On the 16th day after contextual fear conditioning training,the mice were put back into the contextual fear box for recalling the fear memory without any stimulation.The mice were taken out after 5 minutes of free exploration,and the freezing time of mice during this period was recorded.At 24 h after the fear memory was recalled,the mice were again placed in the contextual fear box,allowing them to explore freely for 3 minutes,and the freezing time of mice during this period was recorded;the fear response of mice was indicated by the percentage of freezing time.After the behavioral experiment,the mice were anesthetized by intraperitoneal injection of pentobarbital sodium,and three mice from the two groups were taken and rapidly opened the chest to expose their hearts,the perfusion needle was inserted into the left ventricle from the tip of heart and then was perfuse with 40 g·L-1 paraformaldehyde(pH=7.4)which precooled at 4 ℃ until the involuntary convulsion disappeared and the body limbs of mice were stiff.The brain tissue of mice was taken and fixed with paraformaldehyde solution,and then placed in sucrose solution for dehydration.The brain tissue of mice was coated with tissue embedding agent.The number of microglial cells in the hippocampus of mice in the two groups was detected by immunohistochemistry.The remaining mice in the two groups were taken and quickly decapitated to obtain brain tissues,and the hippocampus tissues of two sides were separated on ice.The expressions of phosphorylated bromodomain-containing protein 4(pBRD4),gasdermin D(GSDMD)and mixed lineage kinase domain-like protein(MLKL)in the hippocampus of mice were detected by Western blot.Results In the stage of contextual fear conditioning training,the percentage of freezing time of mice in two groups increased with the increase of the number of footshock(P<0.05),but there was no significant difference in the percentage of freezing time of mice after the first,second,third,fourth and fifth footshock between the two groups(P>0.05).There was no significant difference in the percentage of freezing time of mice between the two groups during recall period(P>0.05).The percentage of freezing time of mice in the experimental group was significantly lower than that in the control group at the phase of memory test(P<0.05).The number of microglia in CA1,CA3 and DG regions of hippocampus of mice in the experimental group was significantly lower than that in the control group(P<0.05).The relative expressions of pBRD4,GSDMD and MLKL in hippocampus of mice in the experimental group were significantly lower than those in the control group(P<0.05).Conclusion Microglia inhibitor Pexidartinib can injury the reconsolidation of remote contextual fear memory,which may be related to its inhibition of microglial cell activation and the down-regulation of the expressions of pBRD4,GSDMD and MLKL.
Objective To explore the application effect of high-functioning group therapy model in female patients with depression.Methods A total of 80 female patients with depression who were hospitalized in the Department of Psychiatry,the Second Affiliated Hospital of Xinxiang Medical University from April 2021 to April 2022 were selected as the research sub-jects.According to the treatment methods,the patients were divided into the observation group and the control group,with 40 patients in each group.The patients in the two groups were treated with routine anti-depressant drugs based on their condition;the patients in the control group received routine nursing interventions based on their condition;the patients in the observation group received high-function group therapy intervention on the basis of the control group;the patients in the two groups were in-tervened for 1 month.Before and one month after the intervention,the levels of depression and anxiety of patients in the two groups were evaluated by the Hamilton depression scale(HAMD)and the Hamilton anxiety scale(HAMA);the quality of life of patients in the two groups was evaluated by the World Health Organization's quality of life scale(WHOQOL-BREF),while the interpersonal relationships of patients in the two groups were evaluated by the interpersonal relationships comprehensive diagnostic scale.Results Before intervention,there was no statistically significant difference in the scores of HAMA,HAMD,WHOQOL-BREF and interpersonal relationship comprehensive diagnostic scale of patients between the two groups(P>0.05).The HAMA and HAMD scores of patients in the two groups after one month of intervention were significantly lower than those before intervention(P<0.05);after one month of intervention,the HAMA and HAMD scores of patients in the observa-tion group were significantly lower than those in the control group(P<0.05).The WHOQOL-BREF scores in the social,psychological,physiological and environmental domains of patients after one month of intervention were significantly higher than those before intervention in the two groups(P<0.05);after one month of intervention,the WHOQOL-BREF scores in the social,psychological,physiological and environmental domains of patients in the observation group were significantly higher than those in the control group(P<0.05).The scores of interpersonal interaction,conversation,social interaction and hetero-sexual interaction and total scores of interpersonal relationship comprehensive diagnosis scale of patients one month of interven-tion were significantly lower than those before intervention in the two groups after(P<0.05);after one month of intervention,the scores of interpersonal interaction,conversation,social interaction and heterosexual interaction and total scores of interper-sonal relationship comprehensive diagnosis scale of patients in the observation group were significantly lower than those in the control group(P<0.05).Conclusion Compared to conventional nursing interventions,the high-functioning group therapy model can significantly reduce the depression and anxiety of female depression patients,improve interpersonal communication skills and their quality of life.
Objective To explore the application effect of modified installing method of high flexion rotating platform prosthesis trial model in double compartment knee arthroplasty.Methods A total of 20 patients(26 knees)with severe knee osteoarthritis admitted to Hebi People's Hospital from January 2021 to April 2022 and required double compartment knee arthroplasty were selected as the research subjects.Patients were divided into the control group(10 knees)and the observation group(16 knees)based on the installing method of high flexion rotating platform prosthesis trial model.The patients in the control group used the conventional installing method of high flexion rotating platform prosthesis test model(placing the tibial side test model,the femoral side test model and finally the pad test model),while patients in the observation group used the modified installing method of high flexion rotating platform prosthesis test model(matching the femoral test model,pad and tibial test model when the patient flexed 60° to 90°,and placing them together into the knee joint osteotomy groove).The intraoperative bleeding volume and surgical time of patients in the two groups were recorded.Before surgery,2 weeks after surgery and 18 months after surgery,the knee joint function of patients was evaluated by the American hospital for special surgery(HSS)scoring system,and the range of motion(ROM)of knee joint of patients was measured by protractor.Results The intraopera-tive bleeding volume of patients in the observation group was significantly less than that in the control group,and the surgical time was significantly shorter than that in the control group(P<0.05).There was no statistically significant difference in preoperative HSS score and ROM of knee joint of patients between the two groups(P>0.05).At 2 weeks and 18 months after surgery,the HSS score and ROM of knee joint of patients in the observation group were significantly higher than those in the control group(P<0.05).Conclusion Compared with the conventional installing method of high flexion rotating platform prosthesis trial method,the modified installing method of high flexion rotating platform prosthesis trial model in double compart-ment knee arthroplasty for patients with severe knee osteoarthritis can effectively improve surgical efficiency,reduce intraopera-tive bleeding,improve knee function and increase ROM of knee joint.
Food intolerance(FI)is a type of adverse food reaction,which can cause discomfort through immune effects,enzyme deficiency,reduced enzyme activity,pharmacological effects and other mechanisms.FI involves all systems of the body and is associated with migraine,inflammatory bowel disease,irritable bowel syndrome,dermatitis etc.,and even involves some rare diseases.The incidence of FI is high,but due to the insidious onset,the complexity of food types,so its diag-nosis is difficult.At present,the pathogenes of FI is unclear,misdiagnosis and mistreatment prevent effective improvement of symptoms of FI patients,which lower the quality of life of patients.Therefore,it is very important to explore the pathogenesis of FI and further research on more simple and effective diagnosis methods and individualized treatment measures for the preven-tion and treatment of FI.This article mainly reviews the research progress on the pathogenesis,diagnosis,and treatment of FI,in order to provide reference for further research on novel diagnostic,preventive,and therapeutic methods for FI.
Objective To observe the effects of different epidural anesthesia regimens on pain degree and postpartum pelvic floor function of primiparas.Methods A total of 80 primiparas who underwent painless epidural anesthesia delivery in Xiangyang Hospital Affiliated to Hubei University of Traditional Chinese Medicine from September 2021 to September 2022 were selected as the research subjects.The primiparas were divided into control group(n=40)and observation group(n=40)according to the anesthesia plan.The primiparas in the control group received epidural anesthesia with ropivacaine combined with physiological saline,while the primiparas in the observation group received epidural anesthesia with ropivacaine combined with sufentanil.The rate of natural delivery,rate of cesarean section,rate of vaginal delivery and the duration of each labor of primiparas were compared between the two groups;the analgesic effect of primiparas in the two groups was evaluated by pain visual analogue scale(VAS)at 5,60 min after analgesia,full opening of the cervix and postpartum 24 hours;the asphyxia sta-tus of newborns in the two groups was evaluated by neonatal Apgar score at 1,5 min after birth;the complications of nausea and vomiting,cervical or perineal injuries,urinary retention of puerperas and neonatal asphyxia were recorded and the total inci-dence of complications was compared between the two groups.At 6-8 weeks after delivery,pelvic floor functional indicators such as fiber muscle strength,pelvic floor muscle closure contraction force,and static tension of pelvic floor muscles of postpar-tum women in the two groups were measured by biological stimulus feedback instrument.Results The natural delivery rate,vaginal delivery rate and cesarean section rate of primiparas in the control group was 72.5%(29/40),7.5%(3/40),20.0%(8/40),respectively;the natural delivery rate,vaginal delivery rate and cesarean section rate of primiparas in the observation group was 90.0%(36/40),5.0%(2/40)and 5.0%(2/40),respectively.The natural delivery rate of primiparas in the ob-servation group was significantly higher than that in the control group,and the cesarean section rate was significantly lower than that in the control group(P<0.05);there was no significant difference in the vaginal delivery rate of primiparas between the two groups(P>0.05).The first and second stages of labor of primiparas in the observation group were significantly shorter than those in the control group(P<0.05),and there was no significant difference in the third stage of labor of primiparas be-tween the two groups(P>0.05).The VAS score of primiparas in the observation group was significantly lower than that in the control group at 5,60 min after analgesia,full opening of the cervix and postpartum 24 hours(P<0.05).There was no signifi-cant difference in the Apgar score of newborns between the two groups at 1,5 min after birth(P>0.05).The total incidence of complications in the control group and the observation group was 42.50%(17/40)and 20.00%(8/40),respectively;the total incidence of complications in the observation group was significantly lower than that in the control group(x2=4.713,P<0.05).The fiber muscle strength and pelvic floor muscle closure contraction force of postpartum women in the observation group were significantly higher than those in the control group,the static tension of pelvic floor muscles was significantly lower than that in the control group(P<0.05).Conclusion Sufentanil combined with ropivacaine epidural anesthesia can improve the analgesic effect of primiparas during childbirth,reduce the cesarean section rate,shorten the labor process,and facilitate the recovery of pelvic floor function in primiparas,reduce maternal and infant complications.
Objective To investigate the predictive value of serum matrix metalloproteinase-3(MMP-3)and cystatin C(Cys C)levels and their combination for adverse pregnancy outcomes in patients with early-onset preeclampsia.Methods Thirty-one patients with early-onset preeclampsia admitted to the Department of Obstetrics in the First Affiliated Hospital of Bengbu Medical College from November 2021 to August 2022 were selected as the early-onset preeclampsia group,and 44 pregnant women with normal pregnancies who came to our hospital for labor and delivery during the same period were selected as the normal pregnancy group.Patients in the early-onset preeclampsia group were categorized into good pregnancy outcome group(n=13)and poor pregnancy outcome group(n=18)based on whether the pregnancy outcome was accompanied by a neonatal Apgar score of ≤ 7,intrauterine distress,fetal growth restriction,eclampsia seizures,and placental abruption.The levels of serum MMP-3 and cystatin C were detected by enzyme linked immunosorbent assay.The relationship between serum MMP-3,Cys C level and early-onset preeclampsia and pregnancy outcome were analyzed.The value of seurm MMP-3,cystatin C level and their combination in predicting adverse pregnancy outcomes in patients with early-onset preeclampsia were evaluated by using receiver operating characteristic.Results The levels of serum MMP3 and Cys C in the early-onset preeclampsia group were higher than those in the normal pregnancy group(P<0.05).The levels of serum MMP-3 and Cys C in the poor pregnancy outcome group were higher than those in the good pregnancy outcome group(P<0.05).The area unde the curve(AUC),sensitivity,specificity,Youden index of serum MMP-3 in predicting adverse pregnancy outcome in patients with early-onset preeclampsia were 0.885,83.3%,84.6%,0.679,respectively.The AUC,sensitivity,specificity,Youden index of serum Cys C in predicting adverse pregnancy outcome in patients with early-onset preeclampsia were 0.859,83.3%,76.9%,0.603,respectively.The AUC,sensitivity,specificity,Youden index of combined detection of serum MMP-3 and Cys C in predicting adverse pregnancy outcome in patients with early-onset preeclampsia were 0.923,72.2%,100%,0.722,respectively.Conclusion The serum MMP-3 and Cys C levels are overloaded in patients with early-onset preeclampsia,and the serum MMP-3 and serum Cys C levels are related to the pregnancy outcome of patients with early-onset preeclampsia.The combined detection of the two indexes can predict the adverse pregnancy outcome of early-onset preeclampsia to some extent.
Objective To investigate the protective effect and mechanism of piperazine ferulate(PF)on acute kidney injury of rats with sepsis.Methods Twenty-four male Sprague Dawley rats were randomly divided into sham operation group,model group,low-dose PF group and high-dose PF group,with 6 rats in each group.The rats in the low-dose PF group and high-dose PF group were given 50 and 100 mg·kg-1 PF suspension by gavage every day for 7 days,respectively.The rats in the sham operation group and model group were given 9 g·L-1 sodium chloride solution(0.1 mL·kg-1)by gavage every day for 7 days.The rats in the sham operation group underwent cecal exploration and closed their stomachs without undergoing cecal ligation and perforation surgery.The rats in the model group,low-dose PF group and high-dose PF group underwent cecal ligation and perforation surgery to prepare the acute kidney injury models.At 24 hours after modeling,carotid artery blood of rats in each group was collected and the serum was separated.The levels of serum creatinine(Scr)and blood urea nitrogen(BUN)were measured by biochemical methods,and the levels of neutrophil gelatinase related lipid transport protein(NGAL),interleukin-6(IL-6),and procalcitonin(PCT)were measured by enzyme-linked immunosorbent assay.The rats in each group were executed by decapitation,then the kidneys were taken.The pathological changes of the kidney tissues were observed by hematoxylin-eosin(HE)staining.The relative expression levels of nuclear factor-κB(NF-κB),Toll-like receptor 4(TLR4)and NOD-like receptor thermal protein domain associated protein 3(NLRP3)in renal tissues were detected by Western blot method.Results The levels of Scr,BUN and serum NGAL,IL-6 and PCT of rats in the model group and low-dose PF group were significantly higher than those in the sham operation group(P<0.05).The levels of Scr and serum PCT of rats in the high-dose PF group were significantly higher than those in the sham operation group(P<0.05);there was no statistically significant difference in the levels of BUN and serum NGAL and IL-6 of rats between the high-dose PF group and the sham operation group(P>0.05).The levels of Scr,BUN and serum NGAL,IL-6 and PCT of rats in the low-dose PF group and high-dose PF group were significantly lower than those in the model group(P<0.05).The levels of Scr,BUN and serum NGAL,IL-6 and PCT of rats in the high-dose PF group were significantly lower than those in the low-dose PF group(P<0.05).HE staining showed that there were no significant abnormal changes in the renal tissues of rats in the sham operation group.The renal tubular epithelial cells of rats in the model group showed significant swelling and degeneration,with a large amountof red staining in the cytoplasm;the renal tubular had congestion,degeneration,and necrosis,with a large number of inflammatory cells infiltrating the lumen,and cell proliferation in the mesangial-capillaries space.In the low dose PF group,some renal tubular epithelial cells of rats showed mild to moderate swelling,some renal tubular epithelial cells showed degenera-tion and necrosis,and no significant infiltration of inflammatory cells was observed.The renal tubular epithelial cells of rats in the high-dose PF group showed mild swelling,without significant degeneration,necrosis and inflammatory cell infiltration.The levels of TLR4,NF-κB and NLRP3 protein in renal tissues of rats in the model group and low-dose PF group were significantly higher than those in the sham operation group(P<0.05).The level of TLR4 protein in renal tissues of rats in the high-dose PF group was significantly higher than that in the sham operation group(P<0.05);there was no statistically significant difference in the levels of NF-κB and NLRP3 protein in the renal tissues of rats between the high-dose PF group and sham operation group(P>0.05).The levels of TLR4 and NLRP3 proteins in the renal tissues of rats in the low-dose PF group and high-dose PF group were significantly lower than those in the model group(P<0.05);the level of NF-κB in renal tissues of rats in the high-dose PF group was significantly lower than that in the model group(P<0.05);there was no statistically significant difference in the level of NF-κB in renal tissues of rats between the low-dose PF group and model group(P>0.05).The levels of TLR4 and NLRP3 proteins in tissues of rats in the high-dose PF group were significantly lower than those in the low-dose PF group(P<0.05);there was no statistically significant difference in the level of NF-κB in renal tissues of rats between the high-dose PF group and low-dose PF group(P>0.05).Conclusion PF intervention can effectively reduce the levels of Scr,BUN and serum NGAL,IL-6 and PCT of rats with septic acute kidney injury,and play a certain protective role on the kidneys of rats with septic acute kidney injury;its mechanism may be related to inhibiting the expression levels of TLR4,NF-κB and NLRP3,thereby regulating TLR4/NF-κB/NLRP3 signaling pathway.
Objective To explore the correlation between platelet endothelial aggregation receptor-1(PEAR1)gene polymorphisms and vascular access infections in intermittent hemodialysis(IHD)patients.Methods Totally 105 patients who received IHD treatment in Shaanxi Provincial People's Hospital from January 2019 to January 2022 were selected as the obser-vation group,and 50 healthy people who were admitted to the hospital for physical examination at the same period were selected as the control group,and PEAR1 gene polymorphisms of the subjects in the two groups were tested by polymerase chain reaction.In addition,the patients in the observation group underwent imaging,serology,and pathogenesis,and they were divided into infected group and uninfected group according to whether vascular access infections occurred,and the clinical data of patients between the two groups were compared,and the correlation between PEAR1 gene polymorphisms and vascular access infections in IHD patients was analyzed by using univariate and multifactorial logsitic regression.Results There were two alleles G and A at the PEAR1 gene locus of patients in the observation group and control group;and three genotypes were detected,including wild-type(GG),mutant heterozygous(GA),and mutant pure(AA).The frequency of the GG genotype and the frequency of the G allele genotype of the patients in the observation group were higher than those in the control group(P<0.05);the allele frequencies of the gene loci of the subjects in the observation group and the control group were in accordance with the Hardy-Weinberg equilibrium(P>0.05).Univariate analysis showed that there was no statistically significant difference in the sex ratio,body temperature,body mass index,blood pressure,triglyceride,total cholesterol,low-density lipoprotein,high-density lipoprotein,frequency of the GA and AA genotypes,percentage of history of hyperlipidemia,percentage of history of cigarette smoking,and percentage of history of alcohol consumption of patients between the infected group and uninfected group(P>0.05).The age the percentage of dialysis duration>3 years,the percentage of dialysis frequency ≥3 times in 1 week,the frequency of PEAR1 GG genotype,the frequency of PEAR1 G allele,the troponin T level,leukocyte level,platelet count level,hematocrit level,procalcitonin level,neutrophil/lymphocyte ratio,platelet/lymphocyte ratio,hypertension history and diabetes history of patients in the uninfected group were all lower than those in the infected group,while the fre-quency of PEAR1 A allele was higher than that in the infected group(P<0.05).Logistic regression analysis showed that long dialysis duration,older age and high dialysis frequency,high neutrophil/lymphocyte ratio,high platelet/lymphocyte ratio,and PE AR1 G allele were risk factors for developing vascular access infections in patients with IHD(P<0.05).Conclusion IHD patients have PEAR1 genotype expression abnormality,in which the risk of vascular access infection in patients with PEAR1 genotype GG is higher,which can be used as an effective factor for the early prediction of vascular access infection in hemodialysis patients.
Objective To investigate the expression levels of microRNA(miR)-191,miR-23a,and miR-145 in plasma of patients with prostate cancer and their correlation with prognosis.Methods Sixty prostate cancer patients admitted to Zhumadian Central Hospital from December 2019 to December 2021 were selected as the observation group,and 60 healthy subjects who underwent physical examinations during the same period were selected as the control group.The expression levels of miR-191,miR-23a and miR-145 in plasma of subjects in the two groups were measured by real-time fluorescence quantita-tive polymerase chain reaction.The patients in the observation group were divided into poor prognosis group(n=11)and good prognosis group(n=49)according to their prognosis.The expression levels of miR-191,miR-23a,and miR-145 in plasma of prostate cancer patients were compared before and after treatment.The general data of patients was compared between the poor prognosis group and the good prognosis group.The receiver operator characteristic curve was drawed to analyse the predictive value of plasma miR-191,miR-23a,and miR-145 levels on prognosis.Results The relative expression levels of miR-191 and miR-23 a in plasma of patients in the observation group were significantly higher than those in the control group,while the relative expression level of miR-145 was significantly lower than that in the control group(P<0.05).The relative expression levels of miR-191 and miR-23a of patients in the observation after treatment were significantly lower than those before treat-ment,while the relative expression level of miR-145 was significantly higher than that before treatment(P<0.05).There was no significant difference in age,body mass index,smoking history,alcohol consumption history,and prostate volume of patients between the poor prognosis group and the good prognosis group(P>0.05);there was significant differences in TNM staging,Gleason score,serum prostate specific antigen level and relative expressions of miR-191,miR-23a,miR-145 in plasma of patients between the two groups(P<0.05).Plasma miR-191,miR-23a and miR-145 levels were influence factor for prognosis in patients with prostate cancer(P<0.05).There was predictive value of plasma miR-191,miR-23a and miR-145 for prognosis of prestate cancer patients;the combined predictive value of plasma miR-191,miR-23a and miR-145 for the prognosis of prostate cancer patients was significantly higher than that of plasma miR-191,miR-23a and miR-145 alone for the prognosis of prostate cancer patients(P<0.05).Conclusion The expression of miR-191 and miR-23a in plasma increases and the expression of miR-145 in plasma decreases in patients with prostate cancer.The combined detection of plasma miR-191,miR-23a and miR-145 has higher predictive value for the prognosis of prostate cancer.
With the popularization of high-resolution computed tomography and the enhancement of public awareness for health physical examination,the detection rate of small pulmonary nodule(SPN)increases year by year.SPN is often the early manifestations of lung cancer,which need to be surgical resection as soon as possible.Video-assisted thoracoscopic surgery has become the standard operation for the treatment of SPN because of its small trauma and quick recovery.However,SPN is difficult to be accurately located because of its small size,deep location and soft texture.Therefore,how to accurately and quickly locate SPN has become a hot research topic for diagnosis and treatment of SPN in recent years.This article reviews the research progress of preoperative and intraoperative localization methods for SPN.
Stroke is a common nervous system disease with high incidence rate,mortality rate and disability rate.Hydrogen rich water is a type of drinking water containing high concentrations of hydrogen gas,which can be used to treat stroke.Hydrogen rich water mainly plays a role by reducing inflammatory reactions,cell apoptosis,oxidative stress reactions,etc.This article reviews the research progress on the therapeutic effect and mechanism of hydrogen rich water on stroke,in order to provide insights for the treatment plan of stroke.
Objective To investigate the efficacy and safety of fluzopril in the treatment of platinum-sensitive recurrent epithelial ovarian cancer.Methods A total of 107 patients with platinum-sensitive recurrent epithelial ovarian cancer admitted to the First Affiliated Hospital of Henan University of Science and Technology from January 2019 to December 2020 were selected as the subjects.According to treatment methods,the patients were divided into control group(n=50)and observation group(n=57).The patients in the control group received a first-line chemotherapy regimen of paclitaxel combined with platinum:on the first day,intravenous infusion with paclitaxel injection 135 mg·m-2 was administered;on day 1-3,intravenous drip with cisplatin 50-60 mg·m-2 was administered;21 days was one chemotherapy cycle.On the basis of the treatment in the control group,the patients in the observation group were given fluzoparide capsules orally,150 mg each time,twice a day,and the treatment continued until disease progression or unacceptable toxic reactions occurred;21 days was one chemotherapy cycle.The patients in both groups received three consecutive chemotherapy cycles.The clinical efficacy,prognosis within 2 years after chemotherapy,Karnofsky Performance Status(KPS)score before and after chemotherapy,and incidence of adverse reactions during chemotherapy of patients between the two groups were compared.Results After 3 cycles of chemo-therapy,the disease control rate and objective remission rate of patients in the observation group were significantly higher than those in the control group(x2=5.420,4.220;P<0.05).Following up to 24 months,the progression free survival of patients in the observation group was significantly longer than that in the control group(t=6.702,P<0.05);there was no statistically significant difference in 1-year survival rate of patients between the two groups(x2=0.415,P>0.05);the 2-year survival rate of patients in the observation group was significantly higher than that in the control group(x2=5.420,P<0.05).Before chemotherapy,there was no statistically significant difference in KPS scores of patients between the two groups(t=0.537,P>0.05);the KPS scores of patients in the two groups after three cycles of chemotherapy were significantly higher than those before chemotherapy(t=5.604,9.378;P<0.05);after three cycles of chemotherapy,the KPS score of patients in the observation group was significantly higher than that in the control group(t=2.608,P<0.05).The patients in both groups experienced hematological and non-hematological adverse reactions during chemotherapy;the main hematological adverse reactions was bone marrow suppression,most of which were Ⅲ ° and Ⅳ ° adverse reactions;the non-hematological adverse reactions included alo-pecia,gastrointestinal reactions,and liver and kidney dysfunction,most of which were Ⅰ°and Ⅱ°adverse reactions.There were no chemotherapy related deaths of patients in both groups.The incidence rates of anemia,thrombocytopenia,neutropenia,leuko-penia,lymphopenia,nausea,vomiting,fatigue,loss of appetite,hair loss,elevated serum creatinine,and elevated alanine amino-transferase(ALT)of patients during chemotherapy in the control group were 68.00%(34/50),72.00%(36/50),58.00%(29/50),68.00%(34/50),22.00%(11/50),26.00%(13/50),24.00%(12/50),46.00%(23/50),30.00%(15/50),50.00%(25/50),20.00%(10/50),10.00%(5/50),respectively;the incidence rates of anemia,thrombocytopenia,neutropenia,leukopenia,lymphopenia,nausea,vomiting,fatigue,loss of appetite,hair loss,elevated serum creatinine,and elevated ALT of patients during chemotherapy in the observation group were 61.40%(35/57),63.16%(36/57),49.12%(28/57),52.63%(30/57),21.05%(12/57),22.81%(13/57),24.56%(14/57),42.11%(24/57),29.82%(17/57),47.37%(27/57),21.05%(12/57),10.53%(6/57),respectively.There was no statistically significant difference in the incidences of anemia,thrombocytopenia,neutropenia,leukopenia,lymphopenia,nausea,vomiting,fatigue,loss of appetite,hair loss,elevated serum creatinine,and elevated ALT of patients during chemotherapy between the control group and the observation group(x2=0.047,0.000,0.041,0.694,0.056,0.000,0.208,0.041,0.184,0.160,0.233,0.102;P>0.05).Conclusion For patients with platinum-sensitive recurrent ovarian cancer,the combination of paclitaxel and platinum chemo-therapy combined with fluzopril can effectively improve the anti-tumor effect,prolong the progression-free survival,improve survival rate and quality of life,and the adverse reactions are controllable.
Sarcopenia,also known as"oligomyositis""hypomyositis"and"muscle wasting",is an age-related geriatric syndrome characterized by decreased muscle mass,muscle strength and function decline.It can increase the risk of adverse out-comes such as physical disability,poor quality of life,and death.The pathogenic factors of sarcopenia mainly include older age,diseases,lack of exercise,malnutrition,etc.As one of the key factors affecting sarcopenia,the nutritional status of the body play an important role in the development of sarcopenia.Therefore,this paper reviews the role of nutrition in sarcopenia and nutritional interventions for patients with common chronic diseases combined with sarcopenia,aiming to provide a basis for the developing more scientific and rationalized nutritional intervention program for elderly patients with sarcopenia.
目的 探讨肿瘤相关成纤维细胞(CAFs)外泌体对肺腺癌细胞 A549 增殖、迁移、侵袭的影响及机制.方法 取洛阳市中心医院胸外科收治的肺腺癌患者的癌组织和癌旁组织标本,并分离培养原代CAFs和正常成纤维细胞(NFs);收集CAFs和NFs细胞培养上清液,使用差速离心法提取CAFs外泌体和NFs外泌体.取对数生长期的A549 细胞,随机分为对照组、NFs外泌体组、CAFs外泌体组,NFs外泌体组、CAFs外泌体组细胞分别加入NFs外泌体、CAFs外泌体孵育,对照组细胞常规培养.采用细胞计数试剂盒-8(CCK-8)实验检测细胞增殖能力,细胞划痕实验检测细胞迁移能力,Transwell 小室实验检测细胞侵袭能力,Western blot 法检测细胞中磷酸酯酶与张力蛋白同源物(PTEN)/磷脂酰肌醇 3 激酶(PI3K)/蛋白激酶 B(Akt)通路相关蛋白 PTEN、PI3K、磷酸化磷脂酰肌醇 3 激酶(p-PI3K)、Akt、磷酸化蛋白激酶 B(p-Akt)及上皮-间质转化(EMT)相关蛋白上皮钙黏素(E-cad)、神经钙黏素(N-cad)、波形蛋白(VIM)表达.采用随机数字表法将 15 只裸鼠分为A549 裸鼠组、NFs外泌体裸鼠组、CAFs外泌体裸鼠组,每组5 只,将对照组、NFs外泌体组、CAFs外泌体组细胞分别移植到A549 裸鼠组、NFs外泌体裸鼠组、CAFs外泌体裸鼠组小鼠左侧腋下,移植6 周内每周测量小鼠肿瘤体积.结果 培养24h时,3 组细胞的增殖能力比较差异无统计学意义(P>0.05);培养48、72 h时,CAFs外泌体组细胞的增殖能力显著高于NFs外泌体组和对照组(P<0.05);培养48、72 h时,对照组与NFs外泌体组细胞的增殖能力比较差异无统计学意义(P>0.05).培养24、48 h时,CAFs外泌体组细胞的迁移能力显著高于NFs外泌体组和对照组(P<0.05);培养24、48 h时,NFs外泌体组与对照组细胞的迁移能力比较差异无统计学意义(P>0.05).CAFs外泌体组侵袭细胞数显著多于NFs外泌体组和对照组(P<0.05);NFs外泌体组与对照组的侵袭细胞数比较差异无统计学意义(P>0.05).CAFs外泌体组细胞中N-cad、VIM蛋白相对表达量显著高于对照组和NFs外泌体组(P<0.05),E-cad蛋白相对表达量显著低于对照组和NFs外泌体组(P<0.05);NFs 外泌体组与对照组细胞中 N-cad、VIM、E-cad 蛋白相对表达量比较差异无统计学意义(P>0.05).CAFs外泌体组细胞中PTEN 蛋白相对表达量显著低于对照组和NFs外泌体组(P<0.05),p-PI3K/PI3K、p-Akt/Akt值显著高于对照组和NFs外泌体组(P<0.05);NFs外泌体组与对照组细胞中PTEN 蛋白相对表达量及p-PI3K/PI3K、p-Akt/Akt值比较差异无统计学意义(P>0.05).移植第 1 周,3 组小鼠的肿瘤体积比较差异无统计学意义(P>0.05);移植第2~6 周,CAFs外泌体裸鼠组小鼠的肿瘤体积显著大于A549 裸鼠组和NFs外泌体裸鼠组(P<0.05);移植第2~6 周,NFs外泌体裸鼠组与A549 裸鼠组小鼠的肿瘤体积比较差异无统计学意义(P>0.05).结论 CAFs外泌体可促进肺腺癌细胞的增殖、迁移和侵袭,在小鼠体内促进肺腺癌的生长,其机制可能与EMT和PTEN/PI3K/Akt信号通路有关.
目的 探讨巴戟天对骨关节炎小鼠软骨细胞自噬、凋亡的影响及机制.方法 将40 只健康C57BL/6 雄性小鼠随机分为对照组、模型组、低剂量巴戟天组、中剂量巴戟天组及高剂量巴戟天组,每组 8 只.模型组和低、中、高剂量巴戟天组小鼠建立骨关节炎模型,随后模型组小鼠每日给予 1mL生理盐水灌胃,低、中、高剂量巴戟天组小鼠每日分别将 100、200、400 mg·kg-1 巴戟天溶于 1mL生理盐水灌胃,均持续4 周;对照组小鼠制备骨关节炎假模型,随后每日给予 1mL生理盐水灌胃,持续4 周.处死各组小鼠,无菌条件下提取右侧膝关节软骨组织,并制备软骨细胞悬液.原位末端转移酶标记染色检测各组小鼠软骨组织中软骨细胞凋亡率,流式细胞术检测各组小鼠软骨细胞悬液中软骨细胞的凋亡情况,Western blot法检测各组小鼠软骨细胞悬液中磷酸化蛋白激酶B(p-Akt)、磷酸化磷脂酰肌醇-3-激酶(p-PI3K)、自噬效应蛋白 1(Beclin1)、人微管相关蛋白轻链3B(LC3B)蛋白表达,实时荧光定量聚合酶链反应法检测各组小鼠软骨细胞悬液中p-PI3K、p-Akt mRNA的表达.结果 模型组和低、中、高剂量巴戟天组小鼠软骨组织中软骨细胞凋亡率显著高于对照组(P<0.05);低、中、高剂量巴戟天组小鼠软骨组织中软骨细胞凋亡率显著低于模型组(P<0.05);中、高剂量巴戟天组小鼠软骨组织中软骨细胞凋亡率显著低于低剂量巴戟天组(P<0.05);高剂量巴戟天组小鼠软骨组织中软骨细胞凋亡率显著低于中剂量巴戟天组(P<0.05).模型组和低、中、高剂量巴戟天组小鼠软骨细胞悬液中软骨细胞凋亡率显著高于对照组(P<0.05);低、中、高剂量巴戟天组小鼠软骨细胞悬液中软骨细胞凋亡率显著低于模型组(P<0.05);中、高剂量巴戟天组小鼠软骨细胞悬液中软骨细胞凋亡率显著低于低剂量巴戟天组(P<0.05);高剂量巴戟天组小鼠软骨细胞悬液中软骨细胞凋亡率显著低于中剂量巴戟天组(P<0.05).模型组和低、中剂量巴戟天组小鼠软骨细胞悬液中Beclin1、LC3B、p-PI3K、p-Akt蛋白相对表达量及p-PI3K mRNA、p-Akt mRNA相对表达量显著低于对照组(P<0.05);高剂量巴戟天组与对照组小鼠软骨细胞悬液中Beclin1、LC3B、p-PI3K、p-Akt蛋白相对表达量及p-PI3K mRNA、p-Akt mRNA相对表达量比较差异无统计学意义(P>0.05);低、中、高剂量巴戟天组小鼠软骨细胞悬液中Beclin1、LC3B、p-PI3K、p-Akt蛋白相对表达量及p-PI3K mRNA、p-Akt mRNA相对表达量显著高于模型组(P<0.05);中、高剂量巴戟天组小鼠软骨细胞悬液中Beclin1、LC3B、p-PI3K、p-Akt蛋白相对表达量及p-PI3K mRNA、p-Akt mRNA相对表达量显著高于低剂量巴戟天组(P<0.05);高剂量巴戟天组小鼠软骨细胞悬液中Beclin1、LC3B、p-PI3K、p-Akt蛋白相对表达量及 p-PI3K mRNA、p-Akt mRNA 相对表达量显著高于中剂量巴戟天组(P<0.05).结论 巴戟天能通过调控PI3K/Akt信号通路促进骨关节炎小鼠软骨细胞的自噬,抑制软骨细胞凋亡.
