
Sika deer Insulin Growth Factor(IGF-1) mature peptide gene was expressed and purified by prokary-otic expression vector p ET-30 a in this study. Recombinant IGF-1 mature peptide protein from sika deer was ob-tained after splitting decomposition of hydroxylamine and protein renaturation. Chicken embryo fibroblast cell and chicken embryo kidney cell proliferation was determined by MTT assay after recombinant IGF-1 mature pep-tide treatment. The results showed that recombinant IGF-1 mature peptide promoted the proliferation of CEF cell indifferent concentrations. Among them, the most significant impact on CEF cell proliferation was at 48 h, and the finalconcentration was 200 ng/m L. But the effect of recombinant IGF-1 on CEK cell was not significant in different con-centrations and different treatment time. Our findings indicated that IGF-1 mature peptide after renaturation promoted chicken embryo fibroblast proliferation significantly, but its influence on chicken embryo kidney cell proliferation was not significant.
In order to explore the prevention and cure function of microbial source fungicide to cucumber pow-dery mildew, chemicals and water treatment were used as control, a preparation of chitinase microbial sources asmain component was made to prevent and control cucumber powdery mildew. Incidence of powdery mildew of cu-cumber, root activity, and a series of biological properties and quality were determined. Through a microscope, itwas found that the preparation could inhibit fungus reproduction by destruction of powdery mildew pathogen sporescell walls. The plant test showed that incidence of powdery mildew was 8.77% in microbial preparation treatment.The control effect was 89.32%, which was far higher than controls. The yield increased by 15.7%. Quality indexsuch as total amino acid content and root vigor were significantly higher than the control. The research showed thatthe preparation could effectively reduce the incidence of powdery mildew of cucumber and promote plant growth atthe same time. The preparation has potential application value.
In order to understand effects of straws returned into field on environmental effect of soil, a long-term located experiment about returned straws were designed since 2000 to select the best methods of straws re-turned into field. Soil physical and chemical properties of straws returned into field were analyzed. The resultsshowed that soil compactions of every treatment were increased with increase of depth, the maximum is at the depthof 45 cm. In addition, soil compaction of low layer was higher than upper, and treatments of straws returned into fieldwere lower than conventional tillage. Three-phase ration of soil of 20-40 cm of treatments of straws returned intofield were more rational than conventional tillage. The infiltration capacity of treatment of straws returned into fieldwas improved because of increase of porosities in soil. Moreover, soil nutrients were increased significantly, soil or-ganic matter treatments of returning straws into field compared of conventional tillage and the range of increase werefrom 0.26 to 0.74 g/kg.
In the study, a millet variety named‘Fenghonggu'was used for material and the control effects of 5herbicides on weeds in the area were studied. The results suggested that different herbicides have different degreesof inhibition to weed growth. The best results on control effect were 200 g/667m~210 day after 44% Monosulfuronplus Propazine WP spraying and 25 day after 150 g/667m~2 of Atrazine spraying. Compared with other drugs, effectof Atrazine on plant height was more significant. The millet yield were not significant different among spraying of 2,4-D Butylate, 44% Monosulfuron plus Propazine WP and Trifluralin at 150 g/667m~2, but the millet yield whichsprayed of 150 g/667m~2 of 2,4-D Butylate was significant higher than that of spraying of 150g/667m~2 of Atrazine.
After 40 cm and 30 cm deep scarification, photosynthetic characteristics of Zhengdan 958 andTongke 1 were both influenced to different degrees. Leaf area index increased significantly and leaf net photosynthet-ic rate of different positions were also significantly improved. Leaf transpiration rate of different positions under the40 cm deep scarification increased significantly, stomatal conductance of upper and lower leaves of Zhengdan 958 increased significantly under the 40 cm deep scarification treatment while ear leaf intercellular CO2 concentration ofTongke 1 reduced significantly under the 40 cm deep scarification treatment. Yields of the two varieties of differentyear both improved significantly, which illustrated deep scarification promoted the photosynthesis, increased yieldof maize, especially for 40 cm deep scarification treatment.
The study on suitable pregerminating conditions to obtain longer sprouts was conducted through set-ting different temperature and light condition treatment, using minitubers about 40 g of‘Kexin 18u0027, one of majorcultivars in Heilongjiang Province as materials. The results showed that seed potatoes removed from storage cellarshould be placed into indoor of 24℃ under dark and wet conditions to accelerate germination. Then, when the lengthof most sprouts was more than 2.5 cm, seed potatoes was moved to the scattered light conditions for 5 to 10 days.
以稻花香2号为试验材料,基肥采用隔年腐熟的农家肥,追肥使用石家庄希望肥业科技有限公司生产的商品有机肥.在常规施肥(翻地前施有机肥15 000 kg/hm2)的基础上,分别在分蘖初期(6月8日)、分蘖高峰期(6月23日)、孕穗期(7月8日)、齐穗期(8月5日)追施1875 kg/hm2商品有机肥.试验结果表明:分蘖高峰期~齐穗期追施有机肥,可以增加产量4.33%~12.17%.分蘖高峰期追肥单穴平均穗数比对照增加12.00%;分蘖高峰期和孕穗期追肥穗粒数比对照增加17.00%、19.00%;孕穗期和齐穗期追肥饱满粒率比对照提高2.60%、6.53%.各生育时期追施有机肥的作用表现为:分蘖高峰期追肥可以提高有效分蘖数量;分蘖高峰期和孕穗期追肥可以提高每穗平均粒数;孕穗期和齐穗期追肥可以提高混合千粒重和饱满粒率.各生育阶段追施有机肥对稻米品质无明显的不良影响,并且还有助于提高精米率、整精米率,并随着施肥期的后延,垩白粒率也有所降低.
Effect of different planting density and nitrogen application on spring corn yield and biomass produc-tion was studied in the paper. Results indicated that under the middle-high density, the output realized mainlythrough the harvest ear of number, and grain number per ear was key factor affected the yield. Nitrogen insufficientand excessive caused the abortion occurrence and bald-tip ears increased. Seed development was promoted and thenumber of grains per ear increased at moderate nitrogen of 220 kg/hm2, so the yield increased.
Safety is the most basic requirements for probiotics. The safety of newly selected probiotic strainsmust be totally evaluated before its commercial production. In this paper, Bacillus subtilis JAASB4, a newly separat?ed and identified strain, was assessed via the oral acute toxicity and translocation. The results showed that there wasno obvious abnormal impact on indicators of mice, such as weight, behavior, activity, diet, stool and other signs ofdirty body. There were no significant difference in liver, spleen ratio between the experimental group and controlgroup. There is no strain translocation also. Therefore, initial proof showed that Bacillus subtilis JAASB4 was safe.
Effect of light intensity, light quality, seed soaking temperature, pH, salinity, metals and drought onFlaveria bidentis seeds germination rate were examined. The results showed that the germination rate was affectedlittle by light intensity and light quality. The germination percentage reduced as the soaking temperature rose. Noseed emerged when the pH value was lower than 6 or higher than 8. The germination rate decreased gradually withthe increasing of NaCl concentration, which was 43.77% when NaCl concentration was 0.20 mol/L. Fe2+and Mn2+re-duced the germination rate significantly, while Cu2+and Zn2+had strong inhabitation effect on the root growth of F. bidentis seedlings but did not change the germination percentage. The germination percentage reduced as the water os-mosis potential decreased. Cold treatment reduced the seed germination significantly. The inhabitation effect of her-bicides on seed germination of F. bidentis was in such order: paraquat acetochlor atrazine·acetochlor tribenu-ron methyl. In general, the germination of F. bidentis seeds could be affected by temperature, heavy metals, pH val-ue, NaCl, drought, cold and herbicides to some extent.
In recent years, corn planting area in Heilongjiang Province is continuously increasing, the weedcontrol in corn field become the most important link in agricultural production. If the control effect is poor, themaize yield will be seriously affected. In this experiment, several kinds of herbicides were used to deal with theweeds in corn field. The results showed that the control effect of Mesotrione to barnyard grass was better than Nico-sulfuron, and it could be up to 100%. The resistance of barnyard grass to Nicosulfuron in Zhaodong corn field washigher than that in Minzhu field. The control effect of Nicosulfuron was only 68.08%, and the control effect of Nico-sulfuron to abutilon theophrasti medic was worse than Mesotrione and Fluroxypyr. The total control effect of Mesotri-one mixed with Atrazine on weed was the best, it could be up to 99.41%. This can provide theoretical basis for thereasonable control of weeds in corn field.
In this paper, combined with practical maize production in Inner Mongolia Autonomous Region, dif-ferent cultivation patterns of maize were studied from 2011 to 2013. The indexes of plant growth development, yieldand economic benefits were measured and analyzed. The results showed that the soil environment, planting densityand fertilizer proportion of high yield and high efficiency pattern provided good conditions for maize growth bymulti-factors regulation. So the leaf area index and per area dry matter accumulation of maize of high yield and highefficiency cultivation were higher than that of farmers pattern and no fertilizer pattern in each growth stage. Produc-tion data showed that the average yield of 3 years of high yield and high efficiency pattern was higher than farmerspattern and no fertilizer pattern, the yield increased by 13.7% and 35.7%, respectively, and income increased by776.05 yuan/hm2, 1732.64 yuan/hm2, respectively.
The results showed that with the application of chemical fertilizers(N, P, K) alone, concentrations ofheavy metals(Ni, Cu, Zn, As. Pb) in soils changed insignificantly, but the concentration of Cr and Cd increased. How-ever, in plots amended with chemical fertilizers plus organic manure, concentrations of Cr, Cu, Zn, Cd and Pb in soilsincreased, which might raise potential risk of soil heavy metals contamination, especially the pollution of Cr, Cd, Cuand Zn. Application of fertilizers alone or combined with organic manure led to increment in concentrations of Ni, Asand Pb in maize plant, but the concentration of Zn reduced. The accumulation capacity of Cd in maize plant increasedin plots amended with chemical fertilizers alone, but that of Cr, Cu, Cd and Pb in maize plant decreased significantlywith the combined application of chemical fertilizers and organic manure. It was predicted that short term applicationof chemical fertilizers alone would not result in heavy metals pollution in maize seeds, but it was suggested to pay moreattention to the Cr, Zn and Cu pollution caused by application of chemical fertilizers plus organic manure.
The effectiveness of Trichogramma and Beauvenia bassiana on controlling of M. separate was tested.The results showed that the rate of egg mass parasitism of T. chilonis ranged from 55.56% to 77.78%, and the rate ofegg parasitism ranged from 14.24% to 27.69%. The rate of egg mass parasitism of T. dendrolimi ranged from 66.67% to 88.89%, and the rate of egg parasitism ranged from 14.63% to 30.54%. The revised effective of B. bassiana ranged from 20.75% to 54.72%.
Cloransulam-methyl WDG, a kind of sulfonamide, is a effective herbicide to control broadleaf weedsin soybean field. The objective of this study was to determine its control effect to Commelina communis and its safe-ty to different soybean varieties. The results of field trial showed that cloransulam-emthyl 84% WDG at 25.2~50.4g a.i/hm~2 was an effective herbicide to C. communis. and the control effects was from 95.1% to 100% after 45 days.It was safety to ‘Jiyu57' ,‘Jiyu64',‘Jiyu70' and ‘Jiyu74', which showed no any phytotoxicity and it had no anyadverse effects to soybean growth stage and yields.
The differential expression gene and gene expression pattern of the rice cultivar with different salineand alkaline treatments were studied by sequencing analysis of‘Teyou 12'. At the same time, the functional classi-fication and annotation of the differentially expressed genes were classified by biological process. The resultsshowed that the differential expression gene was mainly involved in the oxidation reduction, response to osmoticstress, response to stress, response to oxidative stress, secondary metabolic process, response to stimulus, responseto salt stress, response to chemical stimulus, and response to abiotic stimulus, etc.
选择生产中主推品种"吉薯1号",采取不同密度和不同施肥量配套栽培的种植方式,寻找"吉薯1号"最佳施肥量和种植密度.结果表明:"吉薯1号"最佳组合为施肥量1000 kg/hm2、密度25 cm.产量比对照提高18.6%,纯收入比对照多6418.8元/hm2.
To find a high γ-aminobutyric acid-producing lactic acid bacterium, more than 110 strains of lacto-bacillis isolated from Chinese fermented foods and astrointestinal tract of chickens were screened using thin-layerchromatography and colorimetric methods with glutamic sodium as substrate. The results showed that strain S6-4 ex-hibited high GABA-producing ability with 1.98g/L, which was identified as L. brevis by API 50 CHL kit and 16 S r D-NA sequencing analysis. The factors on transformation of GABA ability of strain S6-4 were analyzed and finally de-termined as follows: MRS medium with beef extract and yeast extract as nitrogen source, subsequently added 1mg/L(w/v) Vit B6 and Vit C as growth factor, initial pH was 6.5, cultured for 36 h under the condition of 27℃, the GABAyield reached 4.05g/L and increased by 105%.
Using the incomplete diallel cross method, correlation analysis between yield and main agronomictraits, path analysis of main agronomic characters and yield per plant was analyzed from 25 combinations of 5 malesterile lines of sorghum and 5 male sterile restorer lines and assembly. The genotypic correlation analysis showedthat the yield was significantly positive correlated with length of ear stem(0.5609), plant height(0.4923), thousandgrain weight(0.4034), and it was significantly positive correlated with yield per plant(0.3846), panicle weight(0.4389). The results of path analysis of main agronomic characters and yield per plant showed that traits influencedthe yield direct in such sequence: panicle weight length of ear stem growth stage stem 1000 grain weight spike length plant height.
Based on the observed data of monthly temperature, monthly rainfall of 34 weather stations and ten-day soil humidity of typical maize agro-meteorological stations during growth period in Jilin Province from 1981 to2010, using statistical method, the changing characters of temperature and rainfall and relationship between soil hu-midity of corn farmland and climate were analyzed. The results were as follows: since 1981, the temperature was in-creasing, the rainfall was decreasing, the soil humidity was descending during growth period, and temperature wasincreasing in each region, rainfall was decreasing in middle region, soil humidity descending in middle and middle-east region was a sudden phenomenon, which occurred in 1990 s. The relation between soil humidity and rainfall wasclosely in each region, the relation between soil humidity and temperature was closely in the middle and west region.Temperature increasing and rainfall decreasing were the main climatic factors caused soil humidity descending.
