
Objective:To explore the effect of corticotrophin-releasing hormone on pain threshold of adjuvant arthritis in rats and regulatory effect of Mongolian Warm Acupuncture.Methods: On the basis of adjuvant arthritis model,30 male Wistar rats were randomly divided into 3 groups(n = 10).① Saline group: inject 10μL saline,no warm acupuncture stimulation.② CRH group: CRH was diluted with distilled water,concentration was 0.lmg/mL,volume of injection volume was 500ng/5μL,amount of lateral ventricle was 10μL(5μL drug + 5μL NS flushing fluid).Injection time was 1min.No warm acupuncture stimulation.③ CRH+ warm Acupuncture group:Injected 1 hour after CRH and took Mongolian organs total points,with a special silver needle(diameter 0.5mm,length 3cm),then connected MLY-1 Mongolian warm acupuncture instrument.The current intensity of 100mA,40℃,to stimulate 15min.Every 15min measured pain threshold.Total four times.Results: Pain threshold AA rats injected NS was not significant different.CRH significantly increased the pain threshold,and injected CRH plus temperature stimulation can enhance pain threshold rats.
Objective:We aim to study the forensic pathological characteristics of sudden death caused by cerebral vascular malformation.Methods:We analyzed 39 cases of cerebral vascular malformation.The samples were collected from Inner Mongolia Medical College between 1998 and 2010.Results:34 cases were young men in our samples(91.89%).Thus young men were the major people died of the lesion.Fight and wound was the main remote cause of the lesion.24 cases(61.54%)belonged to this situation.Vascular rupture and bleeding was the most death cause of subjected people.Arteriovenous malformation was the most common pathological type of cerebral vascular malformation.The malformation type occupied92.30%(36cases).Cerebral vascular malformation often occurred in brain bottom,subarachnoid,lateral ventricles,cerebellum and brain stem.The subarachnoid was the most bleeding position which was16cases(41.00%).Conclusion:The identification of cerebral vascular malformation was very difficult.It was probably to make the objective,comprehensive and accurate identification conclusions combining forensic pathological characteristics with other examinations(MRI,cerebral angiography and so on).
Objective:To investigate the efficiency of abnormal fertilization in human IVF.Methods:The 3423 oocytes from 257IVF cycles were retrospectively analyzed by the age of women,the number of retrieved oocytes,the cleavage rate,the pregnancy rate,implantation rate between the two groups of the incidence of normal fertilization rate(2pronuclear,2PN)and abnormal fertilization rate(1PN,3PN,0PN).Results:There was no significant difference of the age of women,cleavage rate,pregnancy rate,implantation rate.The results suggested that there were significant difference of the number of oocytes,the number of can transfer embryos and good embryos,the rate of normal fertilization.Conclusion:The number of retrieved oocytes is the influencing factor of abnormal fertilization,the pregnancy rate is lower of fresh embryo transfer.But abnormal fertilization can get much transfer embryos and get better clinical result,at last.

Objective:study the expression of Autotaxin in gastric cancer and ATX's significance on tumor invasion and transformation.Methods:We investigated the expression of the ATX by immunohistochemically in 73 gastric cancers and in 50 normal gastric mucosa,as well as analyzed the correlation between the expression of ATX and clinicopathological features.Results:ATX were expressed in normal gastric mucosa tissue,gastric tumor tissue and adjacent tissue.ATX were expressed highly in tumor tissue and lowly in normal and adjacent tissue.ATX was expressed highly in normal and adjacent tumor tissue,but lowly in gastric tumor tissue.ATX expression has correlation with Histological type,Grade of Differentiation,intramural invasion and transformation,but has no correlation with patients' gender,age,location,and size of tumor.Tumor tissue with poor differentiation degree,deep invasion and a lymph node metastasis,TNM stage ⅢⅣ expressed in ATX were obviously higher than good differentiation degree,shallow invasion,without lymph node metastasis and TNM stage ⅠⅡ.Conclusion:High expression of ATX had important meaning in gastric tumor invasion transformation.

objective:To investigate the osteogenesis effect of Adeno-associated virus mediating basic fibroblast growth factor(AAV-bFGF) genetic transfection on human marrow stroma cells(hMSCs).Methods:hMSCs were isolated from health volunteers,cultured and purified in vitro,and randomly divided into 4 groups.①AAV-bFGF group,bFGF recombinant Adenovirus-associated virus was added into culture medium(1 ×1010OPU/mL) and incubated for 24h.②AAV-LacZ group,AAV-bFGF was replaced by LacZ recombinant Adenovirus-associated virus,others were same as AAV-bFGF group.③positive control group,dexamethasone(1nmol/L),glycerophosphate(10mmol/L) and ascorbic(50mg/L) were added into culture medium for 2wk.④control group,no special treatment was Administrated.Two weeks after treatment,bFGF expression were evaluated by immunohistochemistry,calcified nodes of hMSCs in each group were tested by Von Kossa staining and alkaline phosphatase(ALP) of culture medium in each group was detected.Results:hMSCs were uniform spindle-shape fibroblast after several passages.After treatment,hMSCs in AAV-bFGF group and in positive control group gradually showed flat shape,but did not In AAV-LacZ and blank control group.In immunohistochemistry,the level of bFGF expression in AAV-bFGF group were higher than the others,Von Kossa staining demonstrated that red bone collagen notes of hMSCs in AAV-bFGF group and positive control group were significantly more than in AAV-LacZ and blank control group.Similarly,ALP of culture medium in AAV-bFGF group and in positive control group were significantly higher than in AAV-LacZ group and blank control group.Conclusions:Adeno-associated virus mediated bFGF gene can transfect hMSCs and promote osteogenesis of hMSCs.
Objective:To detect the expression of MCM5 protein and PCNA in lung cancer tissues,and to evaluate the relationship between their expressions and clinical pathological parameters and their mutual correlation,and further provide theoretical basis for evaluating the occurrence,development,prognosis and treatment of lung cancer.Methods:Immunohistochemical technique was used to detect the expression of MCM5 protein and PCNA in 68 cases of lung cancer tissues and 20 cases of normal tissues,then to analyze the relationship between the expressions and clinical pathological parameters.Results:Positive signals of MCM5 expression lacates at nucleus,but not kytoplasm.Expression of MCM5 located limitedly in 1/3 to 1/2 cells at basilar part of epithelium in normal epithelial tissues,but that was expressed widespread in lung cancer tissues and a large number of such expressions were near to the surfaces of epithelium.①The expression of MCM5 was significantly different in lung cancer tissues and that in tumor adjacent tissues(P0.05).② The expression of MCM5 was closely correlated with differentiation degrees and lymphaden(P0.05),but were not closely correlated with ages and gender of patients(P0.05).③There was no significant correlation between the expression of PCNA and differentiation degrees(P0.05).④For normal tissues,PCNA labeling index was significantly higher than that of MCM5(P0.05).For lung cancer tissues,PCNA labeling index and MCM5 labeling index was not significantly different(P0.05).Conclusions:①The differences of tissue structure based on MCM5 staining and average optical density can comparatively distinguish normal lung tissues and cancer tissues.The expression of MCM5 was significantly correlated with differentiation degrees of lung cancer,hence expression of MCM5 can suggest malignant degree of lung cancer,contribute to judge prognosis and help doctors choose suitable treatment methods.Comparing with other proliferating labeling PCNA,MCM5 was a better cell proliferating marker and was more significantly correlated with differentiation degrees of tumors,and was a better marker and ranking index for lung cancer cells.②PCNA was overexpressed in lung cancer,which suggests that the overexpression of PCNA was closely correlated with occurrence and development of lung cancer.
Objective:To investigate the effect of bFGF in pathological mechanism of myopia by observing ophthalmic artery blood dynamics in cultured rabbit with eyelid suture-induced form deprived myopia.Methods:Form deprivation method was applied to establish the animal models of myopia by suturation of right eye-lid.Fourteen-day of neonatal rabbits were divided into group A,B and C:in group A(n=15),no drug was used,while group B(n=15)received a serious of subconjunctival injection of bFGF+PBS+insuline and group C(n=15)received PBS+insuline with monocular deprivation(MD).Respectively,the left eyes were employed as control groups.60 days later,data including refractive state,axial length(AL),ophthalmic artery resistance and ophthalmic artery blood stream velocity were obtained and analyzed.Results:Compared with normal control eyes,experimental eyes had obvious myopia after eyelid suture-inducing for 60 days in group A and group C.Yet these did not show any difference in group B.Each group in right and left eyes,Ophthalmic artery resistance and ophthalmic artery blood stream velocity flow were showed a significant difference.Speaking form deprived eyes,above targets did not show any difference between group A and group C,but comparison group B with group A and group C had notable significant differences.Conclusion:suturation eye-lid can induce axial myopia,higher ophthalmic artery resistance and slower ophthalmic artery blood stream velocity flow in rabbits.While bFGF can block these changes.
Objective To investigate the expression of Ezrin protein in gastric carcinoma and its clinicopathologic significance.Methods The expression of Ezrin protein was detected by Envision immunohistochemical method in tissues-microarry (TMA).There were 385 cases of gastric carcinoma [without lymphatic metastasis (n =114) and gastric carcinoma with lymphatic metastasis (n =271)].40 paraffin-embedded tissue blocks of normal gastric epithelium (>7 cm away from the edge of the tumor) were selected as the control group.All patients with gastric cancer were treated surgically and diagnosed histologically without preoperative chemotherapy or radiotherapy.Results The positive expression rates of Ezrin were 64.9% (250/385) in gastric carcinoma tissues and 5.0% (2/40) in normal tissues ; the expression of Ezrin protein in gastric cancer was significantly higher than that in normal gastric mucosa(P < 0.05) ; it was 16.6% (64/114) in gastric carcinoma tissues without lymph node metastasis and 48.3% (186/271) in those with lymph node metastasis.Ezrin protein expression was higher in metastatic gastric carcinoma than that in primary gastric carcinoma tissues [40.0% (154/385) vs 14.8% (57/385),P < 0.05].Ezrin protein expression in gastric cancer invasion to serosa was higher than that in the muscle layer only [44.4% (171/385) vs 9.4% (36/385),P < 0.05].Conclusions Ezrin may contribute to the invasion and metastasis of gastric carcinoma through enhancing or up-regulating Ezrin protein.
It 's observed that CPB plays a role in intestinal mucosal barrier function through three ways,including perfusion of intestinal mucosal is low,the contact between blood and the extracorporeal circuit with oxygenators activate the complement and coagulation system,followed by production of proinflammatory and damage of ischemia/reperfusion.The factors of CPB effects on intestinal mucosal barrier function,which include the perfusion of intestinal mucosal,systemic inflammation response to CPB,temperature,anesthesia,damage of ischemia/reperfusion,drugs,and so on.These effects of CPB are closely associated with systemic inflammation response or multiple organ dysfunctions.We summarize the relation between CPB and the mechanism.
Objective:To investigate the effect of RouKouWuWei and Captopril on matrix metolloproteinases(MMPs) and proto-oncogene c-myc expression in rodent model of myocardial infarction(MI).Methods:96 male SD rats were used in the study,six as normal control(C0),90 as MI model by intraperitoneal application of isoproterenol(ISO),which were further randomly divided into 5 groups with 18 rats each:control(Ct),without any treatment;Cap group,treated with Captopril(50mg/kg)orally;RKl,RKm and RKh groups,treated with RowKouWuWei pills in low,mid and high dosages(0.2,0.5 and 1.0g/kg)respectively.On day 1,7 and 28,6 were sacrificed each time and left ventricular weight index was calculated;MMPs measured by zymography;c-myc mRNA semi-quantified by RT-PCR.Results:Left ventricular weight index,bioactivity of MMP-2,MMP-9 and c-myc expression were increased significantly in rat MI models compared with C0;while the parameters decreased in Cap and RKh groups compared with Ct(P0.05 or P0.01).Conclusion:MMPs and c-myc expression increased in rat MI model,which may be one of the major causes of ventricular remodeling in MI;Captopril and RouKouWuWei Pill could suppress the expression of MMPs and c-myc and may be one of the mechanisms in MI treatment.
Refractory epilepsy mainly occurs in symptomatic epileptic patients.The sufferers are usually with learning difficulties,impaired mental abilities and other neurological symptoms due to recurrent and long-lasting seizures,and have poor therapeutic responses to antiepileptic drugs.Human neural stem cells as a source of new therapy becomes attempting as they can self-renew and also integrate into brain tissue and develop into different types of neurons.The main objective of this pilot study is to investigate the efficacy,safety and feasibility of the new therapy.Two refractory epileptic patients with trauma history were enrolled in the study.Human neural stem cells were derived from a seven-week therapeutic aborted foetus brain and expanded in tissue culture and confirmed expressing strong nestin marker.Two months after intravenous injection of the stem cells,both patients were completely free from seizures;their memory and behaviour were also improved remarkably.They were still seizure free and there was no obvious side effect observed over one year observation.The study indicates that observed neural stem cell therapy can be a new choice for refractory epilepsy as it is effective,safe and easy to administrate.

Objective:To study the effects of Sodium Aescinate on brain tissue of pancreatic encephalopathy(PE) in rats.Methods:Sixty Wister rats were divided into control group(n=30) and treatment group(n=30) randomly.Injected phospholipases A2(1000u/100g,1000u/0.1mL) by internal carotid artery,to copy animal model of PE.The control group and the treatment group were intraperitoneal injection of isotonic saline and sodium aescinate 1mg/100g differently,once a day.Model were killed(each group and n=10)respectively after day 1、day 3、day 7.Observe the pathological lesion of brain tissue by light microscope and detect the content of brain tissue water.After homogenate observed performance in the cerebral.Taking rat brain homogenate and peripheral blood,using enzyme-linked immunosorbent assay(ELISA) to determin the change of brain tissue tumor necrosis factor-α(TNF-α),interleukin-10(IL-10).Results:The rat water content of brain in the treatment group reduced significantly than that in the control group;Brain homogenate levels of TNF-α were decreased while IL-10 was increased(P0.05).The pathological lesion of demyelination in brain tissue decreased under light microscope.Conclusions:the effects of Sodium Aescinate on brain tissue of pancreatic encephalopathy(PE) in rats is anti-inflammatory.

Objective:To research the percutaneous process of rhein in the XIAOZHONGSAN paint,and select the optimal formula of paint.Methods:The formula of paint was made by the orthogonal design,and utilizing modified Franz diffusion cells,the content of rhein in the transdermal receptive liquid was determined by Dual-Wave thin layer chromatogram scanner with excised rat skin.The accumulation permeation amounts and permeation rate of rhein were calculated.Results:The optimal formula is as following:matrix 6%;Azone 6%;0.5g/mL drug concentration.The percutaneous penetration of rhein is increased by the transdermal time.Conclusion:The reference data of the XIAOZHONGSAN paint formula was provided by this optimization study.
