Aim: During the study of the aromatase inhibitors effect on metabolic disorders, it was found that exemestane significantly caused ascites.This part of pre-clinical study aimed to investigate the potential dysregulatory effect of exemestan on adrenal synthesis of hormones with miniralocorticoid activity under the conditions of experimental metabolic syndrome. Methods:This study was performed using female hamsters (1 and 2, 5 weeks old), that were fed hypocaloric diet for 6 (+3) weeks.Exemestane was administered orally in the dose of 3086 mg/kg for 3 weeks.Ascites caused by decreased liver function were excepted by measuring albumin content and histologic examination.Serum cortisol content was measured by ECLIA method.Serum aldosteron content was measured by ELISA method.Results: Under the conditions of experimental metabolic syndrome exemestane provoked significant increase of cortisol level by 1.56-1.62times and aldosterone level by 1.79-2.75times depending on the age of animals. Conclusion:Exemestane may cause mineralo-and glucocorticoids imbalance in female hamsters with metabolic syndrome.So the confirmation of exemestane safety in patients with adrenal hormones disorders and metabolic comorbidity needs additional clinical researches.
Aim Platelets provide substantial information about the proteolytic system profile in neurodegenerative diseases. Assessment of autophagy and proteasome target proteins in platelets may reflect tissue proteolytic machinery profile in central nervous system in Alzheimer’s diseases (AD). We aimed to demonstrate the optimum assay conditions and identify target proteins in platelet proteolytic machinery. Methods Platelet samples were obtained from clinically verified AD patients and age-matched non-demented control subjects that were recruited by University of Kansas Alzheimer’s disease Center. Autophagosome participating proteins in platelets were identified by Western blotting analysis. Standard gel electrophoresis and electro transfer apparatus were used for protein transfer onto the membrane. Several antibodies were tested to identify the best working antibodies, and their concentrations were optimized. An ELISA kit was used for platelet proteasome protein determination. Infrared imaging technology was used for visualizing the proteins on the membrane. Results Autophagosome participating proteins showed elevated levels in AD patient platelet cytosol. Only LC3-I autophagosome protein levels were significantly elevated. The concentrations of platelet lysate proteasome were assessed. AD patient’s proteasome levels were elevated but they were statistically not important as compared to controls. Conclusions Platelets can be used for assessing whether proteolytic system is functional. Blood-based sampling from human donors is less-invasive and analyzing platelet proteolytic system profile may help to develop pharmaceutical intervention approaches for neurodegenerative diseases in general.

Schistosomiasis is one of the most prevalent neglected tropical diseases, remains a serious public health problem in many developing countries in the tropics and subtropics and its pathogenesis depends on the parasite-host interaction. Periportal fibrosis of schistosomiasis complicated by portal hypertension results from increased intrahepatic vascular resistance and blood flow through the portal venous system. This leads to portosystemic collateral variceal veins formation which dilates until they finally rupture and bleed due to progressive rising in the portal venous pressure. The clinical impact of esophagogastric varices is critical due to the rebleeding rate of esophageal varices remains high within two years after cessation of acute bleeding from medium or large varices. In the present review, we will discuss hepatosplenic schistosomiasis and oesophagogastric varices pathophysiology and review the current surgical outcomes of distal splenorenal shunt or liver transplantation in schistosomal refractory variceal bleeding.
Aim: To investigate the cytotoxic effects of tamoxifen on the breast cancer cell line (MCF7). Methods:The cytotoxic effects of tamoxifen on MCF7 cells were investigated using caspase-9 activity and high content screening assays.Apoptosis mechanisms following tamoxifen treatment were also investigated. Results:The most significant cytotoxic effect of tamoxifen in MCF7 cells was a half-maximal inhibitory concentration (IC 50 ) of 4.506 µg/mL.A significant increase in caspase-9 activity was also observed when MCF7 cells were treated with tamoxifen (5 µg/mL).Furthermore, increased cell membrane permeability, cytochrome c level, and nuclear intensity were observed with tamoxifen (100 µg/mL) compared with doxorubicin (20 µg/mL) treatment.However, a noticeable decrease in cell viability and mitochondrial membrane permeability was observed with tamoxifen (100 µg/mL) treatment compared with doxorubicin (20 µg/mL) as a positive control. Conclusion:Tamoxifen showed in vitro cytotoxic effects in MCF7 cells as demonstrated by high-content screening and caspase-9 activity assays.Tamoxifen inhibits estrogen mechanisms, although toxic effect was observed.

New methodologies of genome editing as CRISPR/Cas9; ZFN or Talen make it a suitable model to perform studies to pair human genetic diseases as well. This review surveys recent studies; employing zebrafish as experimental model; comparing it with other in vivo and in vitro models; presenting zebrafish as a potent vertebrate tool to evaluate drug toxicity to facilitate more extensive; easy and comprehensive knowledge of new generation drugs.
