Установлено, что в пленках ДНК, сенсибилизированных органическими красителями (на примере акридинового оранжевого), при импульсном возбуждении в полосе поглощения красителя возможна реализация нескольких типов нестационарной голографической записи. Регистрировались динамические голографические решетки на триплетных состояниях красителя, тепловые и рельефно-фазовые голограммы-решетки недеструктивного типа. Эффективность и время существования этих решеток существенно зависят от концентрации красителя-сенсибилизатора, типа связывания его с биополимером и энергии записывающих импульсов. Высокоэффективные рельефные голографические решетки, сформированные вследствие структурных преобразований биополимерной матрицы при переходе в тепло энергии записывающих лазерных импульсов, самопроизвольно исчезают со временем полностью (или почти полностью) при комнатной температуре. Получены данные о распределении молекул акридинового оранжевого по сайтам связывания в матрице ДНК.
It is established that DNA films sensitized by organic dyes (for example, acridine orange), subjected to pulsed excitation in the dye absorption band, allow for several types of time-dependent holographic recording. Dynamic holographic gratings on the dye triplet states and thermal and relief-phase holographic gratings of nondestructive type have been recorded. The efficiency and lifetime of these gratings depend strongly on the concentration of dye, the type of its bonding with the biopolymer, and the recording-pulse energy. High-efficiency relief holographic gratings formed due to the structural transformations occurring in the biopolymer matrix when the energy of recording laser pulses dissipates into heat spontaneously and completely (or almost completely) disappear with time at room temperature. Some data on the distribution of acridine orange molecules over binding sites in the DNA matrix are obtained.
Using electronic absorption, luminescence, and vibrational spectroscopy, we study the properties of DNA-acridine orange biopolymer films. The conformational structure of DNA molecules in the film is examined at different levels of the relative humidity, and the state of dye molecules incorporated into the structure of the biopolymer is estimated. These structures were formed according to the “guest-host” principle and the dye molecule acts as the “guest” and the “host” is the intercalative DNA binding sites. In such system dye molecules demonstrate high luminescence capability and are the uniform optical centers and their fluorescence spectrum doesn’t depend on excitation wavelength.
