目的 观察电针联合穴位贴敷改善正常高值血压大鼠心肌纤维化的疗效及作用机制.方法 8只DR大鼠为空白组,32只DS大鼠随机分为模型组,针刺组,贴敷组,针刺+贴敷组.造模成功后针刺组每日电针"足三里""曲池"穴20min,贴敷组每日贴敷"脾俞"穴6h,针刺+贴敷组联合应用上述干预手段.治疗结束观察各组大鼠心肌胶原容积分数、TLR4、MyD88、NF-κB蛋白、mRNA含量及IL-6、TNF-α含量.结果 与空白组比较,模型组的心肌胶原容积分数、TLR4、MyD88、NF-κB mRNA、蛋白含量、IL-6,TNF-α含量升高(P<0.05).与模型组比较,治疗组上述指标均降低(P<0.05),针刺+贴敷组降低更明显(P<0.05).结论 电针联合穴位贴敷改善正常高值血压大鼠心肌纤维化与抑制TLR4/NF-κB信号通路活化有关.
Objective To develop an HPLC method for the simultaneous determination of chlorogenic acid and sinapine thiocyanate in Shiweiyipi granula(traditional Chinese medicines).Methods Chromatography was performed on a Kromasil100A C18 column(200 mm×4.6 mm,5 μm)with the mobile phase of acetonitrile-0.08 mol · L-1 ammonium dihydrogen phosphate solution,gradient elution.The flow rate was 1.0 mL · min-1.The column temperature was 30 ℃ and the detection wavelength was set at 326 nm.Results The linear calibration curve were obtained in the concentration range of 0.858-21.500 mg · L-1for chlorogenic acid and 2.140-53.500 mg · L-1for sinapine thiocyanate.The mean recoveries of chlorogenic acid and sinapine thiocyanate were 98.7%(RSD=1.7%,n=9)and 99.0%(RSD=1.9%,n=9),respectively.Conclusions The method can be used to control the quality of Shiweiyipi granula,which provides the advantage of quickness,simplicity and repeatability.
