目的研究放射性125I粒子植入不同时间对家兔肠管吻合口的影响。方法将60只家兔制成吻合口标本,随机分为对照组和实验组,每组30只。在吻合口两侧采用近距离治疗计划系统制定靶区,周边剂量:110Gy,植入3颗活度0.7mCi125I粒子,于术后第7、14、30、60、120天分别观察家兔一般情况,吻合口愈合情况,羟脯胺酸(HPC)含量,组织病理学改变和胶原纤维超微结构变化,评估碘125I粒子对吻合口的影响。结果对照组和试验组家兔一般情况良好,2组均未见腹腔感染及吻合口漏。血常规、肝肾功能、HPC含量,2组差异无统计学意义(P〉0.05),组织病理学改变结构变化不大。结论术中给予125I粒子周边剂量为110Gy时,家兔术肠管吻合口愈合无近期急性不良反应。
Objective To investigate effect of compound Kushen injection on the immune function of patients with gastrointestinal cancer treated by 125I interstitial brachytherapy.Methods 44 cases of gastrointestinal cancer were randomly divided into two groups.Patients in two groups were treated by 125I interstitial brachytherapy.And treatment group received compound Kushen injection.Lymphocyte subsets of CD4+ and CD8+ and IgG,IgA,and IgM levels in two groups were detected after a month.Results There was statistical significance between before and after treatment on CD4+,CD8+ and CD4+/CD8+ and IgG,IgA,and IgM levels in treatment group(P0.05).But there was no statistical significance between between before and after treatment in control group(P0.05).There was statistical significance between two groups on CD4+,CD8+ and CD4+/CD8+ and IgG,IgA,and IgM levels after treatment(P0.05).Conclusion Compound Kushen injection can enhance the immune function of patients with gastrointestinal cancer treated by 125I interstitial brachytherapy.
随着放射性粒子植入治疗肿瘤临床应用范围的扩大,对其引起的并发症也越显重要.穿刺恶性肿瘤能否引起针道医源性种植转移尚存争论[1,2].笔者对2005-2006年16例粒子植入患者术中所用植入针涂片行细胞学检查,现将术后随访结果报道如下.
To observe the possibility of neoplasm needle track implantation after radioactive seeds implantation and seek preventive measures to avoid it.
Objective To study the effect of spider venom from Macrothele raveni on proliferation and cell cycles of human hepatocellular carcinoma cell line SMMC-7721 and the molecular mechanism of the(effect.) Methods Proliferation of SMMC-7721 cells was determined by MTT assay.DNA synthesis of SMMC-7721 cell pre-and post-treatment with spider venom from M.raveni was tested by -TdR(assay.) The induction of apoptosis and the change of cell cycle in SMMC-7721 cells treated with spider(venom) from M.raveni were investigated by Flow Cytometry.The effect of spider venom from M.raveni on expression of c-myc protein in SMMC-7721 cells was studied by Western Blot.Results MTT assay showed that the proliferation of SMMC-7721 cells in vitro was inhibited by spider venom from M.raveni((P0.05)) in time-dependent and concentration-dependent manners.The DNA synthesis of SMMC-7721 cells was inhibited by spider venom from M.raveni.The apoptosis and inhibition of SMMC-7721 cells in the G_2/M phase were induced after treated with the spider venom from M.raveni.Expression of c-myc protein in SMMC-7721 cells treated with spider venom from M.raveni for 72 h was obviously down-(regulated.) Conclusion Spider venom from M.raveni could inhibit the proliferation and DNA synthesis of SMMC-7721 cells.The potential mechanisms are to induce cell apoptosis,activate the lower expression of c-myc protein and change the cell cycle of SMMC-7721 cells.
目的观察不同剂量 125 I粒子对人胃癌裸鼠移植瘤的杀伤作用及组织损伤。方法建立人胃癌BGC-823细胞裸鼠皮下移植瘤模型,随机分为对照组和实验组,将不同剂量(100、120、140 Gy) 125 I粒子植于实验组。30、60 d:比较移植瘤抑瘤率,病理组织学、局部皮肤反应、裸鼠体重及白细胞计数。结果30 d,100、120、140 Gy组抑瘤率分别为51.93%、79.18%、90.22%,病理组织学反应程度多为RCRG2(45.83%),各实验组组间除120 Gy与140 Gy(P>0.05)组外,及分别与对照组比较,差异有统计学意义(P<0.05);局部皮肤反应各实验组与对照组比较,差异有统计学意义(P<0.05),各实验组组间差异无统计学意义(P>0.05)。60 d,各组抑瘤率分别为97.97%、100%、96.69%,病理组织学反应程度以RCRG1居多(62.5%),与对照组比较,差异均有统计学意义(P<0.05),各实验组组间差异无统计学意义(P>0.05);局部皮肤反应随剂量增高而加重,各实验组组间及其分别与对照组比较,差异有统计学意义(P<0.05)。结论不同剂量的 125 I粒子在不同时间对人胃癌裸鼠皮下移植瘤有显著杀伤作用,但120Gy和140Gy组随剂量累积,损伤明显增加。提示100 Gy可能是治疗人胃癌裸鼠皮下移植瘤的有效剂量。
近年来,在动物毒素的抗肿瘤作用研究中,以蛇毒[1]、蜂毒[2]居多,未见蜘蛛蛛毒对肿瘤细胞作用的报道.本研究目的是从海南捕鸟蛛、虎纹捕鸟蛛、敬钊缨毛蛛、雷氏大疣蛛四种蜘蛛毒素中筛选出抗人肝癌细胞株BEL-7402作用强的一种,并探讨其量效及时效关系,以便进行更深入的研究.
目前,在国外125Ⅰ粒子组织间永久种植已广泛应用于各种恶性肿瘤的治疗,尤其是用于前列腺癌的治疗已相当成熟,但在腹部肿瘤的治疗方面仍在探索中.现就125Ⅰ粒子的应用现状、治疗的特点、优势、植入的剂量概念、原则及方式等方面作一综述.
