目的 观察小儿盐酸普鲁卡因口服溶液对消化系统疾病等所致呕吐患儿的疗效,为该药的后续研究提供参考.方法 收集2018年3月—2019年3月四川省人民医院儿科使用小儿盐酸普鲁卡因口服溶液患儿的临床资料,对其使用年龄、适应证、用法用量、不良反应、治疗效果等进行统计.结果 共收集使用小儿盐酸普鲁卡因口服溶液患儿550例,年龄25 d~13岁;适应证以胃肠炎引起的呕吐为主,也适用于胃炎、扁桃体炎、肠系膜淋巴结炎、上呼吸道感染、胃肠功能紊乱等引起的呕吐;用法用量为新生儿及1岁以下儿童口服用量<1 mL,1岁儿童用量平均为1 mL,每增加1岁用量增加1 mL,临床最大使用剂量为10 mL;不良反应为变态反应、食欲下降;治疗效果为治愈467例,好转78例,无效5例.结论 小儿盐酸普鲁卡因口服溶液对儿童消化系统疾病等引起的呕吐疗效好,不良反应少.
Objective To optimize the ethanol sedimentation process of Liqifuwei oral liquid in order to provide the reference for improving the quality and large-scale production of the oral liquid .Methods By using the anthraquinone transfer ratio of ethanol sedimentation solution and the yield of dry extract as indexes ,the effect of the relative density of concentrated liquor extracted by water , the ethanol concentration and the alcohol precipitation time on the ethanol precipitation process was investigated with orthogonal experi -ment method .Results When anthraquinone transfer rate as detecting index ,the various influence on the ethanol precipitation process were that ethanol concentration >alcohol precipitation time >the relative density .When the yield of dry extract as detecting index ,the various influence on the ethanol precipitation process were that relative density >ethanol concentration >alcohol precipitation time . Conclusion According to the test results,balancing various influences factors and combining with the actual production ,the optimal alcohol sedimentation process is that the relative density of concentrated liquor extracted by water is 1.20~1.25 ,ethanol concentration was 75%and alcohol precipitation time was 24 hours.The optimized process could improve the clarity of Liqifuwei oral liquid ,and pro-vide data supporting its large-scale production as well .
目的 通过探讨参附注射液中间体及不同配比对新生大鼠心肌细胞膜ATP酶活性及细胞内相关离子的影响,揭示参附注射液两种中间体最佳配比.方法 通过胰蛋白酶消化法和差速贴壁法获得心肌细胞,分别给予附子提取物30μl·ml-1、红参提取物30μl·ml-1及附子:红参1∶2、2∶1、1∶1不同配比药液,作用1h,检测心肌细胞的细胞活力、ATP酶的活性及相关离子的浓度.结果 参附注射液中间体及各配比药液对心肌细胞的细胞活力,Ca2-ATP酶、Ca2-Mg2+-ATP酶及Na+-K+-ATP酶的活性及细胞内Na+、Ca2+、K+、Mg2的浓度产生不同影响,其中以附子:红参2:1作用最强,且与前期研究中参附注射液的作用一致,均能提高心肌细胞的细胞活力,提高Ca2+-ATP酶、Ca2+-Mg2+-ATP酶及Na+-K+-ATP酶的活性,降低细胞内Na+、Ca2+的浓度,提高Mg2+、K+的浓度.结论 参附注射液中间体及不同配比均对正常心肌细胞具有保护作用,作用以附子:红参2∶1最强.
目的:探讨人参皂苷类成分对戊巴比妥钠损伤心肌细胞的影响.方法:将培养至第5日的乳鼠大鼠心肌细胞暴露于0.8%戊巴妥钠5min制备大鼠乳鼠心肌细胞损伤模型;选用人参皂苷Rg1为代表,MTT法确定作用浓度;然后选取最佳浓度作为人参皂苷Rgl、Rb1、Re及阳性药物西地兰的作用浓度,作用时间为24h,试剂盒测定心肌细胞内Na+、K+、Ca2+、Mg2浓度和细胞膜上Na+-K+-ATP酶、Ca2-ATP酶、Ca2+-Mg2+-ATP酶活性.结果:在1×108M剂量下,人参皂苷Rg1、Rb1、Re均能不同程度升高模型细胞Ca2-ATP酶、Ca2+-Mg2+-ATP酶的活性,降低Na+-K+-ATP酶的活性,同时纠正因造模引起的细胞内Na+、K+、Mg2+、Ca2+等离子浓度的异常.结论:人参皂苷Rg1、Rb1、Re对心衰心肌细胞具有一定治疗作用.
目的建立一种细胞水平的心律失常模型,以用于抗心律失常药物的筛选和评价。方法采用胰酶消化法体外培养SD乳鼠心肌细胞,通过差速贴壁的方法纯化后,在细胞水平给予传统诱发心律失常药物乌头碱,应用试剂盒测定给予乌头碱后心肌细胞内Na+、K+、Ca2+离子含量及Na+-K+-ATP酶、Ca2+-ATP酶活力的变化。结果乌头碱作用心肌细胞15 min后,使细胞内Na+、Ca2+离子含量增加,K+含量有减少,并呈一定的剂量依赖关系,其中1、2μmol/L乌头碱使细胞内Na+离子含量明显增加,与空白组比较有显著差异(P<0.01);能抑制细胞Na+-K+-ATP酶活力,其中0.5,1.0,2.0μmol/L乌头碱与空白组比较有差异(P<0.05),1.5μmol/L乌头碱与空白组比较有显著差异(P<0.01);有降低细胞Ca2+-ATP酶活力的趋势。结论在细胞水平给予乌头碱能引起细胞内离子含量及酶活性的改变,与心律失常发生机制基本一致。因此乌头碱心肌细胞模型是一种稳定可靠的抗心律失常药物筛选模型。
Objective:To study the effects of Shenfu injection on the Ca2+-Mg2+-ATP ase,Ca2+-ATP ase,Na+-K+-ATP and Na+,Mg2+,K+,Ca2+ in the myocardial cell injured by nembutal.Method: The myocardial cell is gotten by trypsin digestion and purified by different velocity of adherence.After injured by 0.8% nembutal,the myocardial cell is administrated with 3 different concentrations of Shenfu Injection.Then mensurate the vitality of the Ca2+-Mg2+-ATP ase,Na+-K+-ATP ase and Ca2+-ATP ase and Na+,Mg2+,K+,Ca2+ in the cell 1 h later.Result: The Shenfu injection can increase the strength of the myocardial cell beating injured by nembutal,and enhance the activity of Ca2+-ATP ase and the Ca2+-Mg2+-ATP ase,inhibit the the activity of Na+-K+-ATP,enzyme increase the content of Na+,Mg2+ and decrease the content of K+,Ca2+ in the myocardial cell.Conclusion: The Shenfu injection can protect the myocardial cell injured by nembutal,and the function is relevent to the effects of Shenfu injection on the Ca2+-Mg2+-ATP ase,Na+-K+-ATP ase,Ca2+-ATP ase and the content of Na+,Mg2+,K+,Ca2+ of myocardial cell.
Objective:To study the effects of Shenfu Injection on the ATP enzyme and related ion in the myocardial cell.Method: The myocardial cell is gotten by trypsin digestion and purified by different velocity of adherence.After administrated with 3 different concentrations of Shenfu Injection,the myocardial cell is mensurated the vitality of the ATP enzyme and related ion in the cell 1h later.Results: The Shenfu Injection(5,10,20μl/ml) can increase the vitality of the myocardial cell,and enhance the activity of Ca2 +-ATP enzyme,Ca2 +-Mg2 +-ATP enzyme and Na+-K+-ATP enzyme,decrease the content of Na+and Ca2 + in the myocardial cell,and increase the content of K+.Conclusion: The Shenfu Injection can protect the myocardial cell and the function is relevent to the effects of Shenfu Injection on ATP enzyme and related ion in the myocardial cell.
