Objective To improve some former culture methods of primary rat cortical neurons and get pure and long living cells for in vitro study.Methods Trypsin digestion and mechanical dissociation were adopted to conduct the culture.Morphological changes of neuron cells were observed under an inversion phase contrast microscope.Immunostaining of microtubule associated protein 2(MAP2)was applied to assess cell purity of the culture.Results Viability of the conducted cells was higher than before.Cells grew well under the given condition.Most cells had one or two ecptoms grown and their interlacing looked like a net on the fourth day after planting.After the neurons grew successively for 8 d,the cell body became bigger and the prominences were thicker and there were more branches than that at the initial stage.The prominences of neurons interlaced very well even to form a typical nerve fiber net.On the tenth day,purity of the cultured neurons was over 90% by immunohistochemical identification.Conclusions Method introduced in the paper is a simple technique that can be used for in vitro primary culture of rat cortical neurons.
Objective To sum up the application of cell injury models in compounds anti-cerebral ischemia activity screening in order to provide a new idea for the research of anti-cerebral ischemia in medicine.Methods The basic theories which related to acute cerebral ischemia and three cerebral ischemia in vitro cell injure models that used widely to imitate in vivo pathological mechanism progress were expounded.The way that the cell injure models used in new compounds' activity screening was discussed combined with these three cell injure models.Results Three cell injury in vitro models were used,which were now most widely accepted for they could represent three main pathophysiological mechanisms of ischemic cascade.OGD/R induced injury model was the initial injury process,excitotoxicity and oxidative stress had the most serious affects on the final ischemic damage.All models accorded with the pathophysiological injury theory for ischemic stroke and had certain representativeness,and the screeing results were thus comprehensive and might be reliable.Conclusions This method can be used to judge the impact mechanism of compound initially.

Salvianolic acid B is one of the currently studied salvia miltiorrhiza phenolic acids.Its absorption is rapid,but the bioavailability is low.Its distribution depends on the physical and chemical properties,protein binding rate and characters of tissue and organs.Metabolism of salvianolic acid B shows a non-linear pharmacokinetic property,and P450 enzyme involves in the process.The major excretion organ for the component is via bile.In the article,we summarized the studies on the pharmacokinetics of salvianolic acid B,and focused on the following aspects: processing procedures of plasma sample,determination validation,absorption,distribution,metabolism,excretion and pharmacokinetic interaction with the other drugs.
