目的 筛选分析调控氧化应激相关基因双特异性磷酸酶1(DUSP1)的微RNA(microRNA,miRNA,miR)分子,确定调控位点,探讨miRNA通过抑制DUSP1表达参与氧化应激调控的机制.方法 通过荧光素酶报告基因方法筛选出能调控DUSP1的miRNA,通过点突变及Western印迹等手段确定miRNA对DUSP1及下游靶分子的调控作用.结果 发现miR-101、miR-133、miR-144可调控DUSP1的表达,确定了调控位点,并发现它们可通过抑制DUSP1蛋白表达水平提高氧化应激条件下ERK1/2的磷酸化水平.结论 miR-101、miR-133和miR-144在氧化应激过程中能通过调控DUSP1发挥作用.
General disposition programming of ecological management is one ecological landscape area,four cultural landscape elements,six ecological water lake areas,six cultural landscape features attractions for the main Fenghe River.Cultural landscape planning includes the design of historical sites,road traffic,folk diet and leisure tourism.It also puts forward the concept that river regulation and construction of cultural landscape should be well combined in the process of river restoration,which can improve cultural connotation,satisfy culture demand of citizen's travel and protect local cultural sites.
Water diversion project brings great economic development opportunities for intake area,but also brings adverse impact for water source area.In order to formulate an effective compensation mode,realize the self-development of water sources,and ensure the harmonious development of ecology,economy and society,combined with the actual situation of Shangluo water source,this paper makes subject benefit analysis by Nash equilibrium game model,and puts forward 4 kinds of water source compensation modes,i.e.establishing compensation funding support system,developing circular economy and ecological tourism resources,ecological migration,and establishing corresponding supporting mechanisms.
Though Shangluo is an economically underdeveloped region in west China,it has its own economic development advantages.Rich natural resource,good tourist environment,regional geological location and opportunities of West China Development create favourable conditions for the economic development in Shangluo.Based on SWOT,their advantages and shortcomings,the opportunity and threats are discussed,which may bring forward a health economic development in Shangluo city.
通过对商洛市近年降水量和多年平均降水量来对比分析,得到商洛市水资源基本特点及其开发利用存在的主要问题:商洛水资源主要来源于降水,水资源丰富,但开发利用程度低,仅可开发利用9%;近年水资源总量逐年下降,但其用量却呈上升趋势,同时存在补给量不足的问题,提出了可持续开发利用的意见建议.
MicroRNAs (miRNAs) are a class of small regulatory RNAs that are thought to be involved in diverse biological processes by regulating gene expression. Numerous miRNAs have been identified in various species, and many more miRNAs remain to be detected. Generally, hundreds of mRNAs have been predicted to be potential targets of one miRNA, so it is a great challenge to identify the genuine miRNA targets. Here, we generated the cell lines depleted of Drosha protein and screened dozens of transcripts (including Cyclin D1) regulated potentially by miRNA-mediated RNA silencing pathway. On the basis of miRNA expressing library, we established a miRNA targets reverse screening method by using luciferase reporter assay. By this method, we found that the expression of Cyclin D1 (CCND1) was regulated by miR-16 family directly, and miR-16 induced G1 arrest in A549 cells partially by CCND1. Furthermore, several other cell cycle genes were revealed to be regulated by miR-16 family, including Cyclin D3 (CCND3), Cyclin E1 (CCNE1) and CDK6. Taken together, our data suggests that miR-16 family triggers an accumulation of cells in G0/G1 by silencing multiple cell cycle genes simultaneously, rather than the individual target.
目的:利用AdEasy系统构建miR-195的腺病毒表达载体,为研究miR-195的功能提供条件.方法:将miRNA-195前体序列从已构建的pcDNA-miR-195载体中克隆进穿梭载体pAdTrack-CMV,通过与骨架载体pAdEasy-1在大肠杆菌BJ5183中高效同源重组,获得完整的重组腺病毒质粒.利用HEK293细胞包装并扩增重组腺病毒Ad-195.Ad-195感染肺癌细胞A549后,利用miRNA的定量RT-PCR技术检测Ad-195在细胞中表达miR-195情况.随后荧光素酶报告基因实验证明,Ad-195表达的miR-195能够作用于BCL2基因上预测的miR-195靶位点.结果:实时定量PCR检测显示,Ad-195感染肺癌细胞A549后,miR-195表达水平有显著的上调.结论:miR-195的腺病毒表达载体构建成功,能够用于在细胞内过表达具有生物学功能的miR-195.
BitBLT engine performs accelerating graphics copy,bit block data transfer with ROP(Raster Operation) or transparency,monochrome to full color expansion for text,mono-bitmaps,and patterns and so on.The trade-off of speed,power and size should be considered carefully in design of BitBLT engine for small size LCD graphics controller.The functions and architecture of the recently developed BitBLT engine are described in the paper.The circuit implementation of the BitBLT engine is also presented.The techniques for processing acceleration and power optimization in the design are discussed.Finally,the results of the simulation and FPGA verification are presented.

在讨论手持设备中图形加速引擎BitBLT的功能、结构、电路实现的基础上,重点阐述了实现设计中总线宽度、多时钟设计、显示存储器仲裁逻辑、颜色扩展的实现等关键问题,通过对速度、功耗和面积等因素的优化处理和折衷考虑完成了图形加速引擎BitBLT设计,并给出了逻辑仿真及FPGA验证的结果.该设计采用流水线处理结构,能达到非常快的处理速度,数据处理速率可达到1 byte/时钟,同时进行了功耗优化.
