目的 在已建立高温致金黄地鼠神经管畸形(NTDs)模型的基础上,观察晚期糖基化终产物受体(RAGE)在高温致金黄地鼠NTDs神经上皮细胞中的表达变化,研究RAGE在高温致金黄地鼠NTDs发生过程中的作用,进一步探讨高温致NTDs作用的分子机制.方法 将孕鼠随机分为实验组和对照组,实验组孕鼠于妊娠第8天16~17时置42℃水浴持续20min,对照组于妊娠第8天16~17时置37℃水浴持续20min.分别于高温后8,16,24,40,64h(相当于胚龄第8,8.5,9,9.5,10.5天)处死孕鼠,剖腹取鼠胚,制备石蜡切片,应用免疫荧光染色技术方法检测RAGE在NTDs发生中神经上皮细胞的表达变化规律.结果 水浴处理后8,16,24,40h,实验组与对照组RAGE免疫荧光染色弥散分布于鼠胚神经上皮细胞和周围间充质细胞的胞膜上;水浴处理后64h,实验组与对照组均未检测到RAGE阳性表达.8,16,24,40h,实验组与对照组相比,胚胎神经上皮细胞的RAGE阳性表达减弱,差异有统计学意义(P<0.05).结论 RAGE在高温致NTDs过程中表达降低,是高温致NTDs发生的重要途径之一.
Objective To study the expression of E-cadherin ( E-cad) and vascular endothelial growth factor ( VEGF) in gestational trophoblastic disease and provide theoretical basis for its early diagnosis and treatment.Methods Ten cases of hydatidiform mole,20 cases of invasive hydatidiform mole and 20 cases of choriocarcinomas were selected. Ten cases of normal villi tissues served as the control group.Expression of E-cad and VEGF in normal chorion of early gestation,hydatidiform mole,invasive hydatidiform mole and choriocarcinoma were detetected with immunohistochemistry method and image analysis technique.Results ①The expression of E-cad in normal villi,hydatidiform mole,invasive hydatidiform mole and choriocarcinomas was 3.521,3.1045 0.9655 0.7682,gradually declined with the progression of gestational trophoblastic disease (P 0.05).②The expression of VEGF in normal villi,hydatidiform mole,invasive hydatidiform mole and choriocarcinomas was 0.4219,0.7521,0.9611,1.1420,significantly elevated gradually with the progression of gestational trophoblastic disease ( P 0.05).Conclusions ①The lower expression of E-cad is related to the progression of gestational trophoblastic disease and helpful to distinguish choriocarcinomas from hydatidiform mole.The overexpression of VEGF is associated with the deterioration of GTT.②The expression changes of E-cad and VEGF may play an important role in the development of gestational trophoblastic tumor.
In order to explore the molecular mechanism of neural tube defects(NTDs) induced by hyperthermia,which may provide a theoretical basis for the prevention of human NTDs.On the base of establishing the hyperthermia-induced golden-hamster NTDs model,the expressional changes of HMGB1 in the neuroepithelial cells during the occurrence of NTDs were investigated.The pregnant rats were randomly divided into the experimental and control groups.The pregnant rats were sacrificed at 8、16、24、40、64 hours(corresponded the embryonic 8,8.5,9,9.5 and 10.5 days of age) after water bath treatment,then the fetuses were got.After conventional paraffin-embedded,the expression of HMGB1 in the neuroepithelial cells during the occurrence of NTDs were detected using immunofluorescent staining technique.The results showed that in the control and experimental groups,HMGB1 immunoreactivity was all diffusely distributed in the cytoplasm of neuroepithelial cells and mesenchymal cells surrounded the neural tube at 8,16 and 24 hours after water bath treatment.At 40 and 64 hours,the expressional place of HMGB1 emerged from the cytoplasm to the nucleus.After maternal hyperthermia,the staining intensity of HMGB1 immunoreactivity in the neuroepithelia at each experimental group was weaker than that of control group.The above results suggest that the expressional change of HMGB1 is closely linked to the development of the neural tube,and the decreased expression of HMGB1 may be involved in one of the important ways on hyperthermia-induced NTDs.
目的 探讨大鼠胚胎脊髓神经干细胞的体外培养生长及分化情况,为神经干细胞的临床应用提供实验依据.方法 取孕13d SD大鼠胚胎脊髓,在条件培养基中培养获得神经干细胞,应用免疫荧光法鉴定.神经干细胞在含血清的培养基中传代培养,应用免疫细胞化学法检测神经干细胞的分化情况.结果 从大鼠胚胎脊髓可获得具有自我增殖分化能力的神经干细胞,培养7d即可获得神经球.用免疫细胞化学和免疫荧光法鉴定干细胞分化,可检测到神经元和星形胶质细胞.结论 从大鼠胚胎脊髓能分离获得神经干细胞,并可向神经元和星形胶质细胞方向分化.
