OBJECTIVE:To investigate the expression of EZH2 in breast cancer and triple-negative breast cancer(TNBC) and their correlation.METHODS:A total of 20 cases of breast benign lesion and 82 cases of breast cancer were detected for the expression of EZH2 by the immunohistochemistry method,and their relationships were analyzed with the clinical pathologic characteristics and the expression of TNBC.The 5-year disease-free survival(DFS) was analyzed by Kaplan-Meier methods.RESULTS:The EZH2 expression was detected in 20.0% of breast benign lesion cases and in 51.2% of breast cancer cases,and the difference showed a statistically significance(χ2=6.329,P=0.012).In the 82 cases of breast cancer patients,the expression of EZH2 was correlated with the tumor size,pathological grade and lymph node metastasis(P0.05).In contrast,it did not show correlation with patients age,whether or not menopause and family history of breast cancer(P0.05).The EZH2 expression was detected in 89.5% of TNBC and 39.7% of non-TNBC,and the difference showed statistical significance(χ2=14.484,P=0.000).The 5-year DFS was significantly lower in EZH2 positive patients than in EZH2 negative patients(64.3% vs 85.0%),Log-rank=3.995,P=0.046).CONCLUSIONS:The expression of EZH2 is higher in breast cancer than in breast benign lesion.In breast cancer patients,the patients with over expression of EZH2 have an increased possibility of recurrence or metastasis,and more intimate relationship with TNBC.
Objective To explore expressions of HIF-1α,Glut1 and PCNA in gastric cancer, and to analyze the clinical significance.Methods Immunohistochemical staining method was used to detect expressions of HIF-1α, Glut1 and PCNA in 52 specimens of gastric cancer and 20 specimens of normal gastric tissues. Furthermore,their interrelations with biological behaviour of gastric cancer were analyzed.Results None of the normal gastric tissues expressed HIF-1α and Glut1.Whereas positive expression rates of HIF-1α and Glut1 in gastric cancer were 69.2℅ and 75%,respectively. HIF-1α expression correlated with TNM stage(P0.01),lymph node metastasis(P0.01) and Glut1 expression(r1=0.580,P0.01).Glut1 expression correlated with cell differentiation(P0.01),TNM stage(P0.01) and lymph node metastasis(P0.01).PCNA level was higher in gastric cancer(65.19%±20.82%) than that in normal gastric tissues(15.50%±9.72%)(P0.01).The positive expressions of HIF-1α and Glut1 were positively correlated with PCNA level(r2=0.723,P0.01;r3=0.527,P0.01).Conclusion The overexpressions of HIF-1α,Glut1 and PCNA protein are closely associated with carcinogenesis and progression of gastric cancer. And it is of clinical significance to guide the diagnosis and prognosis of gastric cancer.
目的探讨乳腺癌中LRP、GST-π、TOPOⅡ的表达及临床意义,以期为选择化疗方案提供参考,获得更有效的化疗效果.方法应用免疫组化SABC法,对80例乳腺癌的主要耐药指标LRP、GST-π、TOPOⅡ进行检测,并结合临床分期进行研究.结果乳腺癌中LRP表达率为80.00%(64/80),临床Ⅱ、Ⅲ期表达率显著高于Ⅰ期,GST-π表达率为65.00%(52/80),临床Ⅲ期表达率显著高于Ⅰ、Ⅱ期.TOPOⅡ表达率为55.00%(44/80),临床Ⅲ期表达率显著低于Ⅰ、Ⅱ期.LRP与GST-π间有明显相关性,说明它们对肿瘤耐药起作用具有一定的协同性.LRP、GST-π与TOPOⅡ间无明显相关性,说明它们各自的表达从不同方面对肿瘤耐药起作用.结论乳腺癌联合检测LRP、GST-π、TOPOⅡ对化疗药物的选择具有指导作用,对预后的判断有参考价值.
肿瘤细胞多药耐药的形成是化疗成功的一大障碍.肿瘤耐药性主要包括4个方面:①多药耐药(MDR1)基因过度表达介导的,称为典型多药耐药;②由拓扑异构酶Ⅱ(DNA topoisomerase Ⅱ,TOPOⅡ)介导的,称为不典型多药耐药;③谷胱甘肽S转移酶的表达;④肺耐药蛋白(lung resistance protein,LRP)的表达.我们采用免疫组织化学法对80例乳腺癌LRP、TOPOⅡ进行检测,现报告如下.
