О-антигены (О-специфические полисахариды) шигелл Флекснера (Shigella flexneri) - одного их основных возбудителей шигеллеза - отличаются широким разнообразием химических модификаций, происходящих после сборки олигосахаридного О-звена. Настоящий обзор посвящен структурным, серологическим и генетическим аспектам этих модификаций, включая недавно обнаруженные О-ацетилирование и фосфорилирование фосфоэтаноламином. Эти модификации наделяют хозяина специфическими иммунодетерминантами (О-факторами или О-антигенными эпитопами), которые обусловливают антигенное разнообразие шигелл Флекснера, рассматриваемое как один из факторов вирулентности этого патогена. Всего идентифицировано 30 вариантов О-антигенов шигелл Флекснера, и соответствующие им О-факторы охарактеризованы с помощью специфических антител. На основании этих данных предложено существенно расширить схему серотипирования этих бактерий. Идентифицирован ряд генов, ответственных за модификации О-антигенов и вызываемые ими конверсии серотипов S. flexneri. Выявлены генетические механизмы диверсификации О-антигенов, включающие приобретение мобильных генетических элементов, таких как профаги и плазмиды, возможно, с последующей мобилизацией и инактивацией генов. Полученные результаты углубляют наши представления о генетике и антигенности шигелл Флекснера и способствуют контролю над распространением шигеллеза.
О-полисахарид (О-антиген) Salmonella enterica O51 выделен мягкой кислотной деградацией липополисаха-рида, и его структура установлена с помощью углеводного анализа и ЯМР-спектроскопии: ?-D-GlcpNAc 1 v 3 >6)-?-D-Glcp-(1>4)-?-D-Galp-(1>3)-?-D-GalpNAc-(1>3)-?-D-GlcpNAc-(1>. Секвенирование генных кластеров биосинтеза О-антигенов S. enterica O51 и E. coli O23 показало, что они содержат одинаковый набор генов с высоким уровнем гомологии. Сравнение с генетическими базами данных позволило определить функции всех ожидаемых генов биосинтеза общей структуры обоих О-антигенов. Сделан вывод о происхождении генных кластеров обеих бактерий от общего предшественника, тогда как модификация О-антигена E. coli O23, по-видимому обусловленная геном профага вне генного кластера, могла произойти уже после обособления бактерий в отдельные виды.
О-Полисахарид выделен после мягкой кислотной деградации липополисахарида Escherichia coli O108 и исследован методами моносахаридного анализа, а также с помощью одно- и двумерной спектроскопии 1H- и 13С-ЯМР. Обнаружено, что в состав полисахарида входит необычный высший сахар - 5,7-диацетамидо3,5,7,9-тетрадезокси-L-глицеро-D-галакто-нон-2-улозоновая кислота (ди-N-ацетил-8-эпилегионаминовая кислота, 8eLeg5Ac7Ac). Установлена следующая структура тетрасахаридного повторяющегося звена: →4)-б-8eLgp5Ac7Ac-(2→6)-б-D-Galp-(1→3)-б-L-FucpNAc-(1→3)-б-D-GlcpNAc-(1→. The structure of the O-polysaccharide (O-antigen) from Shigella dysenteriae type 8 bacteria (strain 599) was corrected using modern NMR techniques (structure 1). The revisions concerned the position of the Glc residue (in the main, but not the side, chain), the site of its substitution, and the configuration of the O-glycoside linkage of the GlcNAc residue. The S. dysenteriae type 8 bacterium (strain G1221), the second investigated representative, was found to produce another structural variant of the O-polysaccharide. It contains GlcNAc instead of the Glc residue in the main chain (structure 2). This data may lead to approval of division of S. dysenteriae type 8 into two subtypes: [Formula: see text].

The main shortcoming of the modem live and killed vaccines based on gram-negative bacterial strains is their ability to cause adverse reactions. The majority of the adverse reactions are associated with the effect of biological activity of lipopolysaccharide. The report covers the problems concerned with biogenesis of the lipid A, lipopolysaccharide structural component, responsible for its endotoxic activity, as well as with genes determining lipid A synthesis. The special attention is paid to gene-engineering technique for reduction of adverse reactions of vaccine strains that are based on the knock-out mutagenesis of genes waaM and/or waaN responsible for addition of lauroyl and myristoyl residues to the distal glucosamine unit lipid A, generating acyloxyacyl moieties.
An acidic O-specific polysaccharide was obtained by mild acid degradation of the lipopolysaccharide of Proteus mirabilis O10 and studied after full acid hydrolysis and carboxyl reduction by 1H- and 13C-NMR spectroscopy, including two-dimensional correlation spectroscopy (COSY), H-detected heteronuclear 1H,13C multi-quantum coherence (HMQC), and rotating-frame nuclear Overhauser effect spectroscopy (ROESY). It was found that the polysaccharide contains 2-acetamido-2-deoxy-D-glucose, 2-acetamido-2-deoxy-D-galactose, D-galacturonic acid, and L-altruronic acid, and the following structure of the branched tetrasaccharide repeating unit of the polysaccharide was established: [sequence: see text]
Based on acid hydrolysis, methylation, and one- and two-dimensional 1H- and 13C-NMR spectroscopy, including homonuclear and 1H, 13C heteronuclear correlation spectroscopy (COSY) and rotating-frame nuclear Overhauser effect spectroscopy (ROESY); it was found that the O-specific polysaccharide chain of the lipopolysaccharide of the bacterium Proteus mirabilis O30 is a hexosamino-glucuronan built up of tetrasaccharide repeating units having the following structure: [formula: see text] The degree of O-acetylation of 2-acetamido-2-deoxyglucose is about 70%.
The review is devoted to a surface carbohydrate antigen of the bacterium Pseudomonas aeruginosa which is common for the majority of the species and has a lipopolysaccharide nature. The occurrence, detection, isolation, structure, expression on the cell surface, interaction with antibodies, an antibiotic and a bacteriophage as well as the immunotherapeutic potential of the common polysaccharide antigen are discussed.
Data on the composition and structure of O-antigens which represent polysaccharide chains of outer membrane lipopolysaccharides of gram-negative bacteria defining cell immunospecificity are reviewed with special reference to some structural features of O-antigens, such as masked regularity and occurrence in one microorganism of lipopolysaccharides with two structurally distinct polysaccharide chains. Antigenic relationships between the microorganisms belonging to different taxonomic groups are discussed.
Like some Pseudomonas cepacia serogroups studied earlier, serogroups C, I (Nakamura), O1 and O4 (Heidt) are characterized by the presence of at least two structurally different O-antigenic polysaccharide chains in cell-wall lipopolysaccharides. On the basis of acid hydrolysis, methylation, 1H- and 13C-NMR spectroscopy, including computer-assisted 13C-NMR-based analysis, the complete structures of the predominant polysaccharides of serogroups I (I), C and O4 (III) and the minor polysaccharides of serogroups I (II) and O1 (V) were established, and the structure of the predominant polysaccharide of serogroup O1 (IV) established earlier (Cox A. D., Wilkinson S. G.@Carbohydr. Res. 1990. V. 195. No 2. P. 295-301) was confirmed.
The data on the structure and properties of outer membrane lipopolysaccharides of gram-negative bacteria are reviewed with special reference to the general organization of the lipopolysaccharide molecule and the structure of the lipid moiety (lipid A). The correlation between the structure of the lipopolysaccharides and lipid A and their biological properties and functions are discussed.
