The precise identification of associations between diseases and drugs is paramount for comprehending the etiology and mechanisms underlying parasitic diseases. Computational approaches are highly effective in discovering and predicting disease-drug associations. However, the majority of these approaches primarily rely on link-based methodologies within distinct biomedical bipartite networks. In this study, we reorganized a fundamental dataset of parasitic disease-drug associations using the latest databases, and proposed a prediction model called PDDGCN, based on a multi-view graph convolutional network. To begin with, we fused similarity networks with binary networks to establish multi-view heterogeneous networks. We utilized neighborhood information aggregation layers to refine node embeddings within each view of the multi-view heterogeneous networks, leveraging inter- and intra-domain message passing to aggregate information from neighboring nodes. Subsequently, we integrated multiple embeddings from each view and fed them into the ultimate discriminator. The experimental results demonstrate that PDDGCN outperforms five state-of-the-art methods and four compared machine learning algorithms. Additionally, case studies have substantiated the effectiveness of PDDGCN in identifying associations between parasitic diseases and drugs. In summary, the PDDGCN model has the potential to facilitate the discovery of potential treatments for parasitic diseases and advance our comprehension of the etiology in this field. Anterior segment dysgenesis is a severe developmental eye disorder that leads to blindness in children. The exact mechanisms underlying this condition remain elusive. Recently, an increasing amount of studies have focused on genes and signal transduction pathways that affect anterior segment dysgenesis;these factors include transcription factors, developmental regulators, extracellular matrix genes, membrane-related proteins, cytoskeleton proteins and other associated genes. To date, dozens of gene variants have been found to cause anterior segment dysgenesis. However, there is still a lack of effective treatments. With a broader and deeper understanding of the molecular mechanisms underlying anterior segment development in the future, gene editing technology and stem cell technology may be new treatments for anterior segment dysgenesis. Further studies on the mechanisms of how different genes influence the onset and progression of anterior segment dysgenesis are still needed.
OBJECTIVE To observe the effects of moxibustion on the ultrastructure of synovial cells of knee joint and serum cytokines in adjuvant arthritis (AA) rats, and to explore the potential mechanism of moxibustion in treatment of rheumatoid arthritis. METHODS Forty-five Wistar male rats were randomly divided into a normal group, a model group and a moxibustion group, with 15 rats in each group. In the model group and the moxibustion group, the AA model was replicated under wind, cold and humid environment and by injection with complete freund's adjuvant. In the moxibustion group, moxibustion at "Zusanli" (ST 36) and "Shenshu" (BL 23) was used, 20 min each time, once daily, for consecutive 21 days. In the normal group and the model group, no intervention was processed. The scores of the knee joint swelling degree (JSD) and arthritis index (AI) were compared among groups. The ultrastructure of synovial cells of knee joint were observed under transmission electron microscope (TEM). The levels of serum cytokines such as tumor necrosis factor-α (TNF-α), interieukin (IL)-1β, IL-6 and IL-10 were detected using ELISA method. RESULTS Compared with the normal group, JSD and AI scores, the levels of TNF-α, IL-1β and IL-6 were increased (P<0.01), while IL-10 was reduced (P<0.01) in the model group after intervention. JSD and AI scores, and the levels of TNF-α, IL-1β and IL-6 were lower (P<0.05, P<0.01), while the level of IL-10 was higher (P<0.01) in the moxibustion group compared with the model group. Compared with the normal group, the ultrastructure of synovial cell was obviously damaged in the model group, and the damage was attenuated in the moxibustion group compared with the model group. CONCLUSION Moxibustion can reduce the symptoms of arthritis in AA rats, which may be related to the improvement of the ultrastructure of synovial cells and the regulation of cytokines.

In order to promote the extensive development of Time Bank, this paper makes a theoretical analysis on the conversion of time currency value, withdrawal and deposit, the conversion of labor value and time currency value in Time Bank, and the inheritance of time currency savings, so as to create an all-round, wide-area, multi-level and integrated old-age service ecology, create a service-oriented old-age model with China characteristics, and contribute to China's wisdom and China's plan for solving the world's old-age problems.
OBJECTIVE To observe the effect of moxibustion at Zusanli (ST 36) and Shenshu (BL 23) on inflammatory factors and intestinal flora in the rats with adjuvant arthritis. METHODS A total of 36 Wistar rats were randomized into a normal group, a model group and a moxibustion group, 12 rats in each one. In the model group and the moxibustion group, the adjuvant arthritis model was established by a compound method, including the environmental factors, i.e. wind, cold and damp, and Freund's complete adjuvant. In the moxibustion group, moxibustion intervention was exerted at Zusanli (ST 36) and Shenshu (BL 23), for 20 min at each acupoint, once daily, consecutively for 21 days. The paw swelling degree and arthritis index (AI) score were observed before and after intervention in the rats of each group. Using real-time fluorescence quantitative PCR method (real-time PCR) and Western blot method, the mRNA and protein expressions of inflammatory factors of colon tissue, i.e. interleukin (IL) 1β, tumor necrosis factor-α (TNF-α), IL-6, were detected after intervention in the rats of each group. The intestinal flora was detected with 16SrRNA sequencing technology after intervention in the rats of each group. RESULTS Compared with the normal group, the paw swelling degree and AI score were increased in the rats of the model group (P<0.05); after intervention, compared with the model group, the paw swelling degree and AI score were reduced in the rats of the moxibustion group (P<0.05). Compared with the normal group, the expressions of IL-1β, TNF-α and IL-6 mRNA, as well as proteins were increased in the colon tissue of the rats in the model group (P<0.05); compared with the model group, the expressions of IL-1β, TNF-α and IL-6 mRNA, as well as proteins were reduced in the colon tissue of the rats in the moxibustion group (P<0.05). Compared with the normal group, OTUs count was reduced in the rats of the model group (P<0.05); and compared with the model group, OTUs count was increased in the rats of the moxibustion group (P<0.05). Compared with the normal group, Simpson index was increased, Chao1 and Ace were reduced in the rats of the model group (P<0.05); while, compared with the model group, Chao1 and Ace were increased in the rats of the moxibustion group (P<0.05). Compared with the normal group, the relative abundance of uncategorized Clostridium, Lactobacillus, Prevotella, uncategorized Porphyromonadaceae and uncategorized Prevotella was increased (P<0.05), and the relative abundance of uncategorized Spironella was reduced in the model group (P<0.05). While, compared with the model group, the relative abundance of uncategorized Clostridium, Lactobacillus, uncategorized Prevotella was reduced in the moxibustion group (P<0.05) and that of uncategorized Spironella was increased (P<0.05). CONCLUSION Moxibustion at Zusanli (ST 36) and Shenshu (BL 23) relieves the joint symptoms of adjuvant arthritis rats and inhibits the expressions of inflammatory factors, which is probably related to the regulation of the structure of intestinal flora.
OBJECTIVE To observe the clinical effect of moxibustion combined with western medication for rheumatoid arthritis (RA) of liver-kidney deficiency, and explore the mechanism of moxibustion for RA. METHODS A total of 60 patients with RA of liver-kidney deficiency were randomly divided into an observation group and a control group, 30 cases in each group. In the control group,leflunomide tablets were taken orally, once a day. On the base of the treatment as the control group, moxibustion was applied at Sanyinjiao (SP 6), Shenshu (BL 23), Zusanli (ST 36) and ashi points in the observation group, once every other day, three times a week. Totally 8 weeks were required in both groups. Before and after treatment, the TCM syndrome score, health assessment questionnaire (HAQ) score, 28-joint disease activity score (DAS-28) and serum levels of calcium ion (Ca2+), erythrocyte sedimentation rate (ESR), high-sensitivity C-reactive protein (hs-CRP) and rheumatoid factors (RF) were compared, and the clinicial efficacy was evaluated in the two groups. RESULTS After treatment, the TCM syndrome scores, HAQ scores and DAS-28 scores were decreased in the two groups (P<0.01), and those in the observation group were lower than the control group (P<0.01). After treatment, the serum levels of Ca2+ were increased (P<0.01), ESR and hs-CRP were decreased in the two groups (P<0.01), and the serum level of RF was decreased in the observation group (P<0.01). The serum level of Ca2+ in the observation group was higher than the control group (P<0.01), ESR and RF were lower than the control group (P<0.01). The total effective rate was 83.3% (25/30) in the observation group,higher than 60.0% (18/30) in the control group (P<0.05). CONCLUSION Moxibustion combined with western medication can effectively relieve clinical symptoms, improve quality of life in RA patients, the curative effect is better than simple western medication. And its mechanism may be related to the regulation of serum level of Ca2+ and immune inflammatory response to inhibit bone destruction.
At present, the contradiction between the high efficiency of the world’s food production and environmental pollution is becoming increasingly prominent. In order to study the related issues of the world’s food production efficiency and sustainability, this paper uses the method of entropy weight to extract 5 indicators as the environmental pollution assessment model from the environmental pollution of agricultural production, scores 10 major agricultural production countries in the world, and obtains the environmental pollution index. Subsequently, the DEA model was established for these countries, the environmental pollution index was included into the production efficiency system as an unexpected output, the Malmquist index was established to describe the changes in agricultural production efficiency from 2010 to 2018, and the cluster analysis was carried out for it. Subsequently, the OSL and Tobit models were used for regression of the influencing factors. In addition, the comprehensive evaluation model of efficiency and sustainability was established by controlling the amount of fertilizer used, which was applied in different countries.
Although persistent hepatitis B virus (HBV) infection is associated with natural killer (NK) cell dysfunction, it remains obscure whether HBV viral antigens are responsible for NK cell dysfunction in patients with chronic hepatitis B (CHB) infection. In this study, we found that the percentage of NK cells expressing the inhibitory receptor, NKG2A, was increased in CHB patients, and NKG2A blockade restored NK cell function. Furthermore, in CHB patients, the frequency of NK cells expressing NKG2A positively correlated with the number of regulatory T cells (Tregs) and production of interleukin-10 (IL-10) in these Tregs. Moreover, exposure of peripheral blood mononuclear cells (PBMCs) isolated from healthy controls to sera from CHB patients resulted in increased proportion of NKG2A+ NK cells; IL-10 blockade reduced the frequency of NKG2A+ NK cells while increasing the percentage of IFN-γ+ NK cells. In addition, stimulation of NK cells and Tregs from healthy controls with CHB sera together with anti-IL-10 antibody increased IFN-γ production in the culture supernatant. The frequencies of NKG2A+ NK cells and IL-10+ Tregs, along with serum levels of alanine transferase and HBV DNA, were significantly increased in CHB patients positive for the Hepatitis B e antigen (HBeAg, a marker of viral replication) when compared to HBeAg-negative CHB patients. Importantly, exposure of PBMCs from healthy controls to HBeAg resulted in increased IL-10 production but reduced levels of TNF and IFN-γ, and IL-10 blockade rescued the generation of TNF and IFN-γ in this assay. The reduced production of TNF and IFN-γ was also observed in NK cells and Tregs from healthy controls that were stimulated with HBeAg, while IL-10 blockade increased the secretion of these two cytokines. We conclude that HBeAg induces IL-10 production in Tregs, thereby leading to increased expression of NKG2A on NK cells, which contributes to NK cell dysfunction during CHB infection. These data suggest that HBeAg is associated with NK cell dysfunction in CHB.
The ongoing spread of coronavirus disease 2019(COVID-19)constitutes an international concern on an unprecedented scale.To date,over 23 million people have been diagnosed with CO VID-19 worldwide,and this disease has caused more than 800,000 deaths.Hyperinflammation elicited by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has been reported to contribute to illness severity and death.1,2 Humoral immune responses play important roles in therapy and prophylaxis for SARS-CoV-2 infection.Since the receptor-binding domain(RBD)of the SARS-CoV-2 spike(S)glycoprotein binds to angiotensin-converting enzyme 2 to trigger virion endocytosis,antibodies against this domain may be able to neutralize SARS-CoV-2 and possibly provide protective immunity in COVID-19 patients.3 Clinical trials investigating the administration of convalescent plasma and the interleukin(IL)-6 antagonist tocilizumab to treat COVID-19 patients are currently underway,4 but the overly robust expansion of antibody-secreting cells(ASCs)could play a major role in the pathogenicity of SARS-CoV-2 in COVID-19 patients.5 Thus,a detailed characterization of the associations between humoral immune responses and inflammatory factors could result in a better understanding of SARS-CoV-2-host interactions in COVID-19 patients.
Objective: Ras association domain family 1 A (RASSF1A) has been regarded as a biomarker predicting the prognosis of non-small cell lung cancer (NSCLC), but previous findings are inconsistent. This meta-analysis of prospective studies aimed to assess the value of RASSF1A methylation in predicting the prognosis of NSCLC patients. Methods: Studies were searched in PubMed and Web of Science. The estimates of the effects and the corresponding 95% confidence intervals (95% CIs) were used for the analyses. The overall effects of RASSF1A methylation on overall survival (OS) were estimated, after which subgroup analysis based on regions was conducted. Sensitivity analyses were conducted to restrict the studies with certain features. Results: A total of 16 studies with 2210 participants were included in this meta-analysis. The overall analysis result indicated that RASSF1A methylation had no statistically significant effects on OS of NSCLC patients (HR = 1.28; 95% CI 0.86-1.70), which were confirmed by the subgroup analysis. However, the sensitivity analysis indicated that RASSF1A methylation from lung cancer tissues was significantly associated with lower OS (HR = 1.24; 95% CI 1.04-1.45). Conclusion: RASSF1A methylation in lung cancer tissue can serve as a prognostic factor of NSCLC. More studies are needed to uncover the underlying mechanisms.
Background: Hydroa vacciniforme–like lymphoproliferative disorder (HVLPD) is a rare Epstein–Barr virus (EBV)-associated lymphoma that mainly affects children. Objectives: To examine the similarities and differences in the clinical pathological features, EBV infection status, and gene rearrangements in adults and children patients with HVLPD. Methods: We compared the clinical manifestations, histopathology, immunophenotypical features, EBV infection status, and T-cell receptor gene rearrangements in the adult and children HVLPD groups. Results: Clinical manifestations differed between children and adults groups. The children were characterized by blisters and severe facial swelling, whereas the adults were characterized by mild facial swelling and papules. Mosquito bite was significantly related to morbidity in the children group. Histologically, the number of mast cells in the adult group was greater than in the children group (P < 0.05). There were no significant differences in EBV infection status or TCR-γ gene rearrangements between 2 groups. Conclusions: There were differences in clinical pathology and prognosis between the 2 groups. A higher mast cell count and T-cell phenotype might be associated with a poor prognosis.
In the present study, DNA methylation level of CD4+ T cells exposed to ultraviolet B (UVB) was investigated and its potential mechanisms were also explored.
Background: Ankylosing spondylitis (AS) is an inflammatory rheumatic disease and strongly associated with an increased risk of fractures. A great proportion of patients with AS are suffering from sustaining fractures and the aim of this study is to evaluate and quantify the association between the site of the fracture and AS by performing a meta-analysis. Methods: A systematic literature search was performed on Medline database from 1966 to August 15, 2016 and Embase database from 1980 to August 15, 2016. Studies were evaluated by 2 independent reviewers and quantitative estimates regarding the association between ankylosing spondylitis and the risk of any, hip, or vertebral fracture were presented. After the heterogeneity of selected studies was assessed by using Cochran I2 statistics, the random effect model was used to combine effect size. Publication bias was measured by Egger and Begg's regression tests. Results: A total of 6 articles were involved in our study. The results of meta-analysis revealed that AS was strongly associated with the risk of vertebral fracture (odds ratio [OR] = 4.25, 95% confidence interval [CI] = 1.07–7.42) and was not significantly associated with the risk of any fracture (OR=2.00, 95%CI = 0.94–3.06) or hip fracture (OR=1.28, 95%CI =0.16–2.40). Conclusion: In the present study, a general knowledge of the association between AS and the risk of 3 kinds of fractures were presented, which could improve the ways of prevention of fracture in the patients with AS.
OBJECTIVE:Elevated serum IgG4 levels are an important hallmark for diagnosing IgG4-related disease (IgG4-RD) but can also be found and reported in other diseases. The present study intended to compare the serum IgG4 levels in both IgG4-RD and non-IgG4-RD and determine the serum IgG4 levels in patients with IgG4-RD before and after glucocorticoid therapy.METHODS:The study included 323 patients from Anhui Medical University Affiliated Provincial Hospital (China) and was conducted from July 2014-January 2016. A total of 25 patients were eventually diagnosed as having IgG4-RD, according to the IgG4-RD diagnostic criteria. Our study also included 108 patients with connective tissue disease, 94 patients with pancreatic lesions, 66 patients with bile duct lesions, 13 patients with carcinoma of the duodenal papilla and 20 control participants. The assay for serum IgG4 detection was peformed using the nephelometric method.RESULTS:Elevated levels of serum IgG4 (>1.35g/L) were detected in all patients with IgG4-RD, and reduced levels of serum IgG4 (<1.35g/L) were found in all patients with non-IgG4-RD. The serum IgG4 level in patients with IgG4-RD after glucocorticoid therapy was significantly lower than that before glucocorticoid therapy (t = 2.426, P = 0.04).CONCLUSIONS:High levels of IgG4 were observed in IgG4-RD. However, a diagnosis of IgG4 disease can not only be dependent on the detection of elevated serum IgG4 levels but also may need clinical manifestations, serology, histopathology and other comprehensive information for verification.
Systemic lupus erythematosus (SLE) is an autoimmune disease. The friend leukemia insertion site 1 (Fli-1) belongs to the Ets family of transcription factors. Recent findings suggested that expression of Fli-1 was abnormal in SLE patients and lupus mice. In addition, functional analysis indicated that Fli-1 plays a key role in the development of this complex autoimmune disorder. Here, we review the updated evidence indicating the roles of Fli-1 in autoimmune lupus. Hopefully, the information obtained may result in a better understanding of the pathogenesis of the systemic autoimmune disease.
The aim of this study was to investigate the mRNA expression levels of suppressor of cytokine signaling 1 (SOCS1) in patients with systemic lupus erythematosus (SLE) compared with healthy controls. The associations of systemic lupus erythematosus disease activity index scores and clinical features of SLE with the expression levels of SOCS1 mRNA were also evaluated. Real-time quantitative reverse transcription-polymerase chain reaction was applied to detect the mRNA expression levels of SOCS1 in peripheral blood mononuclear cells from 34 patients with SLE and 34 healthy controls. The mRNA expression level of SOCS1 was significantly decreased in SLE patients in comparison with healthy controls (Z = −4.207, P < 0.001). Lower SOCS1 mRNA expression was detected in active SLE patients when compared with inactive ones (Z = −2.428, P = 0.015). There was no significant difference found for the SOCS1 mRNA levels between SLE patients with nephritis and those without (Z = −0.642, P = 0.521). The presence of photosensitivity, proteinuria, positive antinuclear antibody, and C4 decline were associated with SOCS1 mRNA levels in SLE patients (all P < 0.05). Furthermore, the SOCS1 mRNA expression was negatively correlated with disease activity (r s = −0.372, P = 0.030). Our results suggest that the dysregulation of SOCS1 might be associated with the pathogenesis of SLE.
Objective: The aim of this study was to determine whether CTLA-4 gene variants were associated with susceptibility to inflammatory bowel disease (IBD).Methods: Meta-analysis was conducted on the association between CTLA-4 variants and IBD using: (I) allelic contrast, (2) the recessive model, and (3) the dominant model.Results: A total of 9 relevant studies including 1739 Crohn's disease (CD) cases, 10 relevant studies containing 1017 ulcerative colitis (UC) cases and 2685 healthy controls were involved in this meta-analysis. Overall, CTLA-4+49A/G, -318C/T and CT60 variants were not associated with IBD susceptibility in all genetic models (P > 0.05). Stratification by ethnicity indicated a significant association between the CTLA-4+49A/G variant and CD in Caucasian group (GG vs. GA + AA: OR = 0.723, 95% CI = 0.564-0.926, P = 0.010). In Asian group, meta-analysis showed a significant association between the CTLA-4 CT60 variant and UC (AA vs. AG + GG: OR = 0.375, 95% CI = 0.163-0.861, P = 0.021).Conclusions: Based on the published literature, this meta-analysis suggests that the CTLA-4+49A/G variant may be related to CD susceptibility in Caucasians, and the CTLA-4 CT60 variant may be associated with UC susceptibility in Asians. 2013 American Society for Histocompatibility and Immunogenetics. The aim of this study was to determine the risk factors for avascular necrosis (AVN) in patients with systemic lupus erythematosus (SLE). Four electronic databases (PubMed, EMBASE, Ovid, and Science Direct) were searched for. The search was performed to identify the articles as to SLE with AVN before September 2013. The clinical and laboratory data were extracted, and a meta-analysis was performed to identify the risk factors for AVN in patients with SLE. Publication bias was assessed with funnel plot and Egger’s test. A total of 995 papers were found from the four databases; 16 studies were finally included. Pooled analysis showed the following result. The result showed that arthritis (odds ratio (OR) = 2.448, 95 % confidence interval (CI) = 1.617–3.707), cushingoid (OR = 3.890, 95 % CI = 1.591–9.510), gastrointestinal involvement (OR = 2.054, 95 % CI = 1.283–3.290), hypertension (OR = 1.482, 95 % CI = 1.093–2.008), oral ulcers (OR = 1.877, 95 % CI = 1.182–2.979), pleuritis (OR = 2.302, 95 % CI = 1.325–4.001), renal disease (OR = 1.475, 95 % CI = 1.124–1.936), and vasculitis (OR = 2.591, 95 % CI = 1.358–4.944) were relevant with AVN in SLE patients. Cytotoxic drug (OR = 1.834, 95 % CI = 1.065–3.156, P = 0.029), the total cumulative dose (Standard Mean Difference (SMD) = 1.104, 95 % CI = 0.118–2.090, P = 0.028), maximum daily dose (SMD = 0.484, 95 % CI = 0.288–0.678, P < 0.001), and mean daily dose (SMD = 1.305, 95 % CI = 0.061–2.549, P = 0.040) were significantly higher in AVN group. There were no significantly laboratory features that appeared in this pooled analysis. We conclude that arthritic, cushingoid, gastrointestinal involvement, hypertension, oral ulcers, pleuritis, renal disease, vasculitis, cytotoxic drug, and steroid treatment may contribute to AVN in SLE patients.
目的 检测转录因子Fli-1蛋白在外周血T细胞与B细胞上的表达水平,分析其表达水平与系统性红斑狼疮(systemic lupus erythematosus,SLE)患者疾病活动度、实验室指标及临床表现之间的关系.方法 选取来 自安徽医科大学附属省立医院的28例SEE患者作为病例组,选择25例健康志愿者作为正常对照.利用流式细胞仪(flow cytometry,FCM)检测外周血单核细胞(peripheral blood mononuclear cells,PBMCs)T细胞和B细胞中Fli-1蛋白表达情况.结果 SLE患者外周血T细胞上Fli-1平均荧光强度(mean fluorescence intensity,MFI)高于对照组,差异有统计学意义(t =3.28,P=0.003),而SLE病例与对照组间,Fli-1在外周血B细胞上的表达差异无统计学意义(Z=-0.87,P=0.383).SLE病人外周血T/B细胞上Fli-1的表达水平与糖皮质激素的平均用量间均呈正相关(均有P<0.05),但并未发现Fli-1在T细胞和B细胞上的表达水平与SLE疾病活动指数间的关联.对多项临床实验室检测指标与Fli-1的关联分析结果显示,低补体的SEE患者,其Fli-1在外周血T细胞与B细胞上的表达水平均高于无低补体表现的SLE患者(均有P<0.05).结论 SLE病人外周血T细胞上Fli-1表达水平增加,提示Fli-1可能参与了SLE的发病,其具体作用机制仍有待进一步研究.
OBJECTIVEConsidering inconclusive and heterogenous results, the aim of this study was to investigate the risk of thyroid cancer in systemic lupus erythematosus (SLE) by a quantitatively systematic review with meta-analysis.METHODSElectronic database of PubMed was searched for studies characterizing the associated risk of thyroid cancer in patients with SLE. The meta-analysis procedure was used to combine standardized incidence rates (SIRs) with 95% confidence intervals (CIs) to evaluate the association.RESULTSSeven cohort studies fulfilled the inclusion criteria and were subjected to the final analysis in the meta-analysis. Homogeneity was confirmed across the included studies. The pooled SIR based on a fixed-effect model was 2.22, with a 95% CI of 2.11-2.34. Sensitivity analyses suggested that the summary statistics obtained should approximate the actual average.CONCLUSIONFindings from this meta-analysis revealed the positive association between thyroid cancer and SLE risk. Individuals with SLE have a heightened risk of developing thyroid cancer.
