本研究旨在观察肝癌大鼠模型经射频消融术后乏氧微环境中缺氧诱导因子(HIF)-1α、基质金属蛋白酶(MMP)-9、血管内皮生长因子(VEGF)、转化生长因子-β1(TGF-β1)表达的影响,现报道如下。
Objective: To investigate the effects of EPBD combined with LC on operation, abdominal pain, inflammatory factors, serum amylase level and adverse reactions. Methods: 102 cases of cholecystolithiasis with choledocholithiasis treated in the digestive department of our hospital from December 2018 to December 2020 were selected as the research object. According to the surgical treatment scheme, they were divided into EST experimental group and EPBD experimental group, with 51 people in each group. The EST group was treated with LC + EST and the EPBD experimental group was treated with LC + EPBD. The operation and postoperative conditions of the two groups were compared and analyzed. Results: there was no significant difference in stone clearance between the two groups (P > 0.05). There was no significant difference in operation time, postoperative exhaust time and hospital stay between EPBD experimental group and EST experimental group (P > 0.05), but the amount of intraoperative bleeding in EPBD experimental group was significantly better than that in EST experimental group (P < 0.05). There was no significant difference in abdominal pain, inflammatory factors and serum amylase between EPBD experimental group and EST experimental group (P > 0.05). The incidence of adverse reactions in EPBD experimental group was significantly lower than that in EST experimental group (P < 0.05). Conclusion: EPBD combined with LC is effective in the treatment of cholecystolithiasis complicated with choledocholithiasis, and the incidence of postoperative complications is low, which is conducive to the rehabilitation of patients.
The present study aimed to investigate the effect of β-asarone treatment in a rat model of depression induced by chronic unpredictable mild stress (CUMS) and to further explore the underlying molecular mechanisms. A rat model of depression was established by subjecting rat to CUMS and treated with various concentrations of β-asarone (12.5, 25 and 50 mg/kg/day) and fluoxetine (20 mg/kg/day). Next, behavioral tests, including an open field, sucrose preference and forced swimming tests, were performed. In addition, the apoptosis of hippocampal neuronal cells was determined by flow cytometry, gene expression levels were detected by reverse transcription-quantitative polymerase chain reaction and protein levels were determined by western blot assay. The results revealed that β-asarone significantly mitigated CUMS-induced depression-like behavior, evidenced by the increased sucrose intake, crossing and rearing numbers, and decreased immobility time in the forced swimming test. Furthermore, β-asarone significantly decreased the apoptosis rate of hippocampal neuronal cells in rats subjected to CUMS. β-asarone was also found to enhance CREB, BDNF, Trk-B and Bcl-2 levels, and reduce Bad level in the hippocampus of CUMS-treated rats. In addition, the activation of extracellular signal-regulated kinase pathway inhibited by CUMS was promoted by β-asarone treatment. In conclusion, the present study findings indicated the antidepressant-like effects of β-asarone on CUMS-induced depression in rats.
目的:通过观察和厚朴酚对TGF-β/Smad信号通路相关指标表达的影响,进而探讨和厚朴酚抗肝纤维化-肝癌的作用机理.方法:将雄性SD大鼠随机分为空白对照组、模型组、和厚朴酚干预组和复方鳖甲软肝片组共4组,每组10只.采用二甲基乙硝铵(DEN)诱导大鼠慢性肝脏损伤后炎症—肝纤维化—肝硬化—肝细胞癌模型,应用实时定量PCR和Westernblot检测TGF-β1、Smad3和Smad7的表达.结果:PCR和Westernblot检测结果显示,与空白对照组比较,模型组TGF-β1与Smad3的mRNA与蛋白表达均明显增高(P<0.05),而Smad7的mRNA与蛋白表达均明显降低(P<0.05);与模型组相比,和厚朴酚组和复方鳖甲软肝片组TGF-β1与Smad3和Smad7的mRNA与蛋白表达水平均明显降低(P<0.05),Smad7的mRNA与蛋白表达水平均明显升高(P<0.05).结论:和厚朴酚可通过调控TGF-β/Smad信号通路来发挥抗肝纤维化-肝癌的作用.
目的 探讨砷中毒患者心彩色多普勒超声变化情况.方法 对本院接收治疗66例急性砷中毒患者为研究对象纳入实验组,以本院进行健康体检者20例为对照组.比较两组实验室检查指标尿砷浓度、血常规指标、肝肾功能指标;通过心电图、彩色多普勒超声检查探讨砷中毒患者心电图及心彩色多普勒超声变化情况.结果 实验组患者尿砷浓度显著高于对照组(P<0. 05);血常规检查中白细胞计数、红细胞计数以及血红蛋白均与对照组无显著性差异(P>0. 05),实验组血小板浓度低于对照组,差异具有统计学意义(P<0. 05);肝功能检查显示,实验组异常者有52例(丙氨酸转氨酶ALT以及天门冬氨酸转氨酶AST均显著升高),对照组患者均正常;肾功能检查显示:实验组有7例肾功能异常,主要表现为尿素氮、尿蛋白、肌酐显著升高;心电图结果显示实验组中男性异常者30例、异常率为78. 9%,女性异常率为20例、异常率为66. 7%;患者主要表现为Q-T间期延长、且心动过缓、房室早、ST段以及T波改变、心律不齐、心动过速;对照组心电图异常率为15. 0%.治疗前,实验组患者多普勒超声结果异常者28名、异常率为42. 42%,多表现为三尖瓣闭合不全、心包积液、肺动脉瓣异常、舒张功能减低;对照组异常率为0%;治疗后实验组患者多普勒超声结果异常者4名、异常率为6. 06%,对照组异常率为0%,治疗后正常率显著增高(P<0. 05).结论 砷中毒对患者心脏功能有一定损害,彩色多普勒超声及心电图检查可准确判断砷中毒患者的病变情况.
研究丹参酮ⅡA对缺血再灌注损伤大鼠心肌细胞损伤的保护作用及机制.方法利用培养的新生大鼠心肌细胞造成缺血再灌注损伤模型,CCK-8检测细胞增殖,BD流式细胞仪Annexin V-FITC/PI双染法检测细胞凋亡.结果:在不同剂量的丹参酮ⅡA干预缺血再灌注损伤大鼠心肌细胞模型后,细胞活性明显增加,差异有统计学意义(P<0.05).细胞凋亡结果显示,与Control组相比,I/R组心肌细胞凋亡显著增加,差异有统计学意义(P<0.05),丹参酮ⅡA高低中剂量组心肌细胞凋亡显著减少,差异有统计学意义(P<0.05).结论丹参酮ⅡA可以有效保护缺血再灌注损伤大鼠心肌细胞的损伤.
