目的:探讨影响Ramsay-Hunt综合征的预后因素,为临床进一步治疗及预后判断提供科学依据.方法:对2016年9月~2021年3月在西安市中医医院接受治疗的52例Ramsay-Hunt综合征患者的人口学资料、临床特征及治疗情况进行回顾性分析,以House-Brackmann(HB)分级法作为面神经功能的评估标准,根据患者治疗后的面瘫恢复情况分为预后良好组(n=37)和预后不良组(n=15),采用Logistic多因素回归分析影响Ramsay-Hunt综合征面瘫的预后相关因素.结果:性别、发病侧别、冠心病、高脂血症、有无听力下降及头晕、针灸治疗时间与Ramsay-Hunt综合征面瘫的预后相关性无统计学差异.多因素回归分析显示,发病年龄、糖尿病、病初面瘫程度、开始治疗时机、治疗方案对预后判断有统计学意义(P<0.05).结论:年龄、发病时面瘫严重程度、糖尿病、开始治疗时机及治疗方案是影响Ramsay-Hunt综合征面瘫的预后独立因素.
目的:探讨甲状腺良恶性肿瘤患者血清维生素D水平与甲状旁腺激素(parathyroid hormone,PTH)及血清钙离子的相关性.方法:收集本院102例甲状腺乳头状癌(PTC)及结节性甲状腺肿患者,检测甲状腺良恶性肿瘤患者血清25-羟基维生素总D3、PTH、血清钙离子水平,比较血钙与维生素D及PTH的相关性.结果:PTH与维生素D呈负向关系(r=-0.161,P=0.105),相关系数无统计学意义(P>0.05).血钙与P T H呈正向关系(r=0.116,P=0.247),相关系数无统计学意义(P>0.05).血钙与维生素D呈正向关系(r=0.144,P=0.148),相关系数无统计学意义(P>0.05).就相关系数而言,相较于PTH,血钙水平与维生素D关系更密切.结论:甲状腺良恶性肿瘤患者血清维生素D与甲状旁腺激素呈负向关系,但无线性相关;血钙、维生素D、甲状旁腺激素三者之间均无线性相关.
目的:初步探讨北方汉族人DNA修复能力(DNA repair capacity,DRC)的水平与头颈鳞癌发病风险的相关性,为头颈鳞癌的诊断提供新的检测标志物.方法:收集71例头颈鳞癌患者和65例健康对照,均为我国北方地区汉族人.通过宿主细胞再活化(host cell reactivate,HCR)实验检测研究对象外周血淋巴细胞DRC的表达水平.对头颈鳞癌病例组和对照组之间一般特征的差异进行卡方检验,通过t检验及Wilcoxon秩和检验分析两组间DRC水平的差异.通过logistic回归模型计算优势比(OR值)及95%可信区间(95% CI).此外,我们通过logistic模型计算ROC曲线下面积,进一步评价DRC模型的诊断价值.结果:头颈鳞癌组中DRC的水平在统计学上低于对照组(P=0.007).在logistic回归模型分析中,矫正完年龄、性别、吸烟状况和饮酒因素后,DRC的水平与头颈鳞癌患病风险关系的ORs,在低水平与其DRC高水平相比为2.35(95% CI,1.11-4.98).此外,DRC的水平降低与头颈鳞癌风险增加之间也存在剂量反应关系.最后,ROC曲线模型提示DRC模型中曲线下面积有所改善(P=0.068).结论:北方汉族人中DRC水平的降低与头颈鳞癌发病风险的增加相关.本研究结果需在更大样本的后续研究中进一步验证.
Objective To investigate the effect of siRNA interference β-catenin gene expression on the proliferation,apoptosis and inva-sion of Hep-2 cells in larynx cancer. Methods The larynx cancer Hep-2 cells were randomly divided into blank control group,negative control group and interference group. The siRNA of target β-catenin were transfected in the interference group,and blank siRNA in negative control group were transfected. The expression of β-catenin mRNA and protein were detected by fluorescent quantitative PCR and Western-blot,cell proliferation was detected by MTT,cell apoptosis was detected by flow cytometry. Transell assay was used to observe the cell invasion ability. Re-sults The β-catenin mRNA and protein expression in the interference group were(0.304 ± 0.015)and(0.234 ± 0.065),significantly lower than those in the blank control group and negative control group(P <0.05). The absorbance of cultured 48 h and 72 h in the interference group were(0.721 ± 0.250)and(0.942 ± 0.321),which were significantly lower than those in the blank control group and the negative control group (P <0.05). The apoptotic rate in the interference group was(19.10 ± 1.24)%,which was significantly higher than that in the blank control group and the negative control group(P <0.05). The number of cells through the cell membrane in the interference group was(22.41 ± 3.25), which was significantly lower than that in the blank control group and the negative control group(P <0.05). Conclusion Interfering β-cate-nin gene expression can inhibit the proliferation and invasion,and promote cell apoptosis of larynx cancer Hep-2 cells.
OBJECTIVE To determine the effect of JAK kinase inhibitor AG490 on proliferation and apoptosis of nasopharyngeal carcinoma cell line C666-1 and explore the inhibitory effect of AG490 on STAT3 signal transduction pathway. STUDY DESIGN An experimental study. PLACE AND DURATION OF STUDY Xi'an Ninth Hospital, Xi'an, China, from May 2015 to December 2016. METHODOLOGY AG490 was applied to C666-1 cells. Cell proliferation was detected by MTTassay. Cell cycle and apoptosis were detected by flow cytometry. The expression of STAT3 and p-STAT3 protein was detected by immobilized Western blot. RESULTS AG490 could effectively inhibit proliferation of C666-1 cellsin vitro, and the inhibitory effect was characterized by time and concentration dependence. AG490 induced apoptosis of nasopharyngeal carcinoma cells, and apoptosis rate increased with prolongation of the time. AG490 could inhibit expression of STAT3 and p-STAT3 protein in C666-1 cells. CONCLUSION AG490 can down-regulate expression of STAT3 and p-STAT3 protein in C666-1 cells, inhibit proliferation of nasopharyngeal carcinoma cells and promote apoptosis of nasopharyngeal carcinoma cells.
目的:探讨巴曲酶与血管扩张剂治疗突发性耳聋的临床疗效及安全性.方法:将96例突发性耳聋患者随机分为观察组与对照组,观察组48例给予巴曲酶治疗,对照组48例给予血管扩张剂治疗,对比两组患者治疗临床疗效,治疗前后血浆纤维蛋白原(Fg)、凝血酶时间(TT)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)等,并观察两组患者不良反应发生情况.结果:与对照组相比,观察组总有效率明显升高(P<0.05);与治疗前比较,治疗后观察组与对照组患者的FG水平明显降低,TT、PT及APTT明显延长,且观察组较对照组各指标变化更显著(P<0.05);与对照组比较,观察组患者眩晕、耳鸣及听力缓解时间均明显缩短(P<0.05).结论:巴曲酶较血管扩张剂治疗突发性耳聋疗效更显著,且能改善患者凝血功能状态.
Objective To explore the functions of microRNA(miR)-19b and miR-185 on proliferation and apoptosis of laryngeal carcinoma cells.Methods The laryngeal squamous carcinoma cell line (Hep-2 cell) as control group,construction the miR-19b and miR-185 deletion mutants by the technology of Crisp-Case 9 as miR-19b and miR-185 deletion groups,and building miR-19b and miR-185 overexpression mutants by cell transfer dyeing technology as miR-19b and miR-185 overexpression groups.Flow cytometry was used to detect the apoptosis of control group,miR-19b,miR-185 deletion groups and miR-19b,miR-185 overexpressing groups.The apoptosis of laryngeal carcinoma cells in five groups was detected by flow cytometry.The proliferation of Hep-2 cell lines (expressed by OD450nm) and the invasive ability (expressed as the number of invasive cells) were measured under different conditions.Results After 64 h culture,the proliferation ability of miR-19b and miR-185 overexpression groups and miR-19b,miR-185 deletion groups were 0.65 ± 0.01,0.63 ± 0.01,1.69 ± 0.03,1.73 ± 0.02,had significant difference with that in control group,which was 1.28 ±0.02 (P <0.01).The apoptosis of miR-19b and miR-185 overexpression groups and miR-19b,miR-185 deletion groups were (12.32 ± 1.39)%,(11.74 ± 1.08) %,(3.28 ± 0.11) %,(3.48 ± 0.12) %,had significant difference with that in control group,which was (5.34 ± 1.27) % (P <0.01).The number of cells in over expression of miR-19b and miR-185 overexpression groups and miR-19b,miR-185 deletion groups were 12 ±2,9 ± 3,47 ± 7,49 ± 8,had significant difference with that in control group,which was 36 ± 5 (P < 0.05).Conclusion miR-185 and miR-19b can significantly inhibit the proliferation and invasion of laryngeal squamous cell carcinoma.
