Candida auris is an emerging multidrug-resistant fungal pathogen causing nosocomial transmission and invasive infections with high mortality. This study aimed to investigate the genetic relationships, enzymatic activities, and drug-resistance profiles of C. auris isolates to evaluate the population and epidemiological diversity of candidiasis in Russia. A total of 112 clinical isolates of C. auris were analyzed from May 2017 to March 2023 in 18 hospitals across Saint Petersburg, the Leningrad Region, and Moscow. Species identification was confirmed by ITS sequencing, and genotyping was performed using 12 short tandem repeat (STR) markers. Antifungal susceptibility was tested using Sensititre™ YeastOne™ plates, and hydrolytic enzyme production was measured by the plate method. ITS sequencing confirmed that all isolates belonged to a single ITS cluster (clades I and III). Fifteen distinct STR genotypes were identified, with genotype I being dominant (n = 53). The most variable of the analyzed markers turned out to be M3-Ia, which was represented in the Russian population by eight different variants. Fluconazole resistance was found in 111 isolates, 17% were resistant to amphotericin B, and 3.6% to 5-flucytosine. Phospholipase activity was strong in most strains, especially in urine isolates (p = 0.014). Conclusion: The predominance of STR genotype I and its variability at the M3-Ia locus suggest its association with nosocomial outbreaks and transmissibility in Russia.
We present a case of fungal peritonitis in a patient secondary to intestinal perforation and repeated laparotomy. An analysis of data from the register of patients with invasive candidiasis is also presented. In the study were included 42 patients with candida peritonitis. The risk factors: antibiotics (100%), vascular catheters (95%), total parenteral nutrition (81%), repeated abdominal surgery (69%), sepsis (69%), bacteremia (50%), repeated perforations on the gastrointestinal tract (43%), oncopathology of the gastrointestinal tract (36%), infected pancreatic necrosis (26%). The etiology agents were C. albicans (50%), C. glabrata (14%), C. parapsilosis (7%). 45% of patients received prophylactic treatment (100% fluconazole). Empirical therapy was carried out by 52% of patients (triazoles – 38%, echinocandins – 14%). After receiving the results of the microbiological study, 3% of patients began to receive antifungal therapy (echinocandins). The 30 days overall survival rate was 66%.
Background The Trichophyton rubrum species group consists of prevalent causative agents of human skin, nail and hair infections, including T. rubrum sensu stricto and T. violaceum , as well as other less well established or debatable taxa like T. soudanense , T. kuryangei and T. megninii . Our previous study provided limited evidence in favour of the existence of two genetic lineages in the Russian T. rubrum sensu stricto population. Objectives We aimed to study the genetic structure of the Russian population of T. rubrum , and to identify factors shaping this structure. Methods We analysed the polymorphism of 12 simple sequence repeat (SSR, or microsatellite) markers and single-nucleotide polymorphism in the TERG_02941 protein-coding gene in 70 T. rubrum isolates and performed a phylogenomic reconstruction. Results All three types of data provided conclusive evidence that the population consists of two genetic lineages. Clustering, performed by means of microsatellite length polymorphism analysis, was strongly dependent on the number of nucleotide repeats in the 5’-area of the fructose-1,6-bisphosphate aldolase gene. Analysis of molecular variance (AMOVA) on the basis of SSR typing data indicated that 22–48% of the variability was among groups within T. rubrum . There was no clear connection of population structure with types of infection, places of geographic origin, aldolase gene expression or urease activity. Conclusion Our results suggest that the Russian population of T. rubrum consists of two cosmopolitan genetic lineages.

Candida auris is an emergent yeast pathogen, easily transmissible between patients and with high percent of multidrug resistant strains. Here we present a draft genome sequence of the first known Russian strain of C. auris, isolated from a case of candidemia. The strain clustered within South Asian C. auris clade and seemingly represented an independent event of dissemination from the original species range. Observed fluconazole resistance was probably due to F105L and K143R mutations in ERG11.
