Изучали влияние острого введения нового ноотропного препарата фенотропила (N-карбамоил-метил-4-фенил-2-пирролидон) в дозе 100 мг/кг на количественные характеристики рецепторов дофамина (DA), серотонина (5-HT), глутамата (NMDA), ГАМК-А (БДЗ) и ацетилхолина (nACh) у крыс, обученных навыку пассивного избегания (УРПИ), в норме и на фоне скополаминовой амнезии в условиях ex vivo. Установлено, что под действием холиноблокатора скополамина происходит существенное увеличение плотности (Bmax) не только для н-холинорецепторов в коре (на 99% по сравнению с контролем), но и для NMDA-рецепторов в гиппокампе (на 93%). Однократное введение фенотропила (100 мг/кг, в/б) противодействует эффекту скополамина, уменьшая количество nACh на 46% и NMDA-рецепторов на 14%, возвращая показатели к контрольным значениям. Антагонистический эффект фенотропила проявляется также в действии скополамина на бензодиазепиновые и дофаминовые D1-рецепторы. Скополамин уменьшает плотность D1-рецепторов на 20% и БДЗ рецепторов на 17%, а препарат увеличивает их количество на 16% и на 25%, соответственно. Фенотропил значительно повышает плотность дофаминовых D2 на 29 % и D3 на 62%, причем сам скополамин также увеличивает количество D3-рецепторов на 44% по сравнению с контролем. Изменений в характеристиках связывания 5-HT2-рецепторов ни при скополаминовой амнезии, ни при действии фенотропила обнаружено не было. Полученные результаты демонстрируют роль изученных рецепторов как в развитии скополаминовой амнезии, так и в нейрохимических механизмах антиамнестического действия ноотропного препарата фенотропила.
The purpose of the work is to asses reversibility bounds of repetitive functional disruptions of two-way avoidance responses in rats provoked by sudden changes of cause-effect («smash» of avoidance responses) and spatial relations in experimental environment and to analyze possibilities of their use for studying neuropsychotropics effects. Statistical significant results have been obtained demonstrating the reversibility of four consecutive functional disruptions of avoidance responses caused by alternation of the smash and the spatial relearning. Our results pointed out that whatever their nature and number, all of the functional disturbances resulted in a deep statistically significant inhibition of avoidance performance followed by its complete restoration. We have shown reproducibility of nootropics piracetam (300 mg/kg) and nooglutyl (10 mg/kg) enhancing effects in the repetitive functional disruptions caused by both the smash and the spatial reverse. Anxiolytic guidazepam (2 mg/kg) precluded avoidance performing only from the smash meanwhile it has no effect on the spatial relearning. Mexidol (100 mg/kg) and IP-5 (0,5 mg/kg) (agents with anxiolytic and nootropic properties) diminished the functional disruption of avoidance responses in case of the spatial relearning but did not precluded them from the smash if it was applied as a second functional disruption. It differs these substances from either nootropics which are effective in both of the models or anxiolytics which are never effective in spatial relearning but effective in the smash of avoidance response.
