High-grade B-cell lymphomas with MYC and BCL2 and/or BCL6 rearrangements (HGBL-D/TH) is a group of aggressive lymphomas with poor clinical outcome following standard immunochemotherapy. A panel of cell lines faithfully representing this lymphoma is desirable for further understanding its biology and developing better treatment regimens. We generated one double hit (COH-DHL) and one triple hit cell line (COH-THL1) from two patients. The two cell lines were extensively characterized, along with two reported lines CS-THL1 and DOGKIT. The data were compared with four previously described D/TH lymphoma lines and one BL line, Jijoye. The D/TH lymphoma cell lines showed similar expression profile to BL and DLBCL with closer similarity to BL. Many of the frequent mutations (CREBBP, KMT2D, EP300 and EZH2) were shared with FL and GCB-DLBCL. Genomic copy number abnormalities (gCNAs) showed that these lines had more gains than losses with some of these harboring know oncogenes or tumor suppressor genes. Genes within the gCNA were enriched in pathways regulating apoptosis, cell cycle, and TGFB, TP53, NF-kB, PKA and interferon signaling. Five D/TH lymphoma cell lines and Jijoye, were treated with 10 different drugs targeting Myc and/or BCL2 and other critical cellular pathways. Silvestrol and Dinaciclib generally showed outstanding activities and the newly derived cell lines tended to be most responsive. The BL cell line Jijoye showed a completely different pattern of response. A panel of well characterized D/TH lymphoma lines would be very helpful in preclinical studies of targeted agents.
BACKGROUND AND OBJECTIVES:Metabolic syndrome (MetS) is an independent risk factor for cardiovascular disease (CVD), in which platelet hyperactivation plays a pivotal role. The purpose of this study was to evaluate platelet function in MetS patients using Platelet Function Analyzer-100 (PFA-100) and to explore the risk factors for platelet hyperactivity in MetS.SUBJECTS AND METHODS:We investigated participants who were enrolled for health check-up in our department. Routine physical examinations and fasting blood sample tests were performed when participants visited the hospital. MetS was defined as ≥3 of the risk factors according to the Harmonised criteria: central obesity, hypertension (HP), hypertriglyceridemia, low high density lipoprotein cholesterol and hyperglycaemia. Participants were divided into a MetS group (≥3), normal control (NC) group (0) and non-MetS group (1-2) according to the numbers of the five risk factors. Platelet function was tested by PFA-100, which measures the time taken for blood to occlude an aperture (closure time [CT]). All continuous data were compared using Student's t test or Mann-Whitney U test according to the data distribution. Categorical data were compared using the chi-square test. Logistic regression was used to investigate the independent risk marker for PFA-100 CT values.RESULTS:A total of 831 participants (611 males and 220 females) was included in our subject. The MetS group had significantly shorter CT values compared with the NC group (106 (52-181) s vs 111 (70-210) s, P < .05) and the non-MetS Group (106 (52-181) s vs 113 (73-197) s, P < .05). Higher body mass index, hypertriglyceridemia and HP were correlated with shorter CT values (P < .05). Logistic regression analyses indicated that hypertriglyceridemia was an independent risk marker for shorter PFA-100 CT values (P < .05).CONCLUSION:Our results indicate the presence of platelet hyperactivation in MetS patients and that hypertriglyceridemia is an independent risk marker for it. Triglyceride-lowering treatment may reduce CVD risk in MetS individuals.
Increasing evidence suggests that uric acid (UA) levels may play a role in the MetS. The purpose of this study was the association of serum uric acid with metabolic syndrome and its components from a single‐clinical centre in China.
By improving the accuracy of sales forecasting, this paper provides support for fashion product sales enterprises to make better inventory management and operational decisions. The deep neural network is introduced into the construction of multimodal features, and the internal structure of different modes, such as historical sales features, picture features, and basic attribute features of products, are fully considered, and finally the sales forecasting model of fashion products based on multimodal feature fusion is constructed. In addition, combined with the actual data of the enterprise, the proposed model is compared with the exponential regression model and shallow neural network model. The paper finds that multimodal features and deep learning representation method has better performance than traditional methods (exponential regression and shallow neural network) in the task of predicting sales of fashion products. The results help enterprises use the deep learning method and the data of multiple modal to make accurate sales forecast.
Purpose: To reduce the miss diagnosis under negative result of magnetic resonance cholangiopancreatography (MRCP), a correlation analysis between biochemical indicators and the common bile duct (CBD) stones was performed in this study. Method: A total of 514 patients following the Laparoscopic cholecystectomy with the CBD exploration were enrolled. The biochemical indicators including gamma-glutamyltransferase (GGT) and common bile duct diameter (CBDD), as well as imagological examination results including MRCP were evaluated to reveal the correlations with CBD stone using receiver operating characteristic (ROC) curve and logistic regression analyses. Result: MRCP and CBDD were two outstanding indexes related with the CBD stone. ROC analysis showed that ALP was the most outstanding index for CBD stone patients with negative MRCP, followed by direct bilirubin (DB) and GGT. Meanwhile, ROC analysis and logistic regression analyses showed that GGT was the most outstanding index for CBD stone patients with the negative MRCP combined to normal width CBDD (CBDD <0.8 cm), followed by alkaline phosphatase (ALP) and DB. Conclusion: ALP, DB and GGT might be potential predictors for CBD stone patients when MRCP is negative. If the result of MRCP was negative, especially combined with CBDD <0.8 cm, a further GGT investigation was suggested to determine whether the LCBDE should be performed. (C) 2019 Elsevier B.V. Objective To investigate arterial endothelial dysfunction in stable chronic obstructive pulmonary disease (COPD) patients and the correlation between the degree of endothelial dysfunction and the severity of COPD.Methods Forty stable COPD patients were enrolled in a COPD group and 30 non-COPD individuals in a control group.The endothelium-dependent flow-mediated vasodilatation (FMD) of the brachial artery and serum eNO value were measured in both groups.Forced expiratory volume in 1 second (FEV1)/prediction of FEV1 was determined and expressed as FEV1 (% pred).Results The mean FMD was significantly lower in the COPD group (11.21±5.19) % than in the control group (19.86±5.24)% (t=6.090,P=0.001).The Pearson's correlation analysis showed FMD was positively correlated with FEV1 (%pred) in COPD patients (r=0.440,P<0.05).The mean serum eNO level in the COPD group (108.58 ± 42.22) μmol/L was significantly lower than in the control group (151.17 ± 97.40)μmol/L (t =2.242,P =0.039).Conclusions The endothelium-dependent flow-mediated vasodilatation is significantly impaired in stable COPD patients,and the degree of impairment is proportional to the FEV1 (% prediction of FEV1) in COPD patients.
Aberrant expression of nephroblastoma overexpressed (NOV) has been evident in certain malignancies.In the current study, we aim to investigate the role played by NOV in colorectal cancer (CRC).NOV expression was determined in a cohort of 359 CRC tissues and 174 normal colorectal tissues.Its impact on CRC cells was investigated using in vitro NOV knockdown and overexpression models.NOV transcripts were reduced in the CRC tumours compared with the paired adjacent normal colorectal tissues (p < 0.01) and was associated with distant metastases.NOV knockdown resulted in increased cell proliferation and invasion of RKO cells, whilst an opposite effect was seen in the HT115 NOV over expressing cells.A positive association between Caspase-3/-8 and NOV was seen in NOV knockdown and overexpression cell lines which contributed to the survival of serum deprived CRC cells.Further investigation showed that NOV regulated proliferation, survival and invasion through the JNK pathway.NOV knockdown in RKO cells reduced the responsiveness to 5-Fluorouracil treatment, whilst overexpression in HT115 cells exhibited a contrasting effect.Taken together, NOV is reduced in CRC tumours and this is associated with disease progression.NOV inhibits the proliferation and invasion of CRC cells in vitro.Inhibition of proliferation is mediated by a regulation of Caspase-3/-8, via the JNK pathway, which has potential for predicting and preventing chemoresistance.
