Objective:To observe the effect of electroacupuncture on reducing the inflammatory response of synovial fibroblasts in rheumatoid arthritis rats by regulating lncRNA NEAT1.Methods:Sixteen 2-month-old rats were randomly divided into a blank control group and an electroacupuncture group using a random number table method,with 8 rats in each group.Rats in the blank control group were not given intervention,while those in the electroacupuncture group were given electroacupuncture intervention,and serum from the two groups was prepared separately.The synovial fibroblasts of 3-week-old SD rats were obtained by continuous enzymatic digestion,and identified by vimentin immunohistochemical staining.Then,the cultured synovial fibroblasts in the 6-well plate were randomly divided into a blank control group,a model control group,and an electroacupuncture group.The blank control group was intervened with serum from the blank control group,while the model control group was induced inflammation by serum with 10 ng·m L -1 interleukin-1β(IL-1β)from the blank control group,and the electroacupuncture was intervened by serum containing 10 ng·m L -1 IL-1β from the electroacupuncture group.Real-time PCR was used to detect the level changes of lnc RNA NEAT1 in synovial fibroblasts of each group.Western blot was used to detect protein expression contents of MMP-3,RELA and TNF-α.Results:After vimentin immunohistochemical staining,the rat synovial fibroblasts were positive for vimentin staining,which confirmed the successful extraction of fibroblasts.Compared with the blank control group,the relative expression level of lnc RNA NEAT1 in the model control group was significantly increased(P < 0.05).Compared with the model control group,the relative expression level of lnc RNA NEAT1 in the electroacupuncture group was significantly reduced(P < 0.05).Western blot results showed that compared with the model control group,the protein expression contents of MMP-3,RELA,and TNF-α in the electroacupuncture group significantly decreased(P < 0.05).Conclusion:Electroacupuncture can alleviate the inflammatory response of synovial fibroblasts in rheumatoid arthritis rats by regulating lnc RNA NEAT1.
玻璃体混浊为眼科常见疾病,目前其患病率逐渐增多,已成为全球流行性疾病。40岁以上的中老年人为此病的高发人群,常造成患者视物模糊、视力下降,给正常的生活带来不便。玻璃体混浊是由于玻璃体变性导致玻璃体中析出有形成分,跟随眼球运动上下移动,从而影响视觉效果。超声波疗法是在常规药物的基础上配合超声波技术进行临床治疗的方法,将治疗药液滴在浸湿的纱布上,应用超声波仪,将圆形声头固定在眼睑上,治疗过程中按照左-右-上-下的顺序,指导患者眼球转动。相关研究表明,超声波治疗玻璃体混浊的疗效显著,能减轻患者临床症状,改善视力水平,有效改善玻璃体变性程度。本文旨在探讨应用超声波方法治疗玻璃体混浊的优势。
目的:探讨乌头汤通过调控软骨细胞炎性焦亡途径延缓膝骨关节炎软骨退变的作用与机制.方法:将 36 只SD大鼠(雄性,8 周龄)随机分为空白对照组、模型对照组和乌头汤组,每组 12 只.空白对照组不造模,模型对照组和乌头汤组采用改良Hulth法造模.空白对照组与模型对照组采用生理盐水(3 mg·kg-1·d-1)灌胃,乌头汤组采用乌头汤(生药浓度 4.2 g·mL-1)灌胃.干预 8 周后,麻醉状态下摘取大鼠膝关节软骨.采用Western blot检测不同组别关节软骨中核苷酸结合寡聚化结构域样受体蛋白 3(NLRP3)、半胱氨酸蛋白酶-1(Caspase-1)、白细胞介素(IL)-1β、IL-18 的蛋白含量表达;采用Real-time PCR检测不同组别关节软骨中NLRP3、Caspase-1、IL-1β、IL-18 的基因相对表达变化.结果:模型对照组和乌头汤组NLRP3、Caspase-1、IL-1β、IL-18 蛋白含量与基因相对表达量均上调,与空白对照组比较,差异有统计学意义(P<0.01);乌头汤组能够下调NLRP3、Caspase-1、IL-1β、IL-18 蛋白含量与基因相对表达量,与模型对照组比较,差异有统计学意义(P<0.05).结论:乌头汤可改善膝骨关节炎大鼠软骨NLRP3、Caspase-1、IL-1β、IL-18 的表达,抑制软骨细胞焦亡,延缓软骨退变.
目的:探讨乌头汤抑制膝骨关节炎软骨细胞氧化应激反应的途径与作用机制.方法:①采用回流法制备乌头汤药物;②软骨细胞培养、LPS诱导软骨细胞退变及Ⅱ型胶原免疫组织化学染色法进行鉴定;③采用ELISA法检测软骨细胞上清液中一氧化氮(NO)与超氧化物歧化酶(SOD)水平变化;④采用Western blot法检测软骨细胞中核因子NF-E2相关因子(Nrf2)、肌动蛋白结合蛋白(Keap1)、诱导型一氧化氮合酶(iNOS)蛋白含量表达.结果:①ELISA检测结果显示,与模型对照组比较,乌头汤各浓度组中的NO水平下降(P<0.01),而SOD水平明显增高(P<0.01);②Western blot检测结果显示,乌头汤组(150μg·mL-1)对脂多糖诱导的软骨细胞干预24 h后,可显著改善软骨细胞中Nrf2、Keap1、iNOS含量表达(P<0.01).结论:乌头汤可抑制膝骨关节炎软骨细胞氧化应激反应,其作用机制与调控Nrf2/Keap1途径相关.

BACKGROUND:Alzheimer disease (AD) is a common degenerative disease of the central nervous system that can be divided into 3 stages, according to the degree of cognitive impairment. The clinical manifestations are cognitive dysfunction and memory loss, impacting the daily activities of the affected individuals. In recent years, studies have demonstrated a relationship between intestinal flora and AD. However, no meta-analysis has documented the correlation between AD and intestinal flora, to the best of our knowledge. Herein, we sought to assess the correlation between different stages of AD and intestinal flora. A systematic and comprehensive understanding of this relationship is of great significance for developing prevention and treatment strategies against AD.METHODS:A comprehensive search of the medical literature in Chinese and English language was performed in databases, such as PubMed, EBSCO, CNKI, web of science, WanFang, Cochrane Library, and CBM databases. Pre-defined search strategies were used to retrieve clinical studies of Alzheimer disease and gut microbiota. The included studies were independently analyzed by the 2 researchers who extracted the data. The quality of the data was evaluated according to the "Cochrane system evaluator manual." Finally, Endnote and RevMan software were used for systematic regression and meta-analysis of evidence.RESULTS:We documented the intestinal flora changes in the 3 stages of Alzheimer disease, according to currently available clinical evidence, and revealed the correlation between the abundance and diversity of flora and treatment efficacy. Review question / Objective: The relationship between intestinal flora and the early, middle and late stages of AD was studied retrospectively to explain the change law of intestinal flora in each stage of AD.The relationship between intestinal flora and various stages of AD was studied retrospectively.Explain the changes of intestinal flora in each stage of AD.The purpose of this study is to use a high-quality system to evaluate the changes of intestinal flora in the three stages of AD (preclinical stage, mild cognitive impairment stage and dementia stage), and to analyze the composition of intestinal flora, the abundance of opportunistic pathogenic bacteria and the abundance of beneficial symbiotic bacteria by NMA method.According to the advantages and disadvantages of exponential effect, the indexes of intestinal bacteria in different stages of AD were sorted, and the best evidence of the relationship between AD and intestinal flora in different stages was selected.This protocol is helpful to understand the impact between the early, middle and late stage of AD and intestinal flora.The understanding of the relationship between AD and intestinal flora may provide information and new treatment options for the phased treatment of AD.Our review will better review the relationship between intestinal microflora and different stages of AD, explain the different effects of different stages of ad on intestinal microflora, the change law of intestinal microflora in different stages of AD, and provide new insights and possible phased treatment schemes for the treatment of intestinal microflora in patients with AD and then change the process of ad to a certain extent.
