В нестабильных атеросклеротических бляшках сонных артерий усиливается экспрессия микроРНК miR-100, что может быть связано с вариабельностью метилирования CpG-сайтов гена MIR100 и его регуляторных регионов. Целью работы был анализ связи уровня метилирования ДНК предполагаемого регуляторного региона гена MIR100 (E-box) с клинически выраженным атеросклерозом сонных артерий методом таргетного бисульфитного секвенирования. В качестве материала использовались ткани сосудов и лейкоциты крови пациентов с клинически выраженным атеросклерозом сонных артерий (n=19), а также лейкоциты периферической крови относительно здоровых индивидов (n=18). В результате выявлена тканеспецифичность паттерна метилирования региона chr11:122,024,891-122,025,506 (сборка генома GRCh37/hg19), включающего E-box — уровень метилирования в непоражённых сонных артериях снижен по сравнению как с венами (p<0,001), так и с лейкоцитами периферической крови (p<0,007). При этом в атеросклеротических бляшках наблюдается значимое гипометилирование относительно непоражённых артерий, однако уровень метилирования ДНК в лейкоцитах крови пациентов с атеросклерозом не отличается от такового в лейкоцитах крови контрольной группы. Полученные результаты позволяют говорить о тканеспецифичности метилирования предполагаемого регуляторного региона гена MIR100 (E-box) и о возможной его связи с атеросклеротическим поражением артерий, однако метилирование исследуемого региона нельзя применять в качестве диагностического биомаркера атеросклероза. Expression of miR-100 is increased in unstable carotid atherosclerotic plaques. It can be associated with CpG sites methylation variability in the MIR100 gene region and its adjacent regulatory regions. The aim of our work was to analyze the association of DNA methylation level in the putative regulatory region of MIR100 gene with advanced carotid atherosclerosis using targeted bisulfite sequencing. We analyzed DNA samples of paired vessel tissues and blood leukocytes of patients with advanced atherosclerosis of the carotid arteries (n=19), as well as blood leukocytes of healthy individuals (n=18). As a result, we revealed tissue-specific methylation pattern of the chr11:122,024,891-122,025,506 region (GRCh37/hg19 genome assembly) including E-box (see Chen D. et al, 2014 for more information). The methylation level in unaffected carotid arteries was decreased compared to both veins (p<0.001) and peripheral blood leukocytes (p<0.007). Significant hypomethylation was observed in atherosclerotic carotid plaques relative to unaffected carotid arteries, but the level of DNA methylation in blood leukocytes of patients with atherosclerosis did not differ from that in blood leukocytes of the control group. Our findings suggest tissue-specific methylation of the putative regulatory region of MIR100 gene (E-box) and its possible association to atherosclerosis. Nevertheless, the methylation of the chr11:122,024,891-122,025,506 region cannot be used as a diagnostic biomarker of atherosclerosis.
Dysregulation of microRNA (miRNA) expression is associated with a susceptibility to many diseases, including atherosclerotic lesions of the coronary and carotid arteries and the development of clinical complications such as coronary heart disease, myocardial infarction, chronic cerebral ischemia, ischemic stroke. Recently, more and more studies analyze the miRNA regulome including a network of regulatory elements for the expression of miRNAs themselves and targets under their control. The review summarizes the data from articles concerned miRNA expression and changes in DNA methylation in the miRNA genes in human atherosclerotic arteries, as well as with the analysis of the association between single nucleotide polymorphisms and copy number variations in the miRNA genes with clinical complications of atherosclerosis.
The pandemic of coronavirus disease 2019 (COVID-19) warrants the identification of factors that may determine both risk and severity of infection. The factors include micro RNAs that have a wide regulatory potential and hence are particularly interesting. The review focuses on the potential roles of human microRNAs and the viral genome as well as microRNAs in SARS-CoV-2 infection and clinical features of COVID-19. The review summarizes the information about the human microRNAs that are thought to specifically bind to the SARS-CoV-2 genome and considers their expression levels in various organs (cells) in both healthy state and pathologies that are risk factors for severe COVID-19. Potential mechanisms whereby SARS-CoV-2 may affect the clinical features of COVID-19 are discussed in brief. The mechanisms include blocking of human microRNAs and RNA-binding proteins, changes in gene expression in infected cells, and possible epigenetic modifications of the human genome with the participation of coronavirus microRNAs.
В исследование были включены 1814 пациентов с артериальной гипертензией, ишемической болезнью сердца, мозговым инсультом и сочетанием этих заболеваний и 885 здоровых индивидов из Центральной России. Проведен молекулярно-генетический анализ 18 полиморфных вариантов генов редокс-гомеостаза (Р-Г) и анализ метилирования 4 генов Р-Г. Обсуждаются молекулярно-генетические и эпигенетические механизмы вовлеченности генов Р-Г в формирование распространенных сердечно-сосудистых заболеваний и их коморбидных форм. A total of 1814 patients with arterial hypertension, coronary heart disease, cerebral stroke and a combination of these diseases and 885 healthy individuals from Central Russia were recruited for the study. Molecular genetic analysis of 18 polymorphisms of redox homeostasis (R-H) genes and analysis of methylation of 4 R-H genes were carried out. Molecular-genetic and epigenetic mechanisms of the involvement of R-H genes in the development of common cardiovascular diseases and their comorbid forms are discussed.
В работе проведен анализ профилей метилирования гена miR-10b и гена транскрипционного фактора TWIST1, регулирующего экспрессию данной микроРНК, в кровеносных сосудах и лейкоцитах крови пациентов с клинически выраженным атеросклерозом и у относительно здоровых лиц. Обнаружено гипометилирование MIR10B в поражённых атеросклерозом артериях относительно непораженных артерий и вен. Профиль метилирования гена TWIST1 был тканеспецифичен и постоянен вне зависимости от наличия заболевания. We analyzed DNA methylation profiles within genes of miR-10b and TWIST1 regulating the expression of this microRNA in blood vessels and blood leukocytes of patients with advanced atherosclerosis and in relatively healthy individuals. MIR10B hypomethylation was detected in arteries affected by atherosclerosis in comparison with unaffected arteries and veins. Methylation profile of TWIST1 gene was tissue-specific and consistent regardless of atherosclerosis.
Методом таргетного бисульфитного секвенирования проанализирован уровень метилирования предполагаемого промоторного региона, CpG-островка и «тела» гена MIR21 в сосудах и лейкоцитах крови у пациентов с атеросклерозом сонных артерий, а также в лейкоцитах крови здоровых индивидов. Выявлено гипометилирование CpG-сайтов в гене MIR21 в сосудах по сравнению с лейкоцитами (вне зависимости от наличия заболевания). В атеросклеротической бляшке 4 последовательных CpG-сайта «тела» гена MIR21 оказались значимо гипометилированы относительно предлежащей макроскопически интактной части сонной артерии. Таким образом, установлена тканеспецифичность метилирования CpG-сайтов в гене MIR21 в сосудах, а также его связь с атеросклерозом сонных артерий. We analyzed the DNA methylation level of MIR21 gene (proposed promoter, CpG island and gene body) by targeted bisulfite sequencing in blood vessels and leukocytes of patients with carotid artery atherosclerosis and in blood leukocytes of healthy individuals. We detected DNA hypomethylation at the CpG sites in the MIR21 gene in the blood vessels compared to the leukocytes (regardless of the disease). At the same time, 4 consecutive CpG sites of the MIR21 gene body were significantly hypomethylated in atherosclerotic carotid plaque compared to nearby macroscopically intact tissue. Thus, we have shown the tissue specificity at the CpG sites in the MIR21 gene for vessels and its association with carotid artery atherosclerosis.
