Существующие методы исследования гликозилирования белков остаются громоздкими. В своем развитии они отстают от высокопроизводительных протеомных технологий. Существует реальная потребность в разработке простого, недорогого и специфичного метода выявления сайтов гликозилирования в белках, совместимого с общепринятыми процедурами подготовки биологического материала к протеомному и фосфопротеомному анализу. В статье описан новый метод дегликозилирования гликопротеинов с помощью бета-элиминирования с последующим введением ковалентной метки и визуализацией паттерна первично негликозилированных и дегликозилированных белков в предобработанном клеточном гомогенате с использованием J-агрегатов, образующихся в геле in situ. Новый метод подготовки материала к протеомному и фосфопротеомному анализу может быть использован при проведении фосфопротеомного анализа в формате RPMA для регистрации изменения фосфорилирования белкового состава клеток, в том числе в условиях развития вирусной инфекции.
Разработка методов перорального введения пептидных ростовых факторов сельскохозяйственным животным, в частности, в составе рекомбинантных пробиотических препаратов, может рассматриваться в качестве перспективного подхода к снижению себестоимости животноводческой продукции и обеспечения конкурентоспособности ее производства. Одним из наиболее эффективных в этом отношении средств является соматолиберин – релизинг-фактор гормона роста. В связи с этим целью работы было создание новых векторных систем для экспрессии полусинтетических генов соматолиберина курицы, способных стабильно функционировать в составе бациллярных штаммов со свойствами пробиотиков. Получены три конструкции, содержащие промотор гена wprА, мини-ген Е-пептида и производные гена соматолиберина, кодирующие либо пептид-предшественник (слитые GHRH и PACAP), либо фрагменты, соответствующие изолированным пептидным гормонам GHRH и PACAP. Сконструированы штаммы B. subtilis, несущие эти конструкции. Таким образом, созданы перспективные продуценты пептидных гормонов, позволяющие проводить испытания биологической активности живых пробиотических препаратов, содержащих производные соматолиберина птицы во внутриклеточной форме, путём их перорального введения.
This review is dedicated to structural and functional properties of IgA1-proteases in pathogenic microorganisms. The interest to structural and functional studies of IgA-proteases is caused essentially by their putatively great impact on set up and progression of infectious diseases. IgA degradation at mucosal surfaces is commonly suggested to be the main function of IgA1-proteases. In turn, this effect may lead to abatement of the immune response efficiency at the epithelial barrier. In our review, structural comparison of “classic” IgA1-proteases with their functional analogues and structural homologues allows to hypothesize IgA-protease dependent effector mechanisms mediated by serum IgA.
Data on structure and specificity of bacterial IgA receptors (IgA-binding M-like proteins Arp4 and Sir22 from hemolytic streptococci of serogroup A, β-antigen from hemolytic streptococci of serogroup B, and SSL family proteins from Staphylococcus aureus) are surveyed in this review. The principal conclusion derived from comparison is the fact that all bacterial receptors bind the same site in the IgA molecule overlapping with the binding site of endogenous human IgA receptor CD89. We assume that this site, consisting of spatially close amino acid strands Leu257-Gly259 in domain Cα2 and Pro440-Phe443 in domain Cα3, is subject to conformational rearrangement induced by the binding of antigen in the IgA active site.
Traditionally, the function of immunoglobulins A (IgA), the major type of secreted antibodies, has been thought to be restricted to binding antigens outside the epithelium basal membrane. Therefore, effector mechanisms eliminating IgA-opsonized targets have not been investigated so far. However, some indirect observations of infectious agents penetrating into tissues and blood from the environment suggest such mechanisms (analogous to IgG/IgM-dependent activation of complement and natural killers). In the present review, we examine details of IgA structure that might contribute to elucidation of IgA-dependent effector functions in human and animal immunity. Special attention is given to a putative transduction of signal about antigen binding in the active center of IgA from the Fab-to the Fc-superdomain via intramolecular conformational rearrangements. Different structure of the IgA subclasses (IgA1 and IgA2) is examined taking into account probable divergence of their functions in immune response.
IgA1-specific proteinases (Igase) are known as a pivotal pathogenicity factor in meningococcus (Neisseria meningitides) and in some related bacteria. These enzymes belong to the trypsin-like clan of serine proteases. They exhibit high substrate selectivity, being able to discriminate between IgA1 and IgA2 and to distinguish the human IgA1 from IgA1 of nonprimate mammalian species [8]. According to recent data, in addition to the conventional IgA1-processing enzymes, meningococci contain alternative enzymes. However, the substrate specificity of the alternative Igase, its role in pathogenesis, and the ability to complement the functionality of the conventional Igase remains obscure. In this study, we investigated the structure of the Igase genes and their products in two highly virulent strains M9 and A208 of N. meningitides serogroup A. In particular, we found both conventional and alternative Igase genes in each genome; the nucleotide sequences of these genes were deposited in the NCBI Gene Bank under the accession numbers AY770504, AY558158, and AY558159. The DNA sequence of the conventional Igase was almost entirely conserved in both strains, whereas the recently discovered alternative Igase (formerly known as type 1 meningococcal adhesin) contained a hypervariable region approximately 900 bp long in the 5′-terminal part of the gene. The conventional genes from both strains were expressed in E. coli in the form of inclusion bodies. The recombinant products were used for immunization of rabbits. The antibodies obtained efficiently reacted with both recombinant and native antigens from the N. meningitides culture medium.
Традиционно функцию иммуноглобулинов A (IgA) - основного типа секретируемых антител - сводят к свя-зыванию антигенов, расположенных на внешней стороне базальной мембраны эпителия. По этой причине до настоящего времени исследователями предпринимались лишь отдельные попытки экспериментального изучения эффекторных механизмов целенаправленного уничтожения мишеней, опсонизированных IgA. Между тем результаты косвенных наблюдений за поведением инфекционных агентов, проникающих в тка-ни и кровь из внешней среды, позволяют предположить существование таких механизмов (по аналогии с IgG- или IgM-зависимой активацией комплемента и натуральных киллеров). В рамках настоящего обзора мы анализируем те детали строения IgA, которые могли бы оказаться важными с точки зрения вскрытия IgA-зависимых эффекторных функций иммунной системы человека и животных. Особое внимание уделе- но исследованию возможности передачи сигнала о связывании антигена в активном центре IgA с Fab- на Fc-супердомен за счет конформационных перестроек. С точки зрения расширения функциональных воз-можностей иммунитета по защите организма рассматриваются и различия в молекулярной организации подклассов IgA - IgAl и IgA2.
