Липосомы - полые частицы, содержимое которых ограничено липидной мембраной. Благодаря своим коллоидным свойствам, контролируемым размерам, поверхностным характеристикам, мембранотропности и биосовместимости липосомы являются перспективными системами доставки лекарственных веществ в органы и ткани. Препараты на основе липосом нашли широкое применение в диагностике и химиотерапии опухолевых заболеваний, в вакцинологии, офтальмологии, пульмонологии и при лечении других патологических состояний. В обзоре рассматриваются методы инкапсулирования в липосомы биологически активных соединений, обладающих различными физико-химическими свойствами, что имеет большое значение для фармацевтического производства липосомальных препаратов.
Liposomes are hollow particles, the internal space of which is bounded by a lipid membrane. Liposomes are promising drug delivery systems for organs and tissues because of their colloidal properties, controlled size, surface characteristics, membrane action, and biocompatibility. Liposomal drugs have found wide use in the diagnosis and chemotherapy of cancer, vaccinology, ophthalmology, pulmonology, and the treatment of other pathologies. This review describes the methods of encapsulation of biologically active compounds with various physicochemical properties in liposomes, which is very important for the production of liposomal drugs.
Выбран оптимальный состав и отработана технология получения липосомальной лекарственной формы лизомустина. С целью повышения стабильности лекарственной формы проведена лиофилизация липосомальной дисперсии с использованием криопротектора - 10 % раствора сахарозы. Для количественного определения содержания лизомустина в липосомах предложен метод спектрофотометрии при длине волны 230 нм. Включение лизомустина в свежеприготовленные липосомы составило 61,2 ± 0,4 %, размер частиц - 170 ± 20 нм.
The optimal composition and production technique were identified for preparation of liposomal formulations of lisomustine. The stability of this formulation was increased by lyophilization of the liposomal dispersion using a cryoprotector, i.e., 10% sucrose solution. The lisomustine content in lysosomes was assayed by spectrophotometry at a wavelength of 230 nm. Uptake of lisomustine into freshly prepared liposomes amounted to 61.2 ± 0.4% and particle size was 170 ± 20 nm.
