A newly developed filter-system (PL 100) for the removal of leukocytes from platelet concentrates was tested. It removes contaminating leukocytes by 99.7 +/- 0.6%. The platelet loss was calculated as 25.7 +/- 9.5%. The filtration process does not influence the filtered platelets; plasma-electrolytes, LDH-concentrations, platelet aggregation curves and serotonin-release remain constant. In addition, platelet ultrastructure is not influenced by the filtration. In spite of the high platelet loss, the tested filter-system seems to be a suitable alternative to the conventional methods for the removal of contaminating leukocytes from platelet concentrates.

Die Lagerung von Thrombozytenkonzentraten über einen längeren Zeitraum stellt seit vielen Jahren ein methodisches Problem dar. Meist werden hierfür DEHP-freie Beutelsysteme verwendet, die aufgrund ihrer speziellen Oberflächenbeschaffenheit eine Lagerung bis zu 5 Tagen ermöglichen. Die vorliegende Arbeit untersucht ultrastrukturelle Veränderungen derartig gelagerter Thrombozyten und vergleicht die Ergebnisse mit denen funktioneller Aggregationsteste. Transmissionsmikroskopische Aufnahmen zeigen mit zunehmender Lagerungsdauer eine deutliche Degranulierung der Zellen sowie eine progrediente Auflösung der Zellmembranen und Organellen. Parallel dazu nimmt die aggregatorische Funktíon der Thrombozyten in vitro ab. Am 5. Tag der Lagerung ist nur noch eine sehr geringe Aggregationsfähigkeit nachweisbar. Diese Ergebnisse legen den Schluβ nahe, daβ auch mit den zur Zeit verwendeten gaspermeablen Beutelsystemen das Problem der Thrombozytenlagerung noch nicht vollständíg gelöst ist.

Transmission microscopy reveals a loss of organelles as well as a progredient destruction of cell membranes during storage. At the same time in vitro aggregability is clearly diminished. Therefore, further investigations seem to be necessary to improve storage conditions of platelets.
In der vorliegenden Arbeit sind die Ergebnisse einer in-vitro-Testung des PL 100-Systems zur Entfernung kontaminierender Leukozyten aus Thrombozytenkonzentraten dargestellt. Die Leukozytenreduktion beträgt 99,7 ± 0,6% 25,7 ± 9,5 % der Thrombozyten werden im Filtersystem zurückgehalten. Die filtrierten Thrombozyten werden durch die Filtration nicht beeinfluβt; sowohl die plasmatischen Elektrolyt- und LDH-Konzentrationen als auch die Thrombozytenaggregationsfähigkeit und die Serotonin-Freisetzungsraten ändern sich nicht; auch die Ultrastruktur der Thrombozyten bleibt nach Filtration unverändert erhalten. Somit scheint trotz des hohen Thrombozytenverlustes das getestete Filtersystem eine geeignete Alternative zu den bisher verwendeten Methoden für die Herstellung Leukozyten-depletierter Thrombozytenkonzentrate darzustellen.
