Covering: up to November 2021Since its isolation in 1818, strychnine has attracted the attention of a plethora of chemists and pharmacologists who have established its structure, developed total syntheses, and examined its complex pharmacology. While numerous reviews on structure elucidation and total synthesis of strychnine are available, reports on structure–activity relationships (SARs) of this fascinating alkaloid are rare. In this review, we present and discuss structures, synthetic approaches, metabolic transformations, and the diverse pharmacological actions of strychnine and its mono- and dimeric analogues. Particular attention is given to its SARs at glycine receptors (GlyRs) in light of recently published high-resolution structures of strychnine–GlyR complexes. Other pharmacological actions of strychnine and its derivatives, such as their antagonistic properties at nicotinic acetylcholine receptors (nAChRs), allosteric modulation of muscarinic acetylcholine receptors as well as anti-cancer and anti-plasmodial effects are also critically reviewed, and possible future developments in the field are discussed.
A series of 2‐amino‐benzimidazoles with various substituents in position 1 and at the amino group has been prepared. The methods used are the displacement of chlorine by an amino group in the position 2 of the benzimidazole ring and the condensation of o‐phenylenediamines with carbodiimides or S‐methyl‐isothioureas.

The products of the condensation of o-phenylenediamine with ethyl acetoacetate were re-investigated. Depending on the reaction conditions, 2-methylbenzimidazole (II), 2-isopropenyl-benzimidazole-2-one (VI) and 4,7-dihydro-5-methyl-1H-2,3-benzo-1,4-diazepin-7-one (IV) were obtained. 2-Acetonylbenzimidazole, which had erroneously been described in the literature, could not be isolated from the above mentioned reaction; it was prepared by condensing o-phenylenediamine with the ketal of ethyl acetoacetate followed by hydrolysis of the ketal group. The investigation was extended to the reaction of o-phenylenediamine with 2-carbethoxycyclohexanone and 2-carbethoxycyclopentanone as well as with N-substituted 3-carbethoxy-4-piperidones.
AbstractThe synthesis of a series of 1‐aminoalkyl‐2‐benzyl‐nitro‐benzimidazoles is described. Many of the 5‐nitro‐benzimidazoles prepared show strong analgesic activity. 1‐(β‐Diethylamino‐ethyl)‐2‐(p‐ethoxy‐benzyl)‐5‐nitro‐benzimidazole (XXXVIII) is 1000 times as potent as morphine and therefore the most active analgesic known until now.
AbstractThe reaction of o‐phenylenediamine with α‐phenyl‐ and γ‐phenyl‐acetoacetic ester has been investigated. The former yields mainly 1‐β‐methylstyryl‐benzimidazole‐2‐one (IX), the latter affords 4,7‐dihydro‐5‐benzyl‐1H‐2,3‐benzo‐1,4‐diazepin‐7‐one (XVI). The structures of these products have been elucidated.
AbstractThe synthesis of 1 H‐4‐phenyl‐pyrido‐[1, 2‐c]‐pyrimido‐1,3(2 H)‐ dione (VI) by condensation of phenyl‐pyridyl‐(2)‐acetamide (I) with diethyl carbonate and sodium ethoxide, as well as the preparation of some of its derivatives, is described.

AbstractDie Isolierung der wichtigsten Glykoside der Samen von Acokanthera friesiorum Markgr. aus Kenya wird beschrieben. Die Samen enthielten als Hauptkomponente ca. 1,7 % des bekannten Acovenosid A. Daneben liessen sich aber auch etwa 0,l% Ouabain isolieren sowie kleine Mengen von Acolongiflorosid E, Acolongiflorosid G, Acolongiflorosid H, Acolongiflorosid‐K‐Acetat und ein Kristallisat, das als „Kristallisat Nr. 4”︁ bezeichnet wird und das ein Gemisch dar‐stellt. Durch Verteilungschromatographie liess es sich in 3 Komponenten zerlegen, von denen eine rnit Acolongiflorosid E identisch war, während die zwei anderen neue Glykoside darstellten, die als Acofriosid L und Acofriosid M bezeichnet werden. – Fur einen Abbau war nicht genügend von diesen zwei Stoffen in reiner Form vorhanden, hingegen wurde die Spaltung von „Kristallisat Nr. 4”︁ mit HClin Aceton durchgeführt. Das in diesem Gemisch erhaltene Acolongiflorosid E blieb dabei weitgehend unverändert. Als Spaltprodukte wurden ein neuer krist. Zucker erhalten, den wir Acofriose (V) nennen, sowie 3,5‐Dianhydro‐periplogenin (VI), das sich zum Vergleich auch teilsynthetisch aus Periplogenin bereiten liess. Wir vermuten, dass diese zwei Spaltstücke aus Acofriosid L stammen, so dass sich fur dieses Glykosid eine Teilformel aufstellen liess.
AbstractExtraktion des frischen Holzes von Periploca nigrescens Afzel mit Wasser und Alkohol lieferte nach üblicher Reinigung einen Extrakt, der reichliche Mengen digitaloider Glykoside, freier Aglykone und wahrscheinlich Aglykonacetate enthielt. Von den zuckerfreien Bestandteilen konnten neben reichlichen Mengen (ca. 0,14%) Strophanthidin etwas Strophanthidol sowie kleine Mengen von zwei weiteren Stoffen isoliert werden, die wir vorläufig als Substanz E. Sche. 12 und Substanz E. Sche. 16 bezeichnen. Diese stellen wahrscheinlich Aglykonacetate dar und besitzen möglicherweise die Formeln C25H34O8 und C25H34O7.
AbstractEine gute Ausführungsform für die Papierchromatographie stark polarer Glykoside wird beschrieben. Das mit Wasser getränkte Papier wurde als ruhende Phase verwendet. Als bewegliche Phase dienten n‐Butanol oder n‐Butanol‐Toluol‐Gemisch.
AbstractNach Fermentierung der Samen von Evonymus europaea L. lässt sich anstelle des Triglykosids Evonosid direkt das entsprechende Monoglykosid Evomonosid isolieren. Dieses besitzt wahrscheinlich die Summenformel C29H44O8 und nicht wie früher vermutet C29H44O9.
Honghelosid G, ein Nebenglykosid aus den Wurzeln von A denium Honghel aus Nigeria, wurde mit der tiefschmelzenden Modifikation von Somalin identifiziert. Dieser Stoff konnte in geringer Menge auch in den Stengeln von Adenium Honghel aus Nigeria gefunden werden.
AbstractDie Isolierung von Odorosid H, „Odorosid‐E‐acetat”︁, Odorosid‐K‐acetat, „Kristallisat J”︁, Scopoletin und Scopolin‐tetracetat aus der Rinde von Nerium odorum wird beschrieben.
