ZusammenfassungIm Rahmen von Untersuchungen zum Rückstandsverhalten von Östradiol‐17β nach unterschiedlicher Applikation beim männlichen Kalb wurden während der Mastperiode (bis zur Schlachtung) physiologische Östrogenbasiskonzentrationen im Plasma und Urin ermittelt. Im Plasma ergaben sich für Gesamtöstrogene 0,084 ± 0,037 ng/ml, für freies Östradiol‐17β 0,011 ± 0,012 ng/ml, für konjugiertes Östradiol‐17α 0,038 ± 0,029 ng/ml; im Urin für Gesamtöstrogene 1,315 ± 1,122 ng/ml.Konzentrationsverschiebungen im Anschluß an die Behandlung wurden vor allem durch die Messung der Gesamtöstrogene sowie von freiem Östradiol‐17β und konjugiertem Östradiol‐17α deutlich (Leitverbindungen). Unabhängig von der Applikationsart (Infusion oder Implantation / u. U. Kombination mit Trenbolonacetat) bewirkt die Verabreichung von Östradiol‐17β einen Anstieg der Östrogenkonzentration im Plasma und im Urin, die aber in keinem Fall Werte erreichen, wie sie unter bestimmten physiologischen Verhältnissen (Östrus, Gravidität) bei Schlachtrindern auftreten können.Aufgrund der zu beobachtenden hohen individuellen Schwankungen der Östrogenwerte bei den behandelten und den unbehandelten Tieren kann i. d. R. im Einzelfall nur über einen sehr kurzen Zeitraum zwischen behandelten und unbehandelten Tieren differenziert werden. Spätestens 30 Tage nach der Implantation liegen die Östrogenwerte sicher in dem für die Kontrolltiere (männliche Kälber) ermittelten physiologischen Bereich. Ein Absetzen der Östrogenzufuhr bewirkt in allen Fällen im Plasma und im Urin binnen kürzester Zeit (24 h) einen Rückgang der Östrogenwerte auf Basisniveau. TBOH konnte entsprechend früherer Beobachtungen stets bis zum Ende der Behandlung im Plasma nachgewiesen werden; die Meßwerte lagen zu diesem Zeitpunkt bei 0,01 bis 0,03 ng/ml Plasma.SummaryStudies on the analytical detection of exogenous administration of 17β‐oestradiol and Trenbolone Acetate in blood plasma and urine in the calfDuring studies on the behaviour of residues of oestradiol‐17β after varying dosage of male calves measurements were made during the fattening period (up to slaughter) of physiological oestrogen base concentrations in plasma and urine. Thirty days at the latest after implantation the oestrogen values differ with certainty from those of the control calves (males). Stopping the oestrogen dosage causes in all cases a fall in oestrogen concentration in plasma and urine to the basal level very rapidly (24 hr). En el plasma se constataron para los estrógenos totales 0,084 ± 0,037 ng/ml, para oestradiol‐17β libre 0,011 ± 0,012 ng/ml, para oestradiol‐17α conjugado 0,038 ± 0,029 ng/ml; en la orina para estrógenos totales 1,315 ± 1,122 ng/ml.Desplazamientos de la concentración a continuación del tratamiento se hicieron evidentes, ante todo, mediante la medición de los estrógenos totales así como de oestradiol‐17β libre y oestradiol‐17α conjugado (nexos de guía). A más tardar, 30 días después de la implantación, se hallan los valores estrógenos, de forma segura, dentro del ámbito fisiológico determinado para los animales testigos (terneros).El cese de aprovisionamiento de estrógeno ocasiona en todos los casos en el plasma y en la orina en un tiempo muy corto (24 h) un retroceso de los valores de estrógenos al nivel de base. ZusammenfassungUnter Verwendung des uterinen zytoplasmatischen Östrogenrezeptors vom Kalb wurde ein Radiorezeptortest zur Messung östrogenwirksamer Verbindungen in Pflanzen eingerichtet. Mit diesem Verfahren wurden 55 Futterproben aus 30 Betrieben mit auffälligen Fruchtbarkeitsstörungen untersucht. Von 18 Maissilageproben konnte keine als östrogenpositiv eingestuft werden, Gras‐ und Heuproben wurden teils negativ teils positiv gefunden, überwiegend positive Ergebnisse mit zum Teil sehr hohen Phytoöstrogenkonzentrationen wiesen Kleeproben oder Proben mit Kleeanteilen auf. Nordafrikanische Trifoliumproben (Trif. subterraneum) wurden über den Zeitraum einer Vegetationsperiode geprüft. Am Anfang derselben wurden die höchsten Phytoöstrogengehalte gemessen, gegen Ende fielen die Werte ab. Grasproben aus dieser Region zeigten mit Ausnahme einer Probe keine östrogene Wirksamkeit. Aus den Ergebnissen war ein Zusammenhang zwischen hohen Phytoöstrogenkonzentrationen und Fruchtbarkeitsstörungen bei Weidetieren in den nordafrikanischen Betrieben abzuleiten. Die Befunde bei den einheimischen Proben lassen es empfehlenswert erscheinen, Phytoöstrogene bei der Suche nach Kausalfaktoren von Fertilitätsstörungen besonders dann in Betracht zu ziehen, wenn Kleefutter eingesetzt wurde. Eine vergleichende Untersuchung von 3 Hopfensorten zeigte, daß nur den Dolden am Ende der Wachstumsperiode eine starke östrogene Wirkung zukam und bestätigte außerdem, daß Hopfen zu den östrogenreichsten Pflanzen zu rechnen ist.SummaryStudies on the demonstration of phyto‐oestrogens in feed plants and hops with the aid of a receptor testUsing the uterine cytoplasmic oestrogen receptors of the calf a radioreceptor test was devised to measure oestrogen‐active compounds in plants. With this test 55 feed samples from 30 premises with marked fertility problems were examined. Of 18 maize silage samples none was oestrogen‐positive. Some grass and hay samples were positive, others negative, but positive results were preponderant, including some very high phyto‐oestrogen concentrations, with clover samples or feed samples containing some clover. Trifolium from North Africa (Trif. subterraneum) was tested over a whole period of vegetative growth. In early stages the highest concentrations of phyto–oestrogen were found, while subsequent values were lower towards the end of growth. Except for one sample, grass samples from this region showed no oestrogen. The results showed a relationship between phyto‐oestrogen concentrations and reproductive disturbances in grazing animals in North African regions. The results with local samples make it advisable to consider phyto‐oestrogens in feed in the search for the causes of impaired fertility whenever clover is fed. A comparative examination of 3 varieties of hops showed that only the umbels at the end of the growth period have a marked oestrogenic effect: the results show that hops must be considered as within the plants richest in oestrogens.RésuméRecherches sur la mise en évidence de phytooestrogènes dans des plantes fourragères et du houblon avec un test de récepteurOn a installé un test de radiorécepteur pour mesurer les liaisons oestrogène‐actives dans des plantes en utilisant des récepteurs d'oestrogène cytoplasmiques utérins du veau. On a examiné de cette manière 55 échantillons de plantes provenant de 30 exploitations présentant des troubles de la fertilité. Aucun échantillon sur 18 de maïs ne fut ostrogène‐positif; les échantillons d'herbe et de foin furent en partie négatifs et positifs; les résultats essentiellement positifs avec parfois de très fortes concentrations en phytooestrogènes ont été trouvés dans des échantillons de trèfle ou contenant du trèfle. Des échantillons de Trifolium (Trif. subterraneum) nord‐africain ont été examinés durant le temps d'une période de végétation. Les taux les plus élevés en phytooestrogènes furent relevés au début et diminuèrent à la fin. Les échantillons d'herbe de cette région à l'exception d'un seul n'ont présenté aucune activité oestrogène. Ces résultats ont permis de faire un rapport entre de fortes concentrations en phytooestrogènes et des troubles de la fertilité chez les animaux au pâturage dans les exploitations nord‐africaines. Les résultats locaux laissent apparaître que la recherche des phytooestrogènes en tant qu'agents étiologiques des troubles de la fertilité entrent en considération lorsque l'alimentation comprend du trèfle. Une recherche comparative sur trois sortes de houblon a montré que seule l'ombelle possède une action fortement oestrogène à la fin de la pétriode de croissance et que le houblon doit être rangé dans les plantes les plus fortement oestrogènes.ResumenEstudios sobre la identificación de fitoestrógenos en plantas forrajeras y lúpulo con ayuda de una prueba a base de receptorUtilizando el receptor citoplasmático uterino de estrógenos en la ternera, se instaló una prueba radiorreceptora para la medición de combinaciones de acción estrógena en los vegetales. Con este procedimiento se examinaron 55 muestras de forraje de 30 explotaciones con trastornos evidentes en la fertilidad. De 18 muestras de maíz ensilado no se pudo encuadrar ninguna como estrógena‐positiva, las muestras de hierba y heno se comportaron en parte negativas, en parte positivas, mientras que los resultados preponderantemente positivos con concentraciones de fitoestrógenos en parte harto elevadas los arrojaban las muestras de trébol o muestras con partes de trébol. Las muestras norteafricanas de Trifolium (Trif. subterraneum) se analizaron durante un espacio de tiempo equivalente al periodo de vegetación. Al principio del mismo se midieron los contenidos más elevados de fitoestrógenos, hacia el final del mismo descendieron los valores. Con la excepción de una muestra, las muestras de hierba procedentes de esta región no mostraban ninguna actividad estrógena. De los resultados obtenidos se pudo derivar una relación entre las concentraciones elevadas de fitoestrógenos y los trastornos en la fertilidad de los animales de pastoreo en las explotaciones norteafricanas. Los hallazgos logrados en las muestras autóctonas recomiendan el que se consideren sobre todo los fitoestrógenos en la búsqueda de los factores causales de los trastornos de la fertilidad cuando se haya empleado pienso a base de trébol. Un estudio comparativo de 3 especies de lúpulo mostró que solo a las umbelas al final del periodo de crecimiento le correspondía una acción estrógena fuerte, confirmando además el que el lúpulo se debe considerar entre los vegetales con mayor riqueza en estrógenos.
In 25, 3-to-13 year old, dairy cows (Braunvieh and Hoehenfleckyieh) FSH, LH and progesterone plasma profiles were determined by RIA. Blood was sampled at 6-hour intervals from parturition to 40–78 days postpartum, and the results correlated with the commencement of cyclic functions. For FSH, generally basic values were recorded, without characteristic features associating any values with the onset of cyclic ovarian activities or the occurrence of the first heat. LH profiles varied greatly between individuals with regard to the onset of elevations, regularity of patterns and peak values. The first preovulatory LH peak was recorded 17.3±9.8 days (range 4–46) postpartum. The first heat occurred on day 28.4±16 (range 6–55) postpartum, indicating that 1323 cows ovulated without behavioral estrus, as reproductive cycles were re-established. Peak LH values increased with progressive cycles (1st peak 5.7±4.8 ng/ml; 2nd peak 11.8±8.7 ng/ml; 3rd peak 13.5±9.9 ng/ml plasma). Progesterone values also showed great variations in the profile of their first postpartum elevation. In 1325 cows the first cycle was shortened (13.1±2.9 days), prolonged in 3 animals (34±4 days) and normal in 7 cows (20.4±1.9 days). Heat, preovulatory LH peak and progesterone profile were normal in all animals on subsequent cycles. Two animals did not start cycling.
Bei einer Gruppe von 8 Rehböcken, die in Gehegen gehalten worden waren, wurden Blutproben vom 8. bis zum 28. Lebensmonat entnommen und die Testosteronkonzentration im Blutplasma gemessen. Während des gleichen Untersuchungszeitraumes wurde der Futterverzehr, die Gewichtsentwicklung, die Trophik und das Wechseln des Haarkleides kontrolliert sowie visuelle (ethologische) Parameter (Hodensichtbarkeit, Aggressivität, Entwicklungszustand der Metatarsaldrüsen) beobachtet.

For determination of early pregnancy in cows milk samples were drawn under field conditions on the 20th day after insemination from 168 animals and mailed to the laboratory for progesterone analysis. Values above 11 ng/ml were considered indicative of pregnancy, and those below 2 ng were classified non-pregnant. Values ranging between 2 and 11 ng/ml were graded according to the progesterone and fat concentration in milk samples. The method was based on results obtained in earlier studies, which showed a highly positive correlation between changes in progesterone concentrations and changes of milk fat concentrations when an active corpus luteum is present. A diagnosis of pregnant was in agreement with results derived from a clinical pregnancy diagnosis by palpation performed 8 to 9 weeks after insemination in 86% of cases; in all cases of a negative diagnosis the cows were found nonpregnant.
Summary. Levels of lh were determined by radioimmunoassay and those of progesterone by a competitive protein-binding technique. Blood samples from eight cows, representing two complete pregnancies (including post-partum periods), and from one cow in the first trimester and another around the time of parturition, were collected. The samples were withdrawn at intervals of 6 hr for lh and every 4 to 5 days for progesterone determination. Levels of lh were consistently low (1·0 to 1·6 ng/ml plasma) with a few single peaks occurring irregularly in individuals during the first 110 days. Distinct lh peaks were observed between Days 7 and 20 post partum. The decrease of progesterone from high levels during pregnancy (plateau between 5 and 9 ng/ml plasma with individual differences) before parturition and the low level following was not reflected in the lh concentration. In an experiment with one animal, an injection of 5 mg flumethasone near term caused a partial decrease in the progesterone concentration, and a larger dose of 10 mg induced parturition. This was preceded by a marked decrease of progesterone similar to that observed before a normal delivery.
The characteristic sex odor, observed in carcasses of boars (boar taint), which is seldom found in female and male-castrate animals, has been the subject of many investigations, most of them using organoleptic approaches. The subjectivity of these methods called for chemical identification of the tainting substances and their demonstration in blood, tissues and excreta. In 1968, Patterson identified 5a-androst-16-en-3-one in fatty tissue of boars as one of these substances which contributes heavily to the sex odor complex. Echternkamp et al. (1969) and Wismer-Pedersen (1968) reported a correlation between the intensity of the odor (detected by organoleptic examination) and the activity of androgens measured by a chick comb assay. However, Gower and Ahmad (1967) could demonstrate that 5a-androst-16-en-3-one is produced by its own biosynthetic pathway different from that of the main androgen testosterone. When measuring testosterone in peripheral plasma of pigs, Hoffmann, Claus and Karg (1970) found concentrations increasing with age, specifically in boars. This presentation deals with the determination of 5a-androst-16-en-3-one in peripheral blood plasma, testicular tissue, fatty tissue and the parotid salivary gland of boars, barrows and gilts. Simultaneously, testosterone was determined in the testicular tissue of the same boars. Emphasis was placed on possible relationships between levels of the main androgen testosterone and 5a-androst-16-en-3-one; and also, on whether the latter compound is stored in fatty tissue and the parotid salivary gland, since organoleptic evaluations are based mainly on examination of these tissues.
