Protein synthesis in the cell, involves particulate elements called ribosomes which translate the information coded as specific nucleotide sequences in messenger RNA molecules into amino-acid sequences of proteins [1]. Ribosomes are frequently found in the cell in the form of aggregates called polysomes. The ribosomes in a polysome are believed to be attached to a single messenger RNA molecule and simultaneously to translate the encoded information to give polypeptides with a specific amino-acid sequence [2]. The basic mechanisms of the control of protein synthesis at the level of translation are not well understood.
The binding curves of oestradiol and testosterone to plasma membranes from male or female rat livers show no specific sites of high affinity. Plasma membranes from male and female rat livers show no disulphide interchange activity. It is concluded that no steroid‐promoted association of ribosome with plasma membranes occurs. Microsomes prepared from a homogenate from which plasma membranes have already been isolated, show a slightly higher concentration of specific high‐affinity steroid‐binding sites than do standard preparations of microsomes. It is concluded that standard microsomal preparations are contaminated only to a slight extent by fragments derived from the plasma membrane.
Smooth reticular membranes from male rat liver contain high affinity binding sites for oestradiol. Similar membranes from females contain analogous sites for testosterone. Rough membranes from which polysomes have been removed by EDTA (degranulated membranes), contain no high affinity binding sites for either hormone. Ethyl acetate extracts of degranulated membranes contain materials which block the high affinity binding sites on smooth membranes of the same but not the opposite sex. These extracts promote the attachment of ribosomes to smooth membranes of the same but not the opposite sex. The ethyl acetate extracts from male and female degranulated membranes contain substances which behave like oestradiol and testosterone, respectively.
