目的 通过研究基质金属蛋白酶(MMP-9)在慢性心衰中的作用,从分子生物学角度探讨中药不同配伍对慢性心衰心室重构的影响,并进一步阐明其作用机理.方法 以冠脉结扎法配合力竭式游泳、减食等方法造成大鼠慢性心衰动物模型,分成模型组、西药组、益气中药组、活血中药组、益气活血中药组、益气活血利水中药组和正常组.利用实时荧光定量PCR技术及SABC免疫组化法检测慢性心衰大鼠MMP-9的变化情况.结果 慢性心衰动物模型组大鼠心肌MMP-9表达明显升高,与假手术组比较,差异有统计学意义(P<0.01);经过治疗,各用药组大鼠心肌组织MMP-9表达明显降低,其中益气活血组、益气活血利水组中药与西药组比较,差异无统计学意义(P>0.05),且益气活血利水组疗效明显优于益气活血组(P<0.01).结论 证明益气活血利水中药可通过抑制心肌组织中MMP-9的表达,抑制或逆转心室重构的过程,从而达到治疗慢性心衰的目的.
Objective:By investigating the effects of different Chinese medicine compounds on expressions of matrix metallopeptidase 2(MMP-2)and tissue inhibitor of metalloproteinase-2(TIMP-2)in chronic heart failure(CHF),we studied the effect of traditional Chinese medicine on further mechanism of ventricular remodeling in CHF on the role of molecular biological sense.Methods:The coronary artery ligation method with exhaustion swimming and reduce feeding was used to cause chronic heart failure rats model.Rats were divided into model group,west medicine control group,Qi-supplementing group,blood-activating group,Qi-supplementing and blood-activating group,Yiqi huoxue Lishui treatment group and normal group.The real-time quantitative PCR technique and SABC immunohistochemical method were used to detect MMP-2 and TIMP-2 changes of CHF rats.Results:MMP-2 expression in animal model of chronic heart failure group increased obviously,and TIMP-2 expression decreased,compared with the surgical group,and differences were statistically significant(P0.01).After treatment,MMP-2 expression in the medication group significantly reduced obviously,and TIMP-2 expression increased.The difference in the supplementing Qi and activating blood group,Yiqi huoxue Lishui treatment group and Western medicine group were no statistically significant(P0.05).And therapeutic effect of Yiqi huoxue Lishui group was much better than that in the supplementing Qi and activating blood group(P0.01).Conclusion:It is proved Chinese compound of supplementing Qi,activating blood circulation and promoting urination can inhibit or reverse ventricular remodeling processon by acting on MMP-2 and TIMP-2 in organizations,so as to achieve the goal of therapy for chronic heart failure.

慢性心衰是临床常见病、多发病,严重危害人类健康。该病中医多归属于"心悸,喘证,胸痹,水肿"等病名范畴内。根据慢性心衰相关临床表现,多数学者认为该病属本虚标实证,以心气阳虚为主,血瘀水停痰湿内阻为标。我们通过文献和临床观察研究,认为慢性心衰以心气虚为主,心血瘀滞,水湿内停为主要病理因素。治疗以益气温阳,活血通脉为主要治法。
慢性心衰为临床危重症,是人类健康的大敌.笔者通过文献及长期临床观察认为心气虚为该病发病的根本,心血淤阻为主要病因,阳气虚衰为病机关键,而心气虚,耗损阳气,鼓动无力,则血行滞缓,血脉淤阻,从而出现心衰.因此,气虚血淤贯穿慢性心衰病程的各个阶段,临床中注重灵活辨证,而不拘泥一方一证,对表现错综复杂、证型相互参杂者,应灵活辨证用药,可获明显疗效.
A novel vapor-permeable composite membrane for air dehumidification has been fabricated. The membrane used porous polyethersulfone (PES) as the support layer, and dense polyvinylalcohol (PVA) as the active separating layer. To facilitate moisture permeation, lithium chloride (LiCl) salts were used as an additive to the PVA solution before it was cast onto the support layer. The effects of the LiCl concentration on the water vapor permeability are investigated. The permeation tests indicate that the addition of LiCl in PVA cast solution can improve vapor permeability substantially. The observations of the microstructures and the characterization of the synthesized material disclosed that the reason behind increased moisture rates is that the addition of LiCl increased the hydrophilicity.
A novel composite supported liquid membrane,which employs LiCl liquid solution immobilized in a porous support membrane to facilitate the transport of moisture,is prepared.To protect the SLM(Supported Liquid Membrane),two hydrophobic PVDF(Polyvinylidene Fluoride) layers are formed on both surfaces of the SLM.The membrane formation and characteristics are investigated.The results show that,the supported liquid layer and the two protective layers are in good contact and the layers are much thinner,from SEM observations;the vapor permeation performance is very good,while the CO2 permeation rate is almost near to zero;the total vapor transfer coefficient,the vapor permeation velocity,the moisture transfer efficiency and the aperture on the surfaces of the membranes increase with the mass fraction of the solid decreasing,when the mass fraction of the additive(PEG2000) was kept to be 2.4%.
The factors and mechanisms leading to the instability of supported liquid membraneswere analyzed.The efforts worldwide devoted to the improvement of the stability of supportedliquid membranes were introduced.The development of new module structures,the compositions ofthe liquid phase,and the fabrication techniques for the supported liquid membranes were discussed.In the end,the prospects for the commercialization of the supported liquid membranes werepresented.
