Coronary artery disease (CAD) is a major atherosclerotic cardiovascular disease and the leading cause of mortality globally. Long non-coding RNAs (lncRNAs) play crucial roles in CAD development. To date, the effect of lncRNA non-coding RNA activated by DNA damage (NORAD) on atherosclerosis in CAD remains unclear. The primary aim of this study was to investigate the effect of lncRNA NORAD on vascular endothelial cell injury and atherosclerosis. Here, ox-LDL-treated human umbilical vein endothelial cells (HUVECs) and high-fat-diet (HFD)-fed ApoE –/– mice were utilized as in vitro and in vivo models. The present study found that lncRNA NORAD expression was increased in ox-LDL-treated HUVECs and thoracic aorta of atherosclerotic mice, and knockdown of lncRNA NORAD alleviated vascular endothelial cell injury and atherosclerosis development in vitro and in vivo . Knockdown of lncRNA NORAD aggravated ox-LDL-reduced or atherosclerosis-decreased vascular endothelial growth factor (VEGF) expression in HUVECs and thoracic aorta of mice to ameliorate vascular endothelial cell injury and atherosclerosis development. Moreover, nucleus lncRNA NORAD suppressed VEGF gene transcription through enhancing H3K9 deacetylation via recruiting HDAC6 to the VEGF gene promoter in ox-LDL-treated HUVECs. In addition, VEGF reduced FUS (FUS RNA binding protein) expression by a negative feedback regulation in HUVECs. In summary, lncRNA NORAD enhanced vascular endothelial cell injury and atherosclerosis through suppressing VEGF gene transcription via enhancing H3K9 deacetylation by recruiting HDAC6. The findings could facilitate discovering novel diagnostic markers and therapeutic targets for CAD.
目的:探讨冠状动脉CT血管成像(CCTA)定量参数评估薄纤维帽粥样硬化斑块(TCFA)易损性的可行性.方法:前瞻性将2018年9月-2020年9月在本院经DSA确诊为冠心病(血管狭窄率>50%)的连续124例患者纳入研究,DSA术中采用血管内超声(IVUS)识别并确定161个TCFA和138个非TCFA.术前所有患者行CCTA检查,测量和计算靶血管的狭窄率、最小管腔面积(MLA)、斑块处血管外径相对值、斑块体积和体积百分比,记录低密度斑块(LAP)、餐巾指环征(NRS)、正性重构(PR)和点状钙化的数量.结果:CCTA测量的冠脉狭窄率与DSA测量结果间的差异无统计学意义(P>0.05).TCFA组的斑块体积及百分比、MLA和RED均明显大于非TCFA组,LAP出现率和CT值小于非TCFA组,但LAP体积及百分比大于非TCFA组,NRS和点状钙化的出现率多于非TCFA组(P<0.05).多因素Logistic回归分析结果显示,LAP的体积和体积百分比以及PR是TCFA的主要危险因素(P<0.05).受试者工作曲线(ROC)分析结果显示,LAP的体积和体积百分比、PR以及LAP体积+LAP体积百分比+PR预测TCFA的符合率分别为0.856、0.902、0.769和0.960.结论:CCTA是无创性评估TCFA易损性质的主要检查工具,斑块相关定量参数中LAP的体积和体积百分比以及PR预测TCFA有较高的准确性,值得临床推广应用.
Objective To explore the value of low-dose multi-slice spiral computed tomography (MSCT) multi-stage scan and three-dimensional reconstruction in diagnosis of patients with gastric carcinoma.Methods A total of 40 patients with gastric carcinoma were selected.All the patients were conducted with low-dose MSCT scan,dynamic contrast scanning and three-dimensional reconstruction scan,and then were processed by multi-planar reconstruction,volume reconstruction and CT virtual endoscopy.Primary lesions in gastric cancer,peripheral infiltration and distant metastasis were observed.Results All the patients were satisfied with the low dose MSCT images,and all of them found primary gastric lesions and had satisfactory diagnosis rates of gastric peripheral infiltration,lymph node metastasis and distant metastasis.Conclusion Low-dose MSCT multi-stage scan and three-dimensional reconstruction can reduce the radiation dose and reveal the primary gastric lesions,gastric peripheral infiltration and distant metastasis.
BACKGROUND AND AIM:To evaluate image quality and radiation dosage in coronary CT angiography using 80-kVp tube voltage combined with low-concentration contrast media (CM) and iterative reconstruction (IR) for coronary CT angiography (CCTA) and employing dual-source CT without heart-rate control.METHODS:154 patients were randomly assigned to Group A (Control Group, 120-kVp tube voltage, high-concentration CM and filtered back projection reconstruction) and Group B (Low-Dose Group, 80 kVp, low-concentration CM and iterative construction). Two experienced radiologists double-blindly evaluated the following parameters: CT attenuation, signal-noise ratio (SNR), contrast-noise ratio (CNR), radiation dose, size-specific dose estimates (SSDE) and total iodine intake. Pearson correlation analysis was used to assess the relationship between SSDE and BMI.RESULTS:98.1% vessel segments in Group A and 97.6% in Group B passed diagnostics, indicating no significant differences; the average aorta scores and CT attenuation values showed no significant differences between groups. Similar SNR and CNR results were obtained for the two groups, although values were slightly lower in Group A compared with Group B. The Effective Dose in Group B was 63% lower than that in Group A (P<.001). SSDE results were significantly different between the two groups (P<.001) but did not correlate with BMI. Finally, the total iodine intake in Group B was 22.9% lower than that in Group A.CONCLUSIONS:Coronary CTA conducted with a low tube voltage of 80 kVp, a low-concentration CM and IR without heart rate control can achieve images of similar quality to those obtained using standard procedures, significantly reducing the associated radiation dose and iodine intake.
Objective To study the diagnostic value of multislice spiral computed tomography (MSCT) including multi-stage scan and post-processing techniques in gastric carcinoma .Methods 25 cases of gastric carcinoma ,which were path-ologically proved ,underwent plain scanning ,triphase enhancement scanning and three-dimensional reconstruction scan-ning ,then ,many kinds of post-processing technology including multi-planar reconstuction ,volume rendering and CT vir-tual endoscopy were performed to observe primary lesions in stomach ,infiltration and distant metastasis .Results All pa-rameters were obtained and the image was satisfactory .It was clearly shown that the primary lesions were in stomach and the infiltration was around them ,same to the lymph node metastasis .Conclusion It could be revealed primary lesions in stomach and infiltration and distant metastasis by MSCT multistage scan and post-processing techniques .It is significant for preoperative evaluation and the choice of therapeutics way of gastric carcinoma .MSCT can increase the detection rate of small focus of infection ,with high accuracy in judgement of invasive depth and regional lymph node metastasis of gastric carcinoma ,and the superiority in evaluating the distant metastases and the involvement of surrounding organs .MSCT multistage scan and post-processing techniques can achieve many-sided information ,which can replace preoperative evalua-tion of gastric carcinoma by X-ray radiography of upper digestive tract .
