With 2019 seeing the launch of the journal, it was an exciting year for the Future Drug Discovery team and, in this Foreword, we discuss our highlights and look forward to what is coming in 2020.Content highlights from 2019Future Drug Discovery publishes quarterly issues and many of our issues carry a themed section surrounding a topic of high interest to the drug discovery community. In 2019, we covered the exciting topic of artificial intelligence (AI), which saw us speak with a number of experts to discuss AI concepts, such as the computational chemist, and discuss some of the challenges the AI field faces, including ethics and intellectual property [1]. Separate Editorials also saw R Lawrence (Cancer Research UK Manchester Institute, UK), E Begoli (Oak Ridge National Laboratory, TN, USA) and D Kusnezov (US Department of Energy, DC, USA) discuss their takes on the topic [2,3]. Begoli and Kusnezov summed the topic up nicely in their conclusion: "Clearly the process of drug discovery requires improvements, but the opportunities to do so with AI are manifest." Various commentaries, research, review and opinion pieces were also published. One of our content highlights is a commentary from Zhang et al., which looks at the potential held by metal polyphenol nanonetworks for drug delivery [4]. This article has been read over 900 times and currently holds our most-cited spot. Our most-read article thus far is a research article by Kanna et al., which saw the team investigate whether their perlecan-targeted nanoparticles could improve drug delivery in triple-negative breast cancer [5].We also published a series of interviews with people from various aspects of the drug discovery field discussing life in their areas, including experts from an independent consultancy, contract research organizations and a start-up in a science incubator. We look forward to continuing this series, as it provides an interesting look into the drug discovery careers landscape.Special issues in 2020The year of 2020 will see us publish a plethora of both general and thematic content. This issue is focused around immuno-oncology and we have upcoming issues on drug-repurposing, anti-microbial resistance and computer-aided drug design. If you would like to discuss ideas for any of these issues, please contact us via email.Maximizing discovery across the globeFuture Drug Discovery utilizes a number of services to help support its researchers and maximize the visibility and impact of work. These include Publons (rewarding peer review), Altmetrics (tracking online discussion of articles, such as via social media or news outlets), Dimensions (tracking citations) and ScienceOpen (disseminating research widely and putting it into context). Download, Dimensions and Altmetrics statistics will be shown on article pages, meaning authors can track the success of their articles. In this interview, he gave further insight into his paper on a novel computational methodology to evaluate chemical optimization [7].Working with the RxNet Editorial team, we were delighted to release the first edition of our Glossary of Drug Discovery last August [8]. Thanks to the help of our expert panel, we compiled a list of all the key terms used in drug discovery – from abbreviated new drug application to Z' factor – and provided handy definitions for your reference. As we celebrate our 5th birthday as a journal, we are proud to maintain our commitment to publishing the key advances in clinical and translational research across this niche, yet fast paced, area of research.In this Foreword, we take a look back at some of our content highlights from 2019, as well as introduce our new resource for concussion researchers: Concussion Zone [1]. We also provide an update regarding the work of our parent, The Drake Foundation [2], on behalf of which the journal is published; looking at the projects they are currently funding in order to help further concussion research.Content highlights of 2019The standard of articles published over the past year has remained at the high quality for which we are known and we would like to thank all of our authors and reviewers, each of whom has worked to ensure that all work follows the highest of standards.Our most read article of the year comes from a research team led by Melissa Hunfalvay of RightEye LLC (MD, USA) and describes their novel eye-tracking tests, which measure both horizontal and vertical saccades to create an objective and quantifiable method of differentiating between severities of traumatic brain injury [3]. An altmetric score of 58 puts this article in the top 5% of all research outputs scored by the metrics system and in the 95th percentile of articles of the same age. A follow-up article from this research group was also published in this issue and we recommend you look out for it.A hot topic of the year has been concussion in school- and college-aged adolescents. First, we had a research article written by Arthur Maerlender et al., reporting on a training model to be given to school personnel that will improve knowledge on how to manage concussion in a student [4]. Based on ten key knowledge competencies, the training method gained positive feedback from those involved in the study, with many reporting the competencies to be informative and useful. For anyone who works in concussion management, this is a key read.Following the theme of concussion in young people, we have another research article, this time with a focus on college students. In this article, the research team, led by Zachary W Bevilacqua (Indiana University, ID, USA), present a new approach to monitor concussed students and determine when they are recovered enough to re-enter the academic setting [5]. This concept, commonly referred to as return-to-learn, has been at the forefront of concussion research and as all student-athletes are first and foremost students, a greater understanding is of vital importance.Our final paper to mention comes from Stanford's (CA, USA) Angela Lumba-Brown and colleagues. In their perspective article, they provide an overview of commonly used post-concussion symptom-rating scales and determine the representation of different subtype-directed symptomatologies [6]. They found that overall, the percentage of symptoms representing each subtype was low, though ocular-motor, vestibular and cervical strain-associated conditions were the least represented. This under-representation of key symptoms marks a clear unmet need in the field of concussion research.Editorial boardTo our Editorial board, we thank you for your continued input, be it in an ambassadorial, advisory or authorship role. Further, in the events channel, you can find our Editor's reports from relevant conferences.Registration to Concussion Zone is free and will provide you with exclusive access to editorials, opinion pieces and interviews from experts within the field of traumatic brain injury, plus optional newsletters via email to keep you up to date with the latest research.Some of our top pieces of content since our launch include a news piece following the publication of the highly anticipated FIELD study, which reported that professional football (soccer) players are 3.5-times more likely to die due to dementia and over five-times more likely to die due to Alzheimer's disease [7] and an editorial in which Claire Baker (Imperial College London, UK) discusses how on-board sensors could be utilized to reduce the effect of head trauma and gives insight into the work being conducted in the AutoTRIAGE project [8].If you have any ideas on what you would like to see on Concussion Zone or are interested in writing for the site, then please get in touch.Update from The Drake Foundation2019 was a big year for The Drake Foundation, with the kick-off of several new projects, as well as progress updates from several others.In January came the announcement of a new research study to evaluate concussion in Premier League football (soccer) [9]. The study is being led by Tony Belli and Patrick O'Halloran (both University of Birmingham, UK), with support from the Premier League and funding from the Foundation. Over the 2018/19 and 2019/20 seasons, saliva and urine samples will be collected from players who sustain a head injury, as well as noninjured controls. These samples will then be tested using the Birmingham Concussion Test to examine the potential of microRNAs as biomarkers of brain injury. The hope is that the test could 1 day be used pitch-side, with the potential to assist in return-to-play decisions and concussion diagnoses across sport, the military and other frontline settings.This year has also seen the start of recruitment for the HEADING (Health and Ageing Data IN the Game of football) study [10], which will recruit 300 former elite football (soccer) players in order to examine the link between a history of concussion and heading the ball with neurodegenerative disease and long-term cognitive function.Another exciting new addition to The Drake Foundation's research portfolio was launched in 2019: The Drake Football Study (DFS) [11]. The DFS comes as a result from a groundbreaking collaboration between the Foundation (as founding funders) and FIFPRO, the PFA and football players unions across Europe. Researchers will measure the health of professional footballers; a population that has been shown to be more likely to experience problems with their mental, musculoskeletal, neurocognitive and cardiovascular health. Until now, available evidence has been established principally from cross-sectional studies focusing on a specific health domain in either active or retired players. The DFS is the first prospective cohort study that monitors the health of professional footballers across domains (mental, musculoskeletal, neurocognitive and cardiovascular) before, during and after their retirement out of sport. This will hopefully fill the gaps in our knowledge around the onset and progression of several health conditions, which will help equip clubs and medical teams to support future players both in their careers and throughout retirement.Outside the research funding, in November 2019 was the 4th Annual UK Sports Concussion Research Symposium, this year jointly hosted by The Drake Foundation, Football Association, Rugby Football Union, England and Wales Cricket Board and British Horseracing Authority. This year's symposium marked a significant year in the concussion field, with media attention and public interest in the long-term effects of sports concussions continuing to grow, particularly around the publication of the first results from the FIELD study. It was fantastic to hear progress from speakers in concussion research, sporting bodies and technology development, with talks centering on diagnostic and recognition tools, understanding of recovery and medium and long-term consequences of head injury. However, before we start this new period, I would like to look back over the past year and our highlights from 2019.Content highlightsOur most-read article of the year comes from Ugonma Chukwueke and Patrick Wen, both of the Dana-Farber Cancer Institute (MA, USA), and reviews the recommendations of the Response Assessment in Neuro-Oncology (RANO) working groups [1]. Distinct RANO groups have been set up for different tumor and treatment types, each group aiming to develop relevant and reliable criteria that can be implemented by clinicians. This review discusses how the recommendations given by the RANO groups may be applied to the care of patients with different brain tumor types and suggests what is still needed for the guidelines to fully reflect the complex nature of each tumor. For anyone interested in neuro-oncology and CNS oncology management, this is a key read. From Issue 1 of this volume, the article has been cited twice and is already the most-read article ever published in CNS Oncology.Next up, we have a preliminary communication written by David E Piccioni et al. (University of California San Diego, CA, USA), describing their study evaluating whether circulating tumor DNA could facilitate genomic interrogation in patients with primary brain tumors [2]. Contrary to previous studies in this area, the group found that 50% of patients included in the study had detectable circulating tumor DNA, making it a clinically viable option for analysis when identifying genomics-based therapy options.Another 2019 article that has hit our most-read list is an editorial that evaluates the benefits of whole-brain radiation therapy versus stereotactic radiosurgery for the treatment of brain metastases [3]. Written by a group from the City of Hope National Medical Center (CA, USA), the article discusses the shift in clinical practice from using whole-brain radiation therapy to stereotactic radiosurgery and looks to the future of radiation therapy for brain metastases.Finally, we have a set of three articles each discussing the Nativis Voyager®, an investigational therapeutic medical device that is being developed for the treatment of both pediatric and adult glioblastoma. With feasibility studies in both American [4] and Australian [5] populations, the researchers found no adverse events that could lead to the discontinuation of the device, and it demonstrated a benign safety profile. However, as stated in an editorial on the device by Victor Levin (MD Anderson, TX, USA), further studies are needed to determine the full impact of the therapy on overall survival of patients with glioblastoma [6].Editorial boardTo our editorial board, we thank you for your continued input, be it in an ambassadorial, advisory or authorship role. However, before we start this new period I would like to look back over the past year and our 2018 highlights.2018 was a year of change for the editorial team at Lung Cancer Management with new faces starting and the journal growing. One of the biggest changes is yet to come with our acceptance to PubMed Central; currently in the final stages of the process, we hope to have all content from the past 2 years and onwards available to search through PubMed in the near future. This is a huge step and can only be beneficial in increasing the reach of the articles we publish.Content highlights of 2018The standard of content that we publish has remained high. This is due to both our authors and our reviewers who utilize our peer review and revisions policy to ensure all work is at its best.The most-read article of 2018 also happens to be our most-read article ever published. The research team from Allegheny Health Network Cancer Institute (PA, USA) discusses the use of stereotactic body radiotherapy for early stage lung cancer and outcomes following its use in biopsy-proven and empirically treated lesions [1]. Watch this space as the authors have written not one but two follow-up articles to this piece, hopefully each proving as successful as the last.Another 2018 article hitting our most read list is from our Editorial Board member Antonio Rossi and colleague Lucia Anna Muscarella (Casa Sollievo della Sofferenza Hospital, Italy), commenting on the presence of the NRG1 gene in cases of non-small-cell lung cancer [2].Looking at the use of imaging methods for staging NSCLC, Vasiliki-Konstantina I Gkogkozotou and the team from the University of Athens (Athens, Greece) investigate the use of PET/CT and brain MRI in detecting the different stages of the cancer in patients, concluding that a combination of the imaging techniques can increase accuracy in staging and may help in reducing avoidable thoracotomies and lower morbidity rates [3].Editorial boardUnfortunately, 2018 saw the loss of our long-term editor-in-chief, Dr David Sugarbaker, who passed away last August. The legacy he leaves behind is great and he will long be remembered as a pioneer in the treatment of mesothelioma. He completed the first lung transplant in the state of Massachusetts, developed innovative multimodality therapies and pioneered the technique of extrapleural pneumonectomy. His work will continue through the mesothelioma program he started during his time at Baylor College of Medicine, serving patients and families from around the world. The original research study from Jasmine Shell and the team from the National Cancer Institute (MD, USA) investigated the use of 68-gallium DOTATATE PET-CT imaging in symptomatic patients with suspected neuroendocrine tumors though negative anatomic imaging [1]. Their study suggested the validity of using this imaging technique for determining a diagnosis in difficult cases as well as the benefit of utilizing this information to guide future treatment.Another 2018 article hitting the most-read list is a review from Kristin Long and Rebecca Sippel, from the University of Wisconsin (WI, USA), that discussed treatments for parathyroid carcinoma, both current and future [2]. With surgery currently being the most common method of treatment, the authors advised on the importance of collaborative research to improve early recognition methods as well as to identify novel adjuvant therapies.An editorial authored by Martyn Caplin of University College London (London, UK) introduced lutetium (177Lu) oxodotreotide following its recent approval in Europe for the treatment of gastroenteropancreatic neuroendocrine tumors [3]. We have received a variety of article types from authors across the world, each providing novel perspectives, as well as interesting and exciting developments in the fields of breast cancer diagnosis and treatment.The most-read article from Volume 7 is a review that also happens to be the most-read article that has ever been published in the journal. In this article, Kimberly Corbin and Robert Mutter, both of the Mayo Clinic (MN, USA), discussed the use of proton beam therapy for treatment of early-stage and locally advanced breast cancer [1]. The ongoing research into this novel treatment shows its promise for various clinical applications and highlights its potential for reducing side effects relative to current treatments. However, as with any new technique, there are multiple challenges to overcome before its clinical use. Further research is needed to establish the future for proton beam therapy and its role in breast cancer therapies.Our reviews have proved particular popular in 2018, with two more making it into our list of most-read articles. Leonie Voorwerk, Marije Kat and Marleen Kok from The Netherlands Cancer Institute Amsterdam (The Netherlands) reviewed the future for biomarkers in predicting an individual's response to immunotherapy [2]. Looking at the breast tumor microenvironment, as well as the putative biomarkers of other tumor types, the team discussed the factors that can guide future studies investigating the possibility of introducing personalized immunotherapy to clinical practice. A second review, written by Jennifer Zocca et al. (Maiden Lane Medical, NY, USA), gives a review of advances in pain management for breast cancer patients as well as potential future directions [3]. Yazeed Sawalha and Anjali Advani from the Taussig Cancer Institute (OH, USA) reviewed the management of acute lymphoblastic leukemia in older generations, discussing the current approaches and the challenges faced by those in this field [1]. Pointing out the poor prognosis for this age group, they stressed the need for more research in the area.Another 2018 article that has hit our most-read list is an editorial from Pratap Neelakantan and Adam Mead, both of the University of Oxford (UK), who described the history of chronic myeloid leukemia and how treatments have developed through the last 5000 years [2]. Remaining hopeful, they ended on a positive note, suggesting that if advancements continue at this rate then a cure could be in sight.We have also published several original research articles; one of our highlights is from the Harbin Institute of Hematology & Oncology (China), which provides an observational study of Chinese adults with relapsed/refractory Philadelphia-negative acute lymphoblastic leukemia [3].
