Novel fluorinated foldamers based on aminomethyl‐1,4‐triazolyl‐difluoroacetic acid (1,4‐Tz‐CF2) units were synthesized and their conformational behaviour was studied by NMR and molecular dynamics. Their activity on the aggregation of the human islet amyloid polypeptide (hIAPP) amyloid protein was evaluated by fluorescence spectroscopy and mass spectrometry. The fluorine labelling of these foldamers allowed the analysis of their interaction with the target protein. We demonstrated that the preferred extended conformation of homotriazolamers of 1,4‐Tz‐CF2 unit increases the aggregation of hIAPP, while the hairpin‐like conformation of more flexible heterotriazolamers containing two 1,4‐Tz‐CF2 units mixed with natural amino acids from the hIAPP sequence reduces it, and more efficiently than the parent natural peptide. The longer heterotriazolamers having three 1,4‐Tz‐CF2 units adopting more folded hairpin‐like and ladder‐like structures similar to short multi‐stranded β‐sheets have no effect. This work demonstrates that a good balance between the structuring and flexibility of these foldamers is necessary to allow efficient interaction with the target protein..

The 5-fluoro triazole amino acid scaffold prepared by halogen exchange has been incorporated into peptides. From the X-ray diffraction of the 5-fluoro triazole motif, the main observation was an important localization on one side of the negative potential surface. The fluorine atom reveals a cylindrical shape in its deformation electron density.
1,2,3-triazoles are important scaffolds in several fields, and there is still a demand to construct these compounds, most notably the disubstituted 4, 5 compounds. The latter are obtained in different ways, but the 4- and 5-halo triazoles are generally the key intermediates in their synthesis. These halogenated compounds are an extraordinary platform leading to a great chemical diversity around the 4 and 5 positions. However, their own pathways are generally put aside at the expense of the substituted triazoles. This review is focused on the main synthesis of 5- and 4-halo 1,2,3-triazoles from cycloaddition followed or not by halogenation reaction. Unlike the halogens (iodine, bromine, and chlorine), the 5- and 4-fluoro 1,2,3-triazoles will be presented separately due to their relatively more difficult and still recently challenging synthesis.

Small selective peptide derivatives and analogues are promising drug candidates for inhibiting this poorly known class of PPIs. In this review, we will highlight the main strategies developed for designing linear and cyclic peptide and peptidomimetic inhibitors of PPIs involving β-sheet structures. These compounds either do not adopt preferred conformations or can mimic protein secondary structures such as β-strands, β-hairpins or α-helices.

The local hydrophobicity of an amino acid residue in a peptide sequence can be determined by measuring the hydrophobicity index (phi(0)) by reversed-phase (RP) HPLC. Herein, the impact on the local hydrophobicity of the replacement of an amide by a monofluoroalkene unit in short peptides is discussed. Monofluoroalkene-containing dipeptides and tripeptides were synthesized, as well as their natural parent compounds, and the hydrophobicity indexes of these short peptides and peptidomimetics were determined. Comparison between the natural parent peptides and their alkene-containing analogues was made, and the dependence of the peptidomimetic analogues' behaviour on the pH and the solvent was studied. It was found that the presence of a monofluoroalkene unit enhanced a peptide's hydrophobicity.
Ces dernières décennies, les foldamères ont émergé comme un outil prometteur pour développer des molécules bioactives mimant les structures secondaires des protéines. Ces foldamères sont principalement utilisés pour moduler les interactions protéine-protéine impliquées dans de nombreuses maladies, notamment les amyloïdoses telles que la maladie d’Alzheimer, la maladie de Parkinson et le diabète de type II (DT2). Les amyloïdoses sont des maladies qui se caractérisent par la présence de dépôts de protéines insolubles dans les tissus. L'agrégation du polypeptide amyloïde des îlots (hIAPP) est liée à la dégénérescence des cellules bêta et à la pathogenèse du diabète de type 2. hIAPP est un polypeptide de 37 résidus sécrété par les cellules bêta du pancréas. On trouve des dépôts amyloïdes de hIAPP dans le pancréas de nombreux patients atteints de DT2. hIAPP est l'un des polypeptides les plus amyloïdogènes connus, et les agrégats de hIAPP sont toxiques pour les cellules bêta du pancréas. Dans le monde, environ 483 millions de personnes sont actuellement atteintes de diabète de type II (DT2), une prévalence qui, selon les prévisions, devrait atteindre 579 millions d'ici 2030. La prévalence croissante du diabète de type II, et les risques cardiovasculaires indésirables qui lui sont associés, sont désormais considérés comme un défi majeur de santé publique. Actuellement, les traitements disponibles pour le traitement du diabète sont symptomatiques et induisent de nombreux effets secondaires. Le ciblage de hIAPP est devenu une stratégie prometteuse pour trouver un traitement étiologique contre le DT2. Dans ce contexte, nous avons conçu des foldamères basés sur les 1,2,3-triazoles fluorés pouvant moduler le processus d’agrégation de hIAPP. Les 1,2,3-triazoles ont été utilisés pour leurs propriétés d’isostères de liaison peptidique avec une meilleure stabilité métabolique. L’atome de fluor, par ses propriétés particulières, permet d’induire des conformations et constitue une sonde en RMN du ¹⁹F, utile pour étudier les interactions ligand-protéine, la stabilité métabolique, le passage de membranes ou même la distribution d’un composé in vivo. D’une part, nous avons réalisé la synthèse d’homo- et d’hétérotriazolamères se basant sur les 1,2,3-triazoles N-difluorométhylés. D’autre part, nous avons effectué des travaux préliminaires sur les 5-fluoro-1,2,3-triazoles comme nouvelle architecture pour concevoir des foldamères fluorés. Les études conformationnelles ont été réalisées par RMN et dynamique moléculaire pour les triazolamères des 1,2,3-triazoles N-difluorométhylés. L’étude conformationnelle des 5-fluoro-1,2,3-triazoles a été réalisée par diffraction aux rayons X. De plus, des études préliminaires ont été réalisées par spectrométrie de fluorescence à la thioflavine-T pour évaluer la modulation de l’agrégation de hIAPP par ces foldamères. L’ensemble de ces travaux de thèse a pour objectif de développer le savoir autour des foldamères fluorés pour concevoir des molécules bioactives. À notre connaissance, aucun des foldamères fluorés décrit dans la littérature n’a d’application médicinale.
