Hintergrund/Einleitung: Patienten mit hereditärer Hämochromatose entwickeln infolge einer erhöhten duodenalen Eisenresorption eine progrediente Eisenüberladung. Es wird vermutet, dass Mutationen im Hämochromatosegen HFE eine Fehlprogrammierung duodenaler Enterozyten im Sinne eines paradoxen Eisenmangelzustands verursachen, mit einer erhöhten Eisentransporterexpression als Folge. Bisherige Studien zur Expression der duodenalen Eisentransporter in Hämochromatosepatienten und Tiermodellen haben jedoch zu kontroversen Ergebnissen geführt.
Background: Interleukin-10 is an immunosuppressive cytokine produced by a variety of cells such as monocytes, macrophages, B-and T-lymphocytes.It was proved that IL-10 is also secreted by different cancer types including ovarian cancer.In patients with advanced ovarian cancer IL-10 is overexpressed in sera and ascites.The role of intracellular expression of IL-10 in ovarian cancer microenvironment remains unclear.The aim of study was to compare the percentage of peripheral blood (PB), peritoneal fluid (PF) and cancer tissue CD4+ and CD8+ T cells producing IL-10 in ovarian cancer patients (OVC).Material and Methods: Three-color flow cytometry was used to evaluate the percentage of CD4+ and CD8+ T cells producing IL-10 from peripheral blood (n = 32), peritoneal fluid (n = 24) and tissue (n = 15) of ovarian cancer patients.Peripheral blood, peritoneal fluid and cancer tissue mononuclear cells were obtained by density gradient centrifugation using gradisol and washed twice with PBS.For CD4+ and CD8+ T cells producing IL-10 detection the cells were stimulated for 4 hours ex vivo with phorbol myristate acetate (PMA) and ionomycin.Cancer tissue was previously fragmented with a surgical blade and digested with an enzyme mixture.Results: The percentage of CD4+ T cells producing IL-10 was significantly higher in the cancer tissue (1.91%) than in the peripheral blood (0.24%).Additionally, we found that the percentage of CD4+/IL-10+ T cells was significantly higher in the peritoneal fluid (0.94%) compared with the peripheral blood (0.24%).There was no significant difference between the percentage of CD4+/IL-10+ T cells between the peritoneal fluid (0.94%) and ovarian cancer tissue (1.91%).The percentage of CD8+ T cells producing IL-10 was also significantly higher in the cancer tissue (0.44%) than in the peripheral blood (0.08%).There were no significant differences between the percentage of CD8+/IL-10+ T cells between the peripheral blood (0.08%) and the peritoneal fluid (0.24%) and between the peritoneal fluid (0.24%) and cancer tissue (0.44%).Conclusions: We demonstrated the presence of IL-10+ T cells in both CD4+ and CD8+ T lymphocytes in ovarian cancer patients.The highest percentage of CD4+/IL-10+ and CD8+/IL-10+ in ovarian cancer tissue suggest that these cells are accumulated in tumor tissue or that they may migrate from the peripheral blood to the tumor tissue.
Einleitung: Die Funktion von FATP4 (fatty acid transport protein 4) ist nach wie vor weitgehend unbekannt. Das FATP4-Gen wird u.a. in Dünndarm, Leber und Fettgewebe exprimiert. Es existieren Daten, die auf eine Rolle von FATP4 für die intestinale Lipidresorption hinweisen und das FATP4-Gen als mögliches Kandidatengen für das humane Insulinresistenzsyndrom identifizieren. Für FATP4 wurden bislang sowohl eine Funktion als originäres Transportprotein als auch als Acyl-CoA-Synthetase diskutiert. Von uns generierte Mäuse mit konstitutiver FATP4-Defizienz zeigen einen dem humanen Krankheitsbild der letalen restriktiven Dermopathie ähnlichen Phänotyp.

Humangenetische Analysen deuten darauf hin, dass FATP4 ein Kandidatengen für das Insulinresistenzsyndrom ist. Von uns generierte Mäuse mit konstitutiver FATP4-Defizienz sterben kurz nach der Geburt; ihr Phänotyp gleicht dem Krankheitsbild der letalen restriktiven Dermopathie beim Menschen. Mit der Generierung einer Mauslinie mit epidermisspezifischer FATP4-Defizienz gelang es uns kürzlich, das FATP4-Gen in der Maus zelltypspezifisch zu inaktivieren.

Der frühe Tod der Mäuse erschwert Untersuchungen zur Bedeutung von FATP4 in inneren Organen an diesem Mausmodell. Um die Funktion von FATP4 in bestimmten Geweben in vivo näher charakterisieren zu können, stellten wir eine Mauslinie mit konditionaler FATP4-Defizienz her. Methoden: Auf der Grundlage einer von uns etablierten Mauslinie mit generalisierter FATP4-Defizienz [1] stellten wir eine Mauslinie mit einem gefloxten, funktionell aktiven FATP4-Allel her. In diese Mauslinie kreuzten wir eine transgene Linie mit induzierbarer epidermisspezifischer Cre-Expression ein. Mittels PCR und Southern Blot analysierten wir die Rekombination von gefloxtem FATP4-Allel zu FATP4-Nullallel in einer Vielzahl von Geweben. Mäuse mit epidermisspezifischer FATP4-Defizienz wurden morphologisch und funktionell charakterisiert. Analog dazu stellen wir aktuell Mauslinien mit enterozyten- bzw. hepatozytenspezifischer FATP4-Defizienz her. Ergebnisse: Die hergestellte Mauslinie zeigt eine epidermisspezifische FATP4-Defizienz. Mäuse mit epidermisspezifischer FATP4-Defizienz entwickeln eine Proliferationshyperkeratose und eine Störung der epidermalen Barriere wie Mäuse mit konstitutiver FATP4-Defizienz. Schlussfolgerung: Die von uns hergestellte Mauslinie mit epidermisspezifischer FATP4-Defizienz ist die erste beschriebene Linie mit konditionaler FATP4-Defizienz. Ihr Phänotyp belegt, dass FATP4 in der Epidermis eine essentielle Bedeutung für die Ausbildung einer normalen Hautstruktur besitzt. Die Charakterisierung von Mäusen mit enterozyten- bzw. hepatozytenspezifischer FATP4-Defizienz wird Rückschlüsse auf die Funktion von FATP4 in Darm bzw. Leber erlauben.
So far, little is known about the physiological role of fatty acid transport protein 4 (Fatp4, Slc27a4). Mice with a targeted disruption of the Fatp4 gene display features of a human neonatally lethal restrictive dermopathy with a hyperproliferative hyperkeratosis, a disturbed epidermal barrier, a flat dermal-epidermal junction, a reduced number of pilo-sebaceous structures, and a compact dermis, demonstrating that Fatp4 is necessary for the formation of the epidermal barrier. Because Fatp4 is widely expressed, it is unclear whether intrinsic Fatp4 deficiency in the epidermis alone can cause changes in the epidermal structure or whether the abnormalities observed are secondary to the loss of Fatp4 in other organs. To evaluate the functional role of Fatp4 in the skin, we generated a mouse line with Fatp4 deficiency inducible in the epidermis. Mice with epidermal keratinocyte-specific Fatp4 deficiency developed a hyperproliferative hyperkeratosis with a disturbed epidermal barrier. These changes resemble the histological abnormalities in the epidermis of newborn mice with total Fatp4 deficiency. We conclude that Fatp4 in epidermal keratinocytes is essential for the maintenance of a normal epidermal structure.
The fatty acid transport protein family is a group of evolutionarily conserved proteins that are involved in the cellular uptake and metabolism of long and very long chain fatty acids. However, little is known about their respective physiological roles. To analyze the functional significance of fatty acid transport protein 4 (Fatp4, Slc27a4), we generated mice with a targeted disruption of the Fatp4 gene. Fatp4-null mice displayed features of a neonatally lethal restrictive dermopathy. Their skin was characterized by hyperproliferative hyperkeratosis with a disturbed epidermal barrier, a flat dermal-epidermal junction, a reduced number of pilo-sebaceous structures, and a compact dermis. The rigid skin consistency resulted in an altered body shape with facial dysmorphia, generalized joint flexion contractures, and impaired movement including suckling and breathing deficiencies. Lipid analysis demonstrated a disturbed fatty acid composition of epidermal ceramides, in particular a decrease in the C26:0 and C26: 0-OH fatty acid substitutes. These findings reveal a previously unknown, essential function of Fatp4 in the formation of the epidermal barrier.
