为探究华中地区犬副流感病毒(CPIV)的流行及遗传进化情况,利用RT-PCR方法对华中地区2020年9月—2022年1月收集的418份病料样品进行了CPIV检测,并利用生物信息学软件对分离的CPIV毒株的F、HN基因进行同源性、氨基酸位点分析及系统发育分析.结果显示:CPIV阳性率为15.8%,扩增获得9株CPIV的F、HN基因的序列,并成功分离培养9株CPIV毒株;同时,将分离株的F、HN基因与19株副流感病毒参考毒株进行序列比对分析,结果表明,F基因核苷酸同源性为95.1%~100%,氨基酸同源性为94.6%~99.8%,HN基因核苷酸同源性为95.9%~99.9%,氨基酸同源性为95.8%~99.6%;CPIV的F及HN基因与参考毒株相比均有部分氨基酸发生了突变;系统发育分析结果显示,本研究获得的8株分离株与美国株CPI-和CPI+处于同一分支上,亲缘关系较近,另一株分离株CS-324与中国株HeN0718和韩国株D277位于同一分支上.本研究丰富了华中地区CPIV的流行病学调查资料,并为该病的综合防控提供了理论依据.
为了解华中地区犬源CDV所属谱系、流行及遗传变异情况,本试验通过RT-PCR方法对华中地区2020年9月~2022年1月收集的418份犬源病料进行了检测,其中83份样品为CDV阳性,阳性率为19.9%.将阳性样品接种Vero细胞,并通过间接免疫荧光方法鉴定,成功获得13株CDV分离株.对13株CDV分离株H基因进行测序,并与32株CDV参考毒株进行同源性分析、遗传进化分析以及H蛋白潜在N糖基化位点预测分析.结果表明,分离到的13株野毒株与参考毒株相比,核苷酸同源性为87.1%~99.8%,氨基酸同源性为82.2%~100%;其中8株分离株在基因型上归属于Asia-1型,5株归属于America-1型;H基因抗原性预测分析表明,分离到的13株野毒株具有6~9个潜在的N糖基化位点.本研究为本土流行性CDV毒株的研究奠定了基础.
To analyze the prevalence of feline viral diseases in China, including feline panleukopenia virus (FPV), feline calicivirus (FCV), feline herpesvirus 1 (FHV-1), and feline coronavirus (FCoV) infectious diseases from 2018 to 2020, swab samples from 304 cats and serum samples from 193 cats in 18 cities were collected. The etiological investigation results of 304 cats showed that 256 (84.21%) cats were positive, infected with at least one virus, and the positive rates for FPV, FCV, FHV-1, and FCoV were 61.51%, 10.86%, 4.61%, and 55.92%, respectively. The mixed infection exhibited high complexity, and a total of eight mixed infection patterns were detected. The risk factor analysis of each pathogen in different clinical scenarios indicated that FPV positive status was significantly related to all the studied diseases, FCV positive status exhibited the most significant association with gingivostomatitis and conjunctivitis, and FHV-1 positive status was significantly related to upper respiratory tract disease, but FCoV positive status was not significantly related to any disease. Additionally, the prevalence of FPV exhibited a strong seasonality and was related to age, while the prevalence of FCV, FHV-1, and FCoV had nothing to do with season or age. FCV infection was sex related in cats, whereas the prevalence of FCV, FHV-1, and FCoV was not sex related. FPV, FCV, FHV-1, and FCoV were unrelated to breed or residential density. Antibody detection results of 193 serum samples by the virus neutralizing method indicated that the current commercial vaccines might not protect hosts against wild strains of FPV, FCV, and FHV-1 in China. In general, this study enriches epidemiological survey data of common viral diseases in cats in China and provides a theoretical basis for further development of vaccines.
为了解华中地区犬细小病毒2型(CPV-2)的流行趋势以及变异情况,本研究于2020年9月至2022年1月间采集华中地区的418份疑似感染犬细小病毒的粪便样品,通过PCR方法检测,利用F81细胞进行病毒分离、间接免疫荧光试验,并对分离病毒的VP2基因进行氨基酸序列及其同源性和遗传进化分析.结果显示,358份病料检测出CPV-2,阳性率为85.65%.病料接种F81细胞后,有23份阳性样品出现明显细胞病变,间接免疫荧光试验观察到明显荧光.VP2基因序列分析结果表明,分离毒株中有New-CPV-2a型12株、New-CPV-2b型5株、CPV-2c型6株.核苷酸同源性为97.9%~99.8%,氨基酸同源性为95.9%~100.0%.遗传进化分析结果显示,分离毒株与国内分离株、伊朗、巴西分离株亲缘关系较近,与意大利、日本、阿根廷分离株亲缘关系较远.本研究为华中地区犬细小病毒防控及研制高效疫苗奠定了基础.
Objective: To estimate the incidence of HIV-1 infection in men who have sex with men (MSM) in key areas of China through HIV-1 limiting antigen avidity enzyme immunoassay (LAg-Avidity EIA), analyze the deviation from the actual results and identify influencing factors, and provided reference for improving the accuracy of estimation results. Methods: Based on the principle of the cohort randomized study design, 20 cities were selected in China based on population size and the number of HIV-positive MSM. The sample size was estimated to be 700 according to the HIV-1 infection rate in MSM. MSM mobile phone app. was used to establish a detection appointment and questionnaire system, and the baseline cross-sectional survey was conducted from April to November 2019. LAg-Avidity EIA was used to identify the recent infected samples. The incidence of HIV-1 infection was calculated and then adjusted based on the estimation formula designed by WHO. The influencing factors were identified by analyzing the sample collection and detection processes. Results: Among the 10 650 blood samples from the participants, 799 were HIV-positive in initial screening, in which 198 samples (24.78%) missed during confirmation test. Only 621 samples were received by the laboratory. After excluding misreported samples, 520 samples were qualified for testing. A total of 155 samples were eventually determined as recent infection through LAg-Avidity EIA; Based on the estimation formula , the incidence of HIV-1 infection in MSM in 20 cities was 4.06% (95%CI:3.27%-4.85%), it increased to 5.53% (95%CI: 4.45%-6.60%)after the adjusting for sample missing rate. When the sample missing rate and misreporting rate were both adjusted, the incidence of HIV-1 infection in the MSM increased to 5.66% (95%CI:4.67%-6.65%). The actual incidence of HIV-1 infection in MSM in the 20 cities might be between 4.06% and 5.66%. Conclusions: Sample missing and misreporting might cause the deviation of the estimation of HIV-1 infection incidence. It is important to ensure the sample source and the quality of sample collection and detection to reduce the deviation in the estimation of HIV-1 infection incidence.
Objective:To identify the status and determinants of sharing personal HIV information with sexual partners among men who have sex with men (MSM) meeting their casual sexual partners on the internet.Methods:A cross-sectional study was conducted in five cities (Hangzhou, Ningbo, Wenzhou, Taizhou and Shaoxing) in Zhejiang province. The recruitment was enrolled by MSM social organization and in voluntary counseling and testing clinics, with a sample size of 793. A self-designed network questionnaire collected essential characteristics, HIV knowledge, sexual behavior, and sharing personal HIV status. SPSS 20.0 software was used for statistical analysis.Results:Among 767 MSM enrolled 302 MSM who reported finding casual sexual partners on the internet were enrolled in the analysis. MSM reported finding casual partners on the internet only, finding sexual partners online and in places were 62.6% (189/302) and 37.4% (113/302), respectively. Among those reporting web-based sexual behavior in the last six months, 54.6% (165/302) informed their partners of their HIV status, 49.2% (146/297) inquired about HIV status, and 42.9% (82/191) knew HIV status before sex intercourse, 75.8% (113/149) reported consistent condom use with HIV negative partners. The multivariable logistic regression analysis showed that related factors of inconsistent inquired HIV status of partners included 25-34 years old (a OR=2.17, 95% CI: 1.20-3.91), >2 partners on the internet in the last six months (a OR=2.13, 95% CI: 1.27-3.57), low-risk perception of HIV infection with online partners (a OR=1.96, 95% CI:1.14-3.35), numbers of HIV testing >1 times (a OR=0.38, 95% CI: 0.22-0.66). Conclusions:The willingness to know the HIV status of partners among MSM who met sexual partners on the internet was high but with a low rate of knowing their sex partner's HIV status in Zhejiang province. However, the successful implementation proportion was low. Therefore, it is necessary to pay attention to people who are elderly, with less conscience about the risk of the sex partners on the internet, have more sex partners, and have received few HIV tests. In addition, peer education was needed to promote related intervention programs.
Objective:To provide information reference for resource allocation and decision-making in related fields, the cost-effectiveness of HIV input among men who have sex with men (MSM) in Ningbo. Different intervention coverages were compared.Methods:Taking MSM as the target population, data were collected and modeled by Optima HIV for the corresponding HIV health output and the budget under different intervention coverages.Results:According to the estimated size of the MSM population, which was 19 584 in Ningbo in 2020, if the coverage of 2020 baseline intervention is maintained in the next ten years, the number of HIV cases, new HIV infections, and HIV-related deaths among this population will show an upward trend. It is estimated that from 2021 to 2030, 7.9% of new infections and 1.7% of deaths can be avoided and the relevant funding investment comed to 2.4 time the baseline if the intervention coverage rate expanded to 3.0 times the 2020 baseline. After the coverage rate of intervention expanded to 3 times the baseline, it continued to grow, the health effect did not increase.Conclusions:At present, expanding the baseline coverage of HIV-related intervention projects among MSM in Ningbo and increasing capital investment will still reverse HIV-related death and reduce new infections. Moreover, there is a saturation point of the intervention effect. Researchers and policymakers must explore more effective interventions/combinations to obtain more significant health outcomes.
Objective: To understand the epidemiological characteristics and transmission routes of newly reported HIV/AIDS cases with non-martial and non-commercial (NMNC) heterosexual behaviors in Zhejiang province. Methods: A retrospective survey was conducted among HIV/AIDS cases that had NMNC heterosexual behavior history and diagnosed with HIV infection in Zhejiang between January 1st, 2017 and September 30th, 2019. The multiple correspondence analysis (MCA) was used to explore the association of social demographic characteristics with NMNC heterosexual partner types in the cases. Results: A total of 406 participants with NMNC heterosexual behaviors before HIV diagnoses were recruited in this study. Most of them were males (67.2%, 273/406), aged 25-49 years at HIV diagnoses (59.1%, 240/406). Prior to HIV infection confirmation, 36.0% (146/406) participants only had casual sexual partners, 52.0% (211/406) only had regular sexual partners, and 12.0% (49/406) had both. Statistical differences in marital status, occupation and income level were found among participants with different types of NMNC heterosexual partners (all P<0.05). Result of MCA indicted that monthly income ≤3 000 yuan RMB, self-employed, being married were only associated with casual NMNC heterosexual partner; working in service industry, education level of junior high school were only associated with regular heterosexual partner; working in enterprise, high school education level or above, monthly income ≥5 000 yuan RMB were associated with both casual and regular sexual partner. Conclusion: The HIV/AIDS cases with NMNC heterosexual behaviors before HIV diagnoses accounted for a large proportion in Zhejiang province during 2017-2019. Active intervention efforts should be made to improve the health awareness of the public to reduce the risk behaviors for HIV transmission.
Objective:To analyze the prevalence of anal sex and related factors in low-tier female sex workers (FSWs) in the demonstration areas of comprehensive AIDS responses (DACAR) in Zhejiang province.Method:In 2013, a survey on low-tier FSWs was carried out in 21 Demonstration Areas of AIDS Responses in Zhejiang. Experience of having anal sex with clients in the last month was taken as dependent variable while independent variables would include perception and characteristics related to behavior of the FSWs. The multivariate logistic regression analyses were conducted to identify the factors related to anal sex in low-tier FSWs.Results:A total of 2 645 low-tier FSWs were interviewed. Among these FSWs, 67.9% (1 796/2 645) were over 26 years old, 89.9% (2 378/2 645) were with education level of junior high school or below, 58.4% (1 546/2 645) were married, 78.2% (2 068/2 645) were from other provinces, 68.6% (1 814/2 645) had income level less than 4 000 Yuan, and rates of anal sex with client in the last month were 5.03% (133/2 645). Results from the multivariate logistic regression analysis of related factors of anal sex with client in the last month showed that among the low-tier FSWs, the venues of having commercial sex were in hair salons/massage parlors ( OR=7.31, 95 %CI: 2.27-23.59), roadside shops ( OR=7.89, 95 %CI: 2.27- 27.40) or other places ( OR=4.65, 95 %CI: 1.23-17.53), when compared with the street-women service. FSWs often engaged in commercial sex business in three or more counties ( OR=1.68, 95 %CI: 1.01-2.81), when compared with FSWs often engaged in commercial sex business just one county. FSWs had sex with only middle-aged and the elderly clients ( OR=0.45, 95 %CI: 0.30-0.66), when compared with FSWs had sex with young or middle-aged and the elderly clients. FSWs had risk perception for sexually transmitted disease (STD) ( OR=2.00, 95 %CI: 1.20-3.32), when compared with FSWs with no risk perception. FSWs had oral sex experience with their clients in the last month ( OR=7.29, 95 %CI: 4.78-11.12), when compared with the ones without oral sex. The above said factors were all related to the incidence of anal sex. Conclusions:Certain numbers of low-tier FSWs had anal sex experiences with their clients, especially those who had oral sex were more likely to have anal sex in DACAR in Zhejiang province. Attention should be paid to anal sex behavior when conducting behavior-related intervention programs, targeting on the low-tier FSWs.
Determining the longevity of natural immunity after SARS-CoV-2 infection is critical for understanding immune protection and optimizing vaccine design. Over six months have been passed since the emergence of COVID-19 in China. We evaluated humoral and cellular responses in 418 patients six months after infection. 95.9% and 98.1% of the patients maintained SARS-CoV-2-specific IgG and neutralizing antibodies. All severe patients were positive for IgG and neutralizing antibodies and had significantly higher titers than mild and moderate patients as well as asymptomatic individuals. The patients had a more robust SARS-CoV-2-specific CD4+ T cell response than CD8+ T cells six months after infection. Unexpectedly, sustained immune activation was observed, which displayed as the evaluated proinflammatory monocytes, non-classical NK cells, CD4+ Treg cells, and activated CD4+ T cells. Objective: To understand the situation and influencing factors related to the promotion of HIV testing program among sex partners in newly diagnosed HIV positive MSM in Zhejiang province during 2015 to 2017. Methods: Newly diagnosed HIV positive MSM in Zhejiang province from 2015 to 2017 were collected and provided four rounds of testing services to their sexual partners so as to study the consequences. Chi-square test was conducted to compare the difference between groups while multivariate logistic regression was conducted to analyze the related influencing factors. Results: A total of 6 269 HIV positive MSM were provided with four rounds of testing services to their sexual partners. 1 925 HIV positive MSM (30.7%, 1 925/6 269) were successfully persuaded in mobilizing their sexual partners to participate in the HIV testing services. However, 4 344 HIV positive MSM (69.3%, 4 344/6 269) refused to do so. A total of 2 126 sexual partners received HIV testing and the HIV positive rates of those sexual partners appeared as 13.0% (277/2 126, 95%CI:11.6%-14.5%) and 78.7% (218/277, 95%CI: 73.8%-83.6%). Results from the multivariate logistic regression analysis indicated that factors as: HIV positive MSM that participated in the study in 2016 (compared with participated in the study in 2015, OR=1.581, 95%CI: 1.370-1.823), in 2017 (compared with participated in the study in 2015, OR=1.394, 95%CI: 1.208-1.608), living in the reporting city (compared with lived outside the reporting city, OR=1.518, 95%CI: 1.320-1.745), being married (compared with unmarried/divorced/widowed, OR=4.449, 95%CI: 3.837-5.160), having education level of junior high school or below (compared with education level of senior high school or above, OR=1.203, 95%CI: 1.058-1.367), numbers of homosexual partners >5, (compared with numbers of homosexual partner from past between 1-5, OR=1.236, 95%CI: 1.095-1.395), active detection (compared with passive detection, OR=1.340, 95%CI: 1.193-1.506) were more likely to relate to the successful persuasion on their sexual partners to receive the HIV testing. There was no statistical difference noticed between HIV-infected homosexual partners and their corresponding HIV positive MSM, in terms of socio-demographic situations. Conclusions: HIV positive MSM should promote their sexual partners to receive HIV testing. This seemed an important role in expanding the HIV testing in this population. However, the program needs to be further improved and included in daily work, focusing on those partners with similar social and demographic characteristics with those infected MSM.
Acute gastroenteritis (AGE) is one of the most frequently occuring illnesses in children and adults worldwide. In February 2017, two AGE outbreaks occurred in two adjacent schools in Huzhou city, Zhenjiang province of China. We detected high percentages of recombinant norovirus GII.P16/GII.2 in one school and chicken anemia virus (CAV) in another school using next generation sequencing (NGS) and specific PCR. The results highlight the importance of continuous surveillance of GII.P16/GII.2, and suggest the need of further studies on whether CAV causes AGE. Objective: To understand the sources and characteristics of newly reported HIV/AIDS cases in Yiwu, Zhejiang province, from 2015 to 2016. Methods: From January 1, 2015 to December 31, 2016, newly reported HIV/AIDS cases in Yiwu were recruited in the study and they were asked to complete an in-depth field epidemiology questionnaire. Information related to their previous sexual behaviors and infection sources or routes were collected in the survey. The data were processed and analyzed with EpiData 3.0 and SPSS 19.0 software. Results: A total of 320 participants from 393 newly reported HIV/AIDS cases in Yiwu during this period were surveyed and recruited in the study. Most of them were males (268, 83.8%), unmarried (141, 44.1%), aged between 25-44 years (166, 51.9%), had census register in other provinces (217, 67.8%) and educational level of junior high school (128, 40.0%). Among 305 cases whose infection times could be determined, 107 (35.1%) were newly infected within one year, and among 267 cases whose infection location could be determined, 200 (74.9%) were infected in Yiwu. New infections were mainly detected in those who had homosexual behaviors or causal sex partners (during the past year) and through active detection approach. Local infections were mainly among those who had local residence and commercial heterosexual behavior in Yiwu. Conclusions: Most newly reported HIV/AIDS cases in Yiwu from 2015-2016 were infected for more than one year and locally infected. Census registration of other provinces, homosexual sexual behavior, and casual sexual behavior give major contribution to HIV/AIDS epidemic in Yiwu.
Methicillin-resistant Staphylococcus aureus (MRSA) has emerged as a serious hospital and community-acquired infection and some strains are associated with greater severity. We investigated the clinical variability and molecular characteristics of MRSA infections in Shenzhen, China through a study at nine sentinel hospitals from January to December 2014. MRSA infections were classified as community-associated (CA-MRSA), healthcare-associated (HA-MRSA), and healthcare-associated community-onset (HACO-MRSA). In total, 812 MRSA isolates were collected and 183 of these were selected for further study. Patients with HA-MRSA infections were generally of greater age compared to other groups. Distinct body site and clinical presentations were evident in infected patients, e.g. CA-MRSA (skin and soft tissue, 53%), HA-MRSA (respiratory tract, 22%; surgical site, 20%; trauma wounds, 20%) and HACO-MRSA (mastitis, 47%). In contrast to HA-MRSA, other categories of strains were significantly more susceptible to gentamicin, sulfamethoxazole/trimethoprim, and tetracycline. No resistance to vancomycin or linezolid was recorded. The predominant clonal lineage within each strain category was CC59-t437-SCCmec IV/V-agr I (CA, 51·4%; HA, 28·9%; HACO, 52·9%) which exhibited characteristics of a traditional CA clone together with agr I which is more often associated with HA clones. In conclusion, for the three categories of MRSA infections, there were significant differences in clinical characteristics of patients, but the predominant clone in each category shared a similar genetic background which suggests that transmission of MRSA strains has occurred between the community and hospitals in Shenzhen.

SUMMARY The aim of this study was to characterize updated HIV subtypes in Yunnan to determine their origins and distribution within the population. RT–PCR of both the gag and env genes were sequenced from Yunnan province inhabitants newly diagnosed with HIV-1. Sequence data from 290 samples were used for statistical analysis of subtype distribution and phylogenetic tree construction. Distribution data were adjusted to account for different geographical distributions of HIV-1 subtypes in the population. Phylogenetic analysis revealed six HIV-1 subtypes in Yunnan, including eight types of unique recombination forms (URFs). The most prevalent subtypes in this province, CRF07_BC (18·9%), CRF08_BC (39·1%), CRF01_AE (22·4%), and URFs (subtype C, 5·9% and subtype B, 4·5%), were all recombinants. We found significant differences in the distribution of these HIV-1 subtypes not only geographically, but also between various ethnic groups and with respect to transmission routes. Our findings indicate a complex population of HIV-1 subtypes, URFs, and recombinant subtypes in Yunnan province. This diversity could make the prevention and control of HIV infection in Yunnan more difficult due to the possibility of virus recombination or infection by multiple subtypes.
OBJECTIVE:To investigate prevalence and risk factors of syphilis infection among female sex workers (FSWs) in Shenzhen, China. METHODS:Observational study among (2009-2012) 1653 FSWs recruited by venue-based sampling using questionnaire-based interviews for socio-demographics, behaviours and syphilis testing results. Logistic regression was used to assess risk factors of syphilis infection. RESULTS:The overall syphilis prevalence was 4.7%, showing a slightly decreasing trend. Factors significantly associated with syphilis infection were inconsistent condom use (OR = 1.87, P = 0.015), illicit drug use (OR = 5.45, P < 0.001) and older age in years (OR = 1.08, P < 0.001). Venues where FSWs were recruited and duration of commercial sex work were not significantly associated with syphilis infection (P > 0.05). CONCLUSIONS:Syphilis is still common among FSWs in Shenzhen, China. Current comprehensive prevention programmes (e.g. condom promotion and peer education) should be continued to maintain and increase safe sexual practices and to reduce illicit drug use among FSWs. Expanding point-of-care syphilis screening programmes may be an important strategy for early diagnosis. We recommend timely and effective treatment programmes to be linked to such screening programmes.
UNLABELLED:In this study, we are trying to investigate the indoor air pollution and to estimate the residents' pollution exposure reduction of energy altering in rural Tibet. Daily PM(2.5) monitoring was conducted in indoor microenvironments like kitchen, living-room, bedroom, and yard in rural Tibet from December 2006 to March 2007. For kitchen air pollution, impact of two fuel types, methane and solid biomass fuels (SBFs), were compared. Questionnaire survey on the domestic energy pattern and residents' daily activity pattern was performed in Zha-nang County. Daily average PM(2.5) concentrations in kitchen, living-room, bedroom, and yard were 134.91 microg/m(3) (mean, n = 45, 95%CI 84.02, 185.80), 103.61 microg/m(3) (mean, n = 21, 95%CI 85.77, 121.45), 76.13 microg/m(3) (mean, n = 18, 95%CI 57.22, 95.04), and 78.33 microg/m(3) (mean, n = 34, 95%CI 60.00, 96.65) respectively. Using SBFs in kitchen resulted in higher indoor pollution than using methane. PM(2.5) concentrations in kitchen with dung cake, fuel wood and methane use were 117.41 microg/m(3) (mean, n = 18, 95%CI 71.03, 163.79), 271.11 microg/m(3) (mean, n = 12, 95%CI 104.74, 437.48), and 46.96 microg/m(3) (mean, n = 15, 95%CI 28.10, 65.82) respectively. Family income has significant influence on cooking energy choice, while the lack of commercial energy supply affects the energy choice for heating more. The effects of two countermeasures to improve indoor air quality were estimated in this research. One is to replace SBFs by clean energy like methane, the other is to separate the cooking place from other rooms and by applying these countermeasures, residents' exposure to particulate matters would reduce by 25-50% (methane) or 20-30% (separation) compared to the present situation.PRACTICAL IMPLICATIONS:Indoor air pollution caused by solid biomass fuels is one of the most important burdens of disease in the developing countries, which attracts the attention of environment and public health researchers, as well as policy makers. This paper gives a pilot research on the indoor air pollution and estimated the effects of some intervention policies in Tibet of China, where the living habits of the residents are quite different from other parts of the world. This work would be an important supply to the indoor air pollution studies, and would be helpful in policy making.
The heat shock 70 kDa protein 5 (HSPA5) gene is known to be involved in stress-associated diseases. In this study, the promoter, exons, 3' untranslated region (3'UTR), and subtotal introns of the HSPA5 gene were sequenced in a sub-population of 161 healthy Han Chinese. Nine single-nucleotide polymorphisms (SNPs) including a new one (-86bp T > A from the estimated translation start site) were found and 15 haplotypes (frequencies > 1%) were inferred. Polymorphisms rs391957 and rs11355458 were completely linked in our population (r (2) = 1.00). Using this information, fellow scientists may be able to decrease the number of SNPs to be genotyped in future disease case-control studies.
