FT-NIR spectroscopy with a fiber optical reflection probe was applied as a process analytical technology tool for the continuous quantitative in-line monitoring of pharmaceutical powder mixing processes in a bladed mixer. Two powders, acetyl salicylic acid as an active pharmaceutical ingredient (API) and α-lactose monohydrate as an excipient were characterized in advance in terms of shear cell tests, flowability tests and particle-size determination to deduce flow properties of the powders. For the quantitative monitoring of the API content, two predictive models were developed with partial-least-squares calibration based on off-line calibration. On the basis of these predictive models, powder agitation and mixing times until blend uniformity were quantitatively monitored. Mixing experiments with systematically varied filling levels and filling protocols showed a strong variation in mixing, but eventually yielded uniform powder blend. Simulation results from the literature were linked to our experimental findings in order to identify and elucidate the effects of convective and diffusive mixing. In accordance to the international conference on harmonization acceptance level of 5% for the nominal API content, UV/Vis reference measurements were performed to verify the blend uniformity as predicted by the NIR measurements.
Streptomyces coelicolor “Müller” DSM3030 excretes a lysozyme comprising both β-1,4-N-acetyl-and β-1,4-N,6-O-diacetyl muramidase activities. The lysozyme is named Cellosyl. Gene libraries have been established using genomic DNA from the wild-type strain, S. coelicolor DSM3030, and from an overproducing mutant, S. coelicolor HP1, which exhibits about a twofold increase in lysozome production. The lysozyme-encoding genes (cel) from both strains were detected by oligodeoxynucleotide hybridization. The nucleotide sequence of the cel genes isolated from both strains was shown to be identical. The different levels of lysozyme production could not be correlated with any mutations at the cel gene locus. The cel gene isolated from the wild-type strain could not be expressed in some other species of Streptomyces. However, self-cloning of the cel gene into S. coelicolor DSM3030 and HP1 resulted in a 2.5-fold increase in lysozyme production.
Low molecular weight β, 1 → 4-glucans (cellodextrins) are favorably prepared by cleavage of cellulose in liquid hydrogen fluoride at temperatures between −15 and −30°C. This is due to the suppression of the reversion reaction in that low temperature range. Under conditions favorable for reversion the presence of water leads to a competing hydrolysis and lower the average degree of polycondensation. For the preparation of defined O-glycosides from glucose and alcohols, hydrogen fluoride is not suitable as a reaction medium. Under reversion conditions, monohydroxy compounds are inferior in their reactivity to the competing carbohydrate molecules, and polyols like sorbitol furnish mixtures of isomeric glycosides. Gaseous hydrogen fluoride represents a highly suitable agent for the degradation of carbohydrate and lignin containing biomass, such as waste wood, for the purpose of providing fermentation raw material. As a model, lignocellulose was studied and the heat of reaction of the hydrogen fluoride sorption and desorption processes were examined. The practically important desorption value was found to be approximately 870 kJ/kg HF.

AbstractDurch eine neu entwickelte, zweistufige Extraktion der Zelltrockenmasse von Bakterien, Hefen oder Pilzen kann fett‐ und nukleinsäurearmes Single Cell Protein (SCP) gewonnen werden. Die erste Stufe dient dem Aufschluß der Zellen, wobei die Lipide aus Zellwand und Membran unter Zerstörung der Zellstruktur in organischen Lösungsmittel/NH3‐Gemischen gelöst werden. Dieser Aufschluß ermöglicht die Extraktion der Nukleinsäuren (DNA + RNA) unter sehr einfachen, schonenden Bedingungen mit Wasser. Nach diesem neuen Verfahren lassen sich SCP‐Produkte mit hohem nutritiven Wert herstellen.
AbstractDas Fünfringsystem des Germadisiladiazans wurde erstmalig gemäß Schema 1 in den Verbindungen A 1, 8, 13 dargestellt. Diese ließen sich zu den Derivaten A 2–7 und 9–12 weiter umsetzen (Schema 2). Alle Verbindungen werden in ihren Eigenschaften beschrieben (Tab. 2). Ihre Konstitution geht aus Elementaranalysen (Tab. 3), Massen‐, 1H‐NMR‐ und IR‐Spektren eindeutig hervor (tab. 4–6).
