Angiogenese und Immuntoleranz sind zwei pathophysiologische Mechanismen, die vom hepatozellulären Karzinom (HCC) beeinflusst werden. Sie dienen daher als therapeutische Angriffspunkte, wie z.B. in der Kombinationstherapie aus Bevacizumab (anti-VEGF, Anti-Angiogenese) und Atezolizumab (anti-PD-L1, Immuncheckpoint Therapie, ICI), welche aktuell die Standardtherapie des fortgeschrittenen HCC darstellt (IMbrave150). Neben Tumorzellen exprimieren insbesondere anti-inflammatorische tumor-assoziierte Makrophagen (TAM) sowohl VEGF als auch PD-L1. Die Regulation der TAM Differenzierung ist dabei u.a. abhängig von Chemokin-Signalwegen.
Einleitung Die Pathogenese der portalen Hypertension ist multifaktoriell und beinhaltet eine pathologisch intrahepatische Angiogenese. Angiogenin ist ein potenter Mediator der Angiogenese, allerdings ist dessen Stellenwert in der Pathogenese der portalen Hypertension bisher noch unbekannt.
Macrophage Migration Inhibitory Factor (MIF) is a proinflammatory cytokine with chemokine-like functions. Analysis of the GEPIA database revealed an increased expression of MIF in human hepatocellular (HCC) tumor tissue and high expression levels were associated with decreased survival in HCC patients. Referring to the MIF receptors, CD74 has been identified as an important target of MIF in malignant diseases. In this study, we analyze the functional role of MIF and its receptor CD74 in HCC progression in vitro and in vivo using an experimental murine model of HCC.
Die Pathogenese des hepatozellulären Karzinoms (HCC) ist durch ein komplexes Wechselspiel zwischen verschiedenen Leber-residenten Zellen und infiltrierende Immunzellen geprägt. Der Chemokinrezeptor CXCR3 ist auf Immunzellen aber auch auf Leber-residenten Zellen und Tumorzellen exprimiert. Bei der Promotion und Progression von HCCs spielen sogenannte Tumor-assoziierte Makrophagen (TAM) eine prominente Rolle. TAMs differenzieren sich aus Monozyten, welche sind über verschiedenen Stadien z.B über das Allgemeine Myeloide Progenitor (CMP)-Stadium und das Granulozyten-Monozyten-Progenitor (GMP)-Stadium, aus hämatopoetischen Stammzellen entwickeln.
Liver fibrosis, caused by fatty degeneration, is a major cause of liver failure, and is also associated with higher levels of hepatocellular carcinoma. Although the pathophysiologic process leading to liver fibrosis is not completely clarified, stellate cells appear to play an important role. In this study, we have investigated a possible interplay between hepatocytes under normal and steatotic conditions. Our hypothesis was that extracellular vesicles (EV) isolated from hepatocytes could influence the behavior and phenotype of stellate cells.
Metabolic zonation of healthy liver lobules is extensively studied. However, little is known how chronic liver disease (CLD) influences lobular zonation. To bridge this gap, we studied metabolic zonation in two mouse models of CLD: repeated intoxication with carbon tetrachloride (CCl4; 1 gram/kg) twice a week up to one year, and bile duct ligation (BDL) for 21 days. Disease progression and its impact on metabolic zonation was analysed by histopathology, immunohistochemistry, image analyses, RT-PCR, as well as RNA-sequencing (RNA-seq) techniques.
Intrahepatic cholangiocarcinoma (ICC) is a highly aggressive cancer that ranks the second most common liver cancer in the world. It is associated with very poor prognosis and limited treatment options. Well-defined animal models recapitulating human disease to dissect molecular mechanisms and identify and evaluate new therapeutic strategies in preclinical settings are sparse. of rise and progression of cholangiocarcinoma animal models are needed. Here we present an murine orthotopic transplant model on B6 background resulting in ICCs with high molecular homology to human ICC within two to four weeks.
Caloric restriction is known to improve inflammatory and autoimmune diseases. However, the mechanisms by which reduced caloric intake modulates inflammation are poorly understood. Here we show that short-term fasting reduced monocyte metabolic and inflammatory activity and drastically reduced the number of circulating monocytes. Regulation of peripheral monocyte numbers was dependent on dietary glucose and protein levels. Specifically, we found that activation of the low-energy sensor 5′-AMP-activated protein kinase (AMPK) in hepatocytes and suppression of systemic CCL2 production by peroxisome proliferator-activator receptor alpha (PPARα) reduced monocyte mobilization from the bone marrow. Importantly, we show that fasting improves chronic inflammatory diseases without compromising monocyte emergency mobilization during acute infectious inflammation and tissue repair. These results reveal that caloric intake and liver energy sensors dictate the blood and tissue immune tone and link dietary habits to inflammatory disease outcome.
HCC progression is linked to the dynamic process of neovascularization, chronic liver inflammation and the crosstalk between tumor cells, peritumoral parenchymal cells as well as non-parenchymal cells e.g. infiltrating immune cells. The chemokine CXCL10 is implicated in these processes and plays a key role during acute and chronic liver injury in men and mice. Here we aimed to investigate the role of CXCL10in HCC progression.
Macrophage migration inhibitory factor (MIF) ist ein inflammatorisches Zytokin mit chemokin-ähnlichen Funktionen. In vorherigen toxischen Leberschädigungsmodellen konnten wir zeigen, dass MIF über CD74/AMPK antifibrotische Eigenschaften vermittelt, sowie in Modellen der nicht-alkoholischen Steatohepatitis (NASH) antisteatotische Effekte aufweist. Ziel der vorliegenden Studie war, die Rolle von MIF in der NASH assoziierten hepatischen Fibrosierung zu definieren.

Hinsichtlich chronischer Lebererkrankungen konnte bisher ein Zusammenhang zwischen der Schwere der autoimmunen Hepatitis und dem Genotyp C/C im SNP -173 G/C dargestellt werden.
Hintergrund: CXCR3-Liganden (CXCL9, CXCL10, CXCL11) spielen eine zentrale Rolle bei der hepatischen Inflammation und Fibrose. Kürzlich konnte gezeigt werden, dass die CXCR3-Liganden CXCL9 und CXCL11 mit der Ätiologie und dem Langzeitüberleben von Patienten mit Leberzirrhose und TIPS-Implantation korrelieren. Hingegen ist die Rolle, die CXCL10 bei schwerer portaler Hypertension spielt noch nicht ganz geklärt.
