Careya arborea Roxb, commonly known as wild guava which belonging to the family lecythidaceae which is popular as Padmaka, Kumbhi in Ayurveda. This tree can be identified by its thick dark grey bark, large showy flowers and leaves which turn red in winter. It is a significant medicinal value plant, hence most of the plant parts used in traditional systems of medicine. It has been used as antimicrobial, astringent, demulcent, antitumor, antipyretic and antipruritic, hepatoprotective, antimicrobial, antioxidant, CNS activity, antileishmanial, antidiarrheal, anticoagulant, analgesic, antitumour and cytotoxic activity. The pharmacological activity of the plant depends upon the chemical constituents present in the plants. It is used as astringent, demulcent, antitumour, antipyretic and antipruritic, hepatoprotective, antimicrobial, antioxidant, CNS activity, antileishmanial, anticoagulant, analgesic, cytotoxic activity. The authenticated barks were procured from Sri Venkateshwara University, Tirupathi. The coarse powder of bark of Careya arborea subjected for extraction process by using soxhlet apparatus by using successive solvents. The methanol solvent produces highest extractive yield. Therefore the methanolic extract of bark was used for optimization of solvent systems for TLC studies. The mobile phase selected was n-butanol: glacial acetic acid: water (4:1:5) showed best pattern of band separation. So that the methanolic extract was subjected to column chromatography. Therefore the extract packed into column and eluted with hexane: acetone to obtain 11 fractions. The 10th fraction showed the three bands with similar chromatographic pattern by TLC identity tests. This fraction was subjected for preparative column for isolation and purification of compound. Purity of the isolated compound was determined by HPLC analysis. Structural determination/ characterization was done by UV, IR spectroscopic analysis.

The authenticated barks were procured from Sri Venkateshwara University, Tirupathi. In the present study the dried and powdered drug was successively extracted with petroleum ether, chloroform, methanol, ethanol and water by continuous hot extraction using soxhlet assembly. The respective percentage yields were recorded with regular intervals. All the extracts were subjected to antimicrobial activity by agar diffusion method out of which the methanolic extract showed significant antimicrobial activity against bacteria and fungus.
