Acute myeloid leukemia (AML) is the most common acute leukemia in adults, associated with poor prognosis and easy relapse of disease. Circular RNAs (circRNAs) were detected to be m6A modified and the role of m6A circRNAs has been reported in other diseases including cancers, however, their role has not been elucidated in AML yet. In the present study, we aimed to investigate the expression profiling of m6A circRNAs in AML. We performed m6A circRNAs microarray analysis to identify differentially expressed m6A circRNAs in bone marrow samples from AML patients and healthy individuals (control). Furthermore, bioinformatics analysis predicted the potential functions and relevant pathways that may be associated with the m6A circRNAs. The circRNA m6A methylation levels were found to be positively associated with the circRNAs expression, suggesting circRNA m6A modification could contribute to circRNA regulation in AML. Further analysis demonstrated that circRNA m6A modification might influence the circRNA-miRNA-mRNA co-expression network that may contribute to the circRNA regulatory network in AML. Our findings provide evidence of the differential expression profile of m6A circRNAs in AML, and circRNA m6A modification may contribute to circRNA regulatory function in AML.
目的 探讨血管紧张素Ⅱ 1型受体(AT1R)基因DNA甲基化和环境因素在冠状动脉粥样硬化性心脏病(冠心病)中的作用.方法 于2014年4月至2015年9月,对福建省福清市、长乐市和南安市共8720人进行流行病学调查和实验室检测.采用病例对照研究方法(选择冠心病患者46例和对照351人),高分辨率熔解曲线(HRM)试剂盒检测唾液标本AT1R基因的DNA甲基化,叉生分析、Logistic回归模型与Delta法分析AT1R基因甲基化水平与环境因素的联合及交互作用.结果 多因素Logistic回归分析结果显示,体质量指数(BMI)偏高(OR=1.712,95% CI 1.289~2.274)和中心性肥胖(OR=1.353,95% CI 1.003~1.825)是冠心病的危险因素;每周饮酒≤3次(OR=0.463,95% CI 0.245~0.876)和体育锻炼(OR=0.804,95% CI 0.686~0.943)是冠心病的保护因素.调整人口学与环境因素后,AT1R基因DNA甲基化水平偏低是冠心病患病的危险因素.叉生分析显示,AT1R基因DNA甲基化程度与中心性肥胖基于相加模型的交互作用有统计学意义(U=2.105,P=0.035),归因比=57.9%,相对超额危险度=4.86.结论 AT1R基因DNA甲基化程度与中心性肥胖对冠心病具有叠加交互作用,提示冠心病是环境因素和表观遗传因素共同作用的结果.
Objective To identify the hospital workplace violence pattern,and to provide a basis for targeted preventive measures.Methods Totally 1950 doctors and nurses were selected by Multi.stage stratified cluster sampling method and investigated with a violence questionnaire.Latent class analysis was used to identify the latent class and classify the violence pattern with different distribution characteristics.Results There were three types of violence pattern:high frequency group,moderate frequency group and low frequency group.The higher the frequency of violence occurred,the lower job satisfaction was.The high risk violence population were Men,aged 30 ~ 40 years,night shift and bachelor degree or above.Conclusion Latent class model can be used to classify the violence pattern for revealing key populations of workplace violence prevention and providing a basis for violence prevention measures.
环状RNA (circular RNA,circRNA)是继微小RNA (microRNA,miRNA)和长链非编码RNA (long noncoding RNA,lncRNA)之后一种新兴的内源性非编码RNA (noncoding RNA,ncRNA),在最近几年逐渐受到广泛关注,是目前RNA研究的热点.近年来越来越多的研究者发现,circRNA参与了心血管疾病的发生发展,它在调控心血管疾病中的巨大潜能,提示其在临床诊断和治疗方面具有广阔的前景.本文将从circRNA的特征、主要功能及其与心血管疾病的关系等方面对近年来国内外的研究进展作一综述.
Objective To evaluate the status of hypertension in community residents ,and analyze the impact of environmental factors on prevalence of hypertension ,and to propose countermeasures against hypertension . Methods Field epidemiology methods were adopted ,physical examinations and laboratory studies were performed ,between April and September ,2014 . The relevant data collected were then ana‐lyzed . Results The total prevalence of hypertension was 20 .9% ,and the difference between male and female was statistically significant . The prevalence of hypertension in population assorted by age was higher in the older population ,with the highest prevalence in the group of age ≥60 . Multivariate logistic regression analysis showed that ,after the adjustment of sex ,age ,education level ,and marital status ,the environmental risk factors of hypertension were high drinking frequency ,high BMI and central obesity . Conclusion High blood pressure of residents is ascribed to many factors in the communities that we stud‐ied . We need to intensify the promotion of healthy lifestyle in community ,and gradually increase the public awareness of hypertension and blood pressure control .
