Objective To explore the effects of a specific COX-2 inhibitor SC236 on biological behavior of renal cell carcinoma RCC-949. Methods Various concentrations of SC236 were incubated with cultured RCC-949 in vitro for 24,48 and 72 hours,and the growth of RCC-949 was detected by the MTT method.RCC-949 was stimulated by 50 and 150 umol/L of SC236 for 48 hours,and then the expression of COX-2 mRNA was detected by RT-PCR.RCC-949 was stimulated by 50 and 100 umol/L of SC236 for 24 hours,and then PGE2 was detected by enzyme linked immunosorbent assay.RCC-949 was stimulated by 100 umol/L of SC236 for 24 hours,and then the cell cycle and apoptosis were detected by flow cytometer(FCM). Results SC236 inhibited the proliferation of RCC-949 in a time-and dose-dependent manner and decreased the expression of COX-2 mRNA.Different concentrations of SC236 could restrain the liberation of PGE2 and induce RCC-949 to apoptosis, and there were statistically significant differences between experimental group and control group(P<0.05). Conclusions Specific COX-2 inhibitor SC236 can inhibit the proliferation of renal cell carcinoma(RCC-949),regulate the delivery of PGE2 and induce RCC-949 to apoptosis.
Objective To explore the expression of COX-2 and Ki-67 in thyroid neoplasm tissue,and to analyze the correlation between the tumor-related factors and their biocharacteristics,the postoperative patients' survival time. Methods Immunohistochemical SP method was used to detect the expression of COX-2 and Ki-67 in thyroid neoplasm tissues of 68 thyroid neoplasm patients and in normal thyroid tissues of 40 healthy controls.All the data were analyzed by SPSS16.0 software. Results The positive expression rates of COX-2 and Ki-67 in thyroid neoplasm tissues(60.29%,67.64%) were higher than those in healthy controls(2.5% and 27.5%),and the difference was statistically significant(P0.05).The expression of COX-2 and Ki-67 in thyroid neoplasm tissue was positively correlated with the size of the tumor,differentiation degree,invasive depth,TNM stage,and distal metastasis(P0.05).The five-year survival rate of the patients with positive expression of COX-2 and Ki-67 was lower than that of healthy controls(P0.05). Conclusions COX-2 and Ki-67 could be regarded as the effective indices for early diagnosis and prognosis prediction of thyroid neoplasm,which could guide the clinical evaluation of biological behaviors of thyroid tumors.
